In vitro immunotoxicity of important mycotoxins detected in commercial dry dog and cat food in relation to internal exposure.

A screening of commercial dog and cat foods revealed considerable variation in metabolite profiles across feed categories, with standard feeds generally showing higher concentrations of Fusarium metabolites, phytoestrogens, and plant metabolites than specialized senior or intestinal diets. Concentrations of aflatoxin B1 (AFB1), deoxynivalenol (DON) DON, zearalenone (ZEN), ochratoxin A (OTA), fumonisin B1 + 2 (FB1 + FB2), and T-2/HT-2 toxins remained below existing guidance values and maximum limits. In vitro assays using peripheral blood mononuclear cells (PBMCs) from dogs and cats demonstrate

Species-specific inhibition of human and rat aromatase by mycotoxins: Integrated enzyme assays, docking, and structure-activity relationship analysis.

In placental tissue, human and rat aromatases convert testosterone to estradiol, which was prioritized for investigation over others due to its susceptibility to endocrine disruption by mycotoxins, yet their effects remain unclear. The objective of this study is to investigate the effects of mycotoxins on human and rat aromatase, including inhibition strength, mechanism, and structural-activity relationships (SAR). Screening was conducted to evaluate mycotoxin inhibition using placental microsomes by HPLC-MS/MS. IC50 and Ki values were determined as well as effects on estradiol secretion in hu

Hazard characterization of the mycotoxins enniatins and beauvericin to identify data gaps and improve risk assessment for human health.

Enniatins (ENNs) and beauvericin (BEA) are cyclic hexadepsipeptide fungal metabolites which have demonstrated antibiotic, antimycotic, and insecticidal activities. The substantial toxic potentials of these mycotoxins are associated with their ionophoric molecular properties and relatively high lipophilicities. ENNs occur extensively in grain and grain-derived products and are considered a food safety issue by the European Food Safety Authority (EFSA). The tolerable daily intake and maximum levels for ENNs in humans and animals remain unestablished due to key toxicological and toxicokinetic dat

Investigating the role of food pollutants in autism spectrum disorder: a comprehensive analysis of heavy metals, pesticides, and mycotoxins.

Food pollutants, including heavy metals, pesticides, and mycotoxins, have been proposed as potential risk factors for autism spectrum disorder (ASD) during pregnancy and early childhood. This paper examines the impact of food pollutants on ASD risk. A systematic search through PubMed, Google Scholar, and Sciverse yielded studies from 1990 to present. Research indicates elevated levels of heavy metals in children with ASD, linking pesticides and toxins to brain development disruptions. Mycotoxins, specifically, show a correlation with ASD and can contaminate food, posing a threat to neurodevelo

Effects of dietary Fusarium mycotoxins on intestinal lymphocyte subset populations, cell proliferation and histological changes in avian lymphoid organs.

An experiment was conducted to investigate the effects of dietary Fusarium mycotoxins on gut immunity, cell proliferation, and histology of avian lymphoid organs. The efficacy of a polymeric glucomannan mycotoxin adsorbent (GMA) was also determined. Seventy-two one-day-old male turkey poults were fed corn, wheat, and soybean meal-based diets for 21 days. Diets included control grains, contaminated grains and contaminated grains +0.2% GMA. The major contaminant was deoxynivalenol (3.9 μg/g) with lesser amounts of zearalenone (0.67-0.75 μg/g), 15-acetyl-deoxynivalenol (0.34 μg/g) and HT-2 toxin

Suppression of immune response in the B6C3F1 mouse after dietary exposure to the Fusarium mycotoxins deoxynivalenol (vomitoxin) and zearalenone.

The effect that dietary exposure to the naturally-occurring Fusarium graminearum toxins deoxynivalenol (DON) and zearalenone (ZEA) may have on immune function was assessed in the B6C3F1 mouse. Dietary DON depressed the plaque-forming response to sheep red blood cells, the delayed hypersensitivity response to keyhole limpet haemocyanin and the ability to resist Listeria monocytogenes. Listerial resistance was similarly decreased in control mice fed restricted diets comparable to the dietary restriction caused by DON-induced feed refusal, whereas equivalent food restriction did not decrease the

Metabolomics and Lipidomics Reveal the Metabolic Disorders Induced by Single and Combined Exposure of Fusarium Mycotoxins in IEC-6 Cells.

Deoxynivalenol (DON), fumonisin B1 (FB1), and zearalenone (ZEN) are typical fusarium mycotoxins that occur worldwide in foodstuffs, posing significant health hazards to humans and animals. Single and combined exposure of DON, FB1, and ZEN leads to intestinal toxicity but the toxicology mechanism research is still limited. In this study, we explored the cytotoxicity effects of DON, FB1, ZEN, and their combination in rat intestinal epithelial cell line 6 (IEC-6) cells. Cell viability results showed that the cytotoxicity potency ranking was DON > ZEN > FB1. Furthermore, both DON + FB1 and DON + Z

Chemical Genetics in C. elegans Identifies Anticancer Mycotoxins Chaetocin and Chetomin as Potent Inducers of a Nuclear Metal Homeostasis Response.

C. elegans numr-1/2 (nuclear-localized metal-responsive) is an identical gene pair encoding a nuclear protein previously shown to be activated by cadmium and disruption of the integrator RNA metabolism complex. We took a chemical genetic approach to further characterize regulation of this novel metal response by screening 41,716 compounds and extracts for numr-1p::GFP activation. The most potent activator was chaetocin, a fungal 3,6-epidithiodiketopiperazine (ETP) with promising anticancer activity. Chaetocin activates numr-1/2 strongly in the alimentary canal but is distinct from metal exposu

Silver Nanolabels-Assisted Ion-Exchange Reaction with CdTe Quantum Dots Mediated Exciton Trapping for Signal-On Photoelectrochemical Immunoassay of Mycotoxins.

Mycotoxins, highly toxic secondary metabolites produced by many invading species of filamentous fungi, contaminate different agricultural commodities under favorable temperature and humidity conditions. Herein, we successfully devised a novel signal-on photoelectrochemical immunosensing platform for the quantitative monitoring of mycotoxins (aflatoxin B1, AFB1, used as a model) in foodstuffs on the basis of silver nanolabels-assisted ion-exchange reaction with CdTe quantum dots (QDs) mediated hole-trapping. Initially, a competitive-type immunoreaction was carried out on a high-binding micropla

Designer Enzymes: Food-System-Oriented Biocatalysts for Mitigating Mycotoxin Contamination and Reducing Postharvest Grain Loss.

Mycotoxin contamination remains a major challenge to food and feed safety, agricultural productivity, and postharvest resource utilization. Although numerous mycotoxin-transforming enzymes have been discovered and increasingly improved through protein engineering, only a limited number have progressed from laboratory characterization to practical food or feed applications. This translational gap arises because catalytic efficiency alone does not address matrix interference, processing stress, scalable production, formulation, degradation-product safety, and application-specific regulatory requ

Species of the Fusarium incarnatum-equiseti species complex associated with panicles of pearl millet and potential for mycotoxin production.

Pearl millet is an expanding crop in Brazil, used as a cover crop and destined for animal food. Fusarium species colonize the panicle and may cause head mold or compromise the safety of grains by producing mycotoxins. We collected samples of panicles from different producing regions of Brazil with and without signs of Fusarium infection to investigate which species are associated with panicles and their potential for producing mycotoxins. A total of 154 Fusarium isolates were obtained and assigned to three species complexes (SC) based on morphology. Fusarium incarnatum-equiseti SC was dominant

Investigating the Correlations Between Weather Factors and Mycotoxin Contamination in Corn: Evidence from Long-Term Data.

Mycotoxins are secondary metabolites produced by certain fungi, posing significant health risks to humans and animals through contaminated food and feed. These fungi, and consequently the mycotoxins which they produce, are strongly influenced by weather, and this shifts over time due to climate change, leading to more frequent and severe events, such as heat waves, storms, and heavy rainfall. This study investigates how long-term weather trends and climatic factors impacted mycotoxin levels in corn samples over a 17-year period (2006-2022) across 12 countries, with a focus on 136 specific weat
