A multimodal neuroimaging study of cerebrovascular regulation: protocols and insights of combining electroencephalography, functional near-infrared spectroscopy, transcranial Doppler ultrasound, and physiological parameters.

Objective. The current paper describes the creation of a simultaneous trimodal neuroimaging protocol. The authors detail their methodologica … orespiratory variables. The EEG-fNIRS-TCD protocol was attached to a 21 year-old male who completed neurovascular coupling/functional hyperemia (finger tapping and 'Where's Waldo/Wally?'), dynamic cerebral autoregulation (squat-stand maneuvers), and cerebrovascular reactivity tasks (end-tidal clamping during hypocapnia/hypercapnia).Main results. In a pilot participant, …

Effects of changes in end-tidal PO2 and PCO2 on neural responses during rest and sustained attention.

Impairments of cognitive function during alterations in arterial blood gases (e.g., high-altitude hypoxia) may result from the disruption of neurovascular coupling; however, the link between changes in arterial blood gases, cognition, and cerebral blood flow (CBF) is poorly understood. To interrogate this link, we developed a multimodal empirical strategy capable of monitoring neural correlates of cognition and CBF simultaneously. Human participants performed a sustained attention task during hypoxia, hypercapnia, hypocapnia, and normoxia while electroencephalographic (EEG) activity and CBF (m…

Cocaine Decreases Spontaneous Neuronal Activity and Increases Low-Frequency Neuronal and Hemodynamic Cortical Oscillations.

Low-frequency oscillations (LFOs) in hemodynamics assessed by fMRI reflect synchronized neuronal activities and are the basis for mapping brain function and its disruption by drugs and disease. Here we assess if cocaine disrupts coupling between neuronal and vascular LFOs by simultaneously measuring cortical field potentials (FP) and cerebral blood flow (CBF) regarding their LFOs (0-1 Hz) spectral bandwidths in the somatosensory cortex of naïve and chronic cocaine-exposed rats at baseline and during cocaine intoxication. While across all conditions the dominant oscillation frequencies for FP a

Assessing paedophilia based on the haemodynamic brain response to face images.

OBJECTIVES: Objective assessment of sexual preferences may be of relevance in the treatment and prognosis of child sexual offenders. Previous research has indicated that this can be achieved by pattern classification of brain responses to sexual child and adult images. Our recent research showed that human face processing is tuned to sexual age preferences. This observation prompted us to test whether paedophilia can be inferred based on the haemodynamic brain responses to adult and child faces. METHODS: Twenty-four men sexually attracted to prepubescent boys or girls (paedophiles) and 32 men

Microvascular dysfunction and aberrant network activity drive reduced brain oxygenation in a mouse tauopathy model.

Tauopathies such as Alzheimer's disease and frontotemporal dementia are leading causes of cognitive impairment, characterized by accumulatio … in the context of pronounced network hyperactivity and hypersynchrony, which combined with impaired neurovascular coupling, lead to reduced oxygen availability, episodic hypoxia, and disturbed metabolic homeostasis. Collectively, these findings identify reduced brain oxygenation driven by an imbalance between neuronal hyperactivity and diminished oxygen delivery as a pr …

Dual-layer microneedles with NO/O2 releasing for diabetic wound healing via neurogenesis, angiogenesis, and immune modulation.

Diabetic wounds present multiple functional impairments, including neurovascular dysregulation, oxidative imbalance, and immune dysfunction, making wound healing particularly challenging, while traditional therapeutical strategies fail to address these complex issues effectively. Herein, we propose a strategy utilizing dual-layer microneedles to deliver therapeutic gases by modulating neurovascular coupling and immune functions for diabetic wound treatment. The microneedle can respond to reactive oxygen species (ROS) in the diabetic microenvironment and subsequently generate oxygen (O2) and ni…

Implicating endothelial cell senescence to dysfunction in the ageing and diseased brain.

Cerebrovascular endothelial cells (CECs) are integral components of both the blood-brain barrier (BBB) and the neurovascular unit (NVU). As the primary cell type of the BBB, CECs are responsible for the tight regulation of molecular transport between the brain parenchyma and the periphery. Additionally, CECs are essential in neurovascular coupling where they help regulate cerebral blood flow in response to regional increases in cellular demand in the NVU. CEC dysfunction occurs during both normative ageing and in cerebrovascular disease, which leads to increased BBB permeability and neurovascu…

Early cerebral amyloid angiopathy-related pathology is associated with localized functional brain connectivity.

BackgroundThe mechanisms of early brain damage in cerebral amyloid angiopathy (CAA), a highly prevalent comorbid condition in Alzheimer's disease, are not completely understood. While current CAA diagnosis relies on late-stage MRI markers, impaired neurovascular coupling (NVC) has emerged as a promising early marker.ObjectiveTo investigate whether early vascular changes in CAA are associated with functional brain consequences, we examined the association between NVC and various functional connectivity metrics.MethodsWe analyzed MRI data from 93 older adults (71 ± 9 years old). The Effect of a Neuronal Nitric Oxide Synthase Inhibitor on Neurovascular Regulation in Humans.

BACKGROUND: Neurovascular coupling (NVC) is a key process in cerebral blood flow regulation. NVC ensures adequate brain perfusion to changes in local metabolic demands. Neuronal nitric oxide synthase (nNOS) is suspected to be involved in NVC; however, this has not been tested in humans. Our objective was to investigate the effects of nNOS inhibition on NVC in humans. METHODS: We performed a 3-visit partially randomized, double-blinded, placebo-controlled, crossover study in 12 healthy subjects. On each visit, subjects received an intravenous infusion of either S-methyl-L-thiocitrulline (a sel…

Hemodynamic responses are abnormal in isolated cervical dystonia.

Neuroimaging studies using functional magnetic resonance imaging (fMRI), which measures brain activity by detecting the changes in blood oxygenation levels, are advancing our understanding of the pathophysiology of dystonia. Neurobiological disturbances in dystonia, however, may affect neurovascular coupling and impact the interpretability of fMRI studies. We evaluated here whether the hemodynamic response patterns during a behaviorally matched motor task are altered in isolated cervical dystonia (CD). Twenty-five CD patients and 25 healthy controls (HCs) underwent fMRI scanning during a paced…

Blood pressure variability compromises vascular function in middle-aged mice.

Blood pressure variability (BPV) has emerged as a significant risk factor for cognitive decline and dementia, independent of alterations in … (myogenic response) in parenchymal arterioles of BPV mice. While sensory stimulus-evoked dilations (neurovascular coupling) were amplified at higher BP levels in control mice, this pressure-dependent effect was abolished in BPV mice. Our findings indicate that chronic BP fluctuations impair vascular function within the neurovascular complex and contribute to cognitive d …

Olig2+/NG2+/BLBP+ astrocyte progenitors: a novel component of the neurovascular unit in the developing mouse hippocampus.

Astrocytes are key components of the neurovascular unit. While we have recently identified Olig2+ astrocyte progenitors (ASPs) in the developing mouse dentate gyrus (DG), their molecular signature remains incompletely characterized. Here we demonstrate that Olig2+ ASPs predominantly express brain lipid-binding protein (BLBP), while only a small population of them expresses gfap-GFP. These Olig2+/BLBP+ ASPs co-express the transcription factors Sox3, Sox9 and the proteoglycan NG2 but not Sox10, a marker for oligodendrocyte progenitors (OLPs). Olig2+ ASPs appear from embryonic day 18 (E18) onward

Calcium dysregulation in neurovascular function in Alzheimer's disease: mechanisms, therapeutic targets.

Neurovascular dysfunction, characterized by impaired cerebral blood flow (CBF) autoregulation and neurovascular coupling (NVC), represents a critical upstream facilitator of Alzheimer's disease (AD) that precedes overt cognitive decline. While traditional therapeutic strategies that focus strictly on amyloid β clearance have achieved limited long-term clinical efficacy, targeting the mechanisms governing neurovascular uncoupling offers a promising frontier. Calcium ions (Ca2+) serve as the quintessential second messenger, linking neuronal metabolic demand to real-time microvascular responsiven…

Molecular mechanisms of blood-brain barrier dysfunction caused by obesity: pathophysiological understandings and treatment targets.

Chronic low-grade inflammation and systemic metabolic dysregulation are linked to obesity, a rapidly expanding worldwide health concern that affects the central nervous system (CNS) in addition to peripheral organs. Dysfunction of the blood-brain barrier (BBB), which impairs selective permeability, tight junction integrity, and neurovascular coupling, is one of its most serious neurobiological effects. The BBB, which is made up of pericytes, endothelial cells, and astrocytic end-feet, depends on specialized transporters and tight junction proteins like occludin, ZO-1, and claudin-5 to control …

Effect of an inhibitor of neuronal nitric oxide synthase 7-nitroindazole on cerebral hemodynamic response and brain excitability in urethane-anesthetized rats.

The role of neuronal nitric oxide synthase (nNOS) in the pathophysiology of epilepsy and seizures remains disputable. One of the reasons why results from the acute in vivo studies display controversies might be the effect on the regulation of cerebral blood flow (CBF) during pharmacologically induced alterations of NO system. We examined neurovascular coupling in the rat sensorimotor cortex in response to transcallosal stimulation under nNOS inhibition by 7-nitroindazole (7-NI). Adult Wistar rats were anesthetized with urethane and epidural silver EEG electrodes were implanted over sensorimoto…

The dental pulp as a high-resolution model for neurovascular niche biology.

Building functional tissue in vitro shares a common ceiling. Current models span vascular co-development, trophic interaction, vascular-to-neural sensitization, barrier function, and even functional neuromuscular coupling, yet none has reconstituted functional neurovascular (NV) coupling. We argue that, on one specific axis, the dental pulp is the most direct single-source substrate for dissecting this coupling. A single, clinically accessible neural-crest tissue supplies the perivascular compartment together with glial and neurotrophic support for the neural compartment, in a volume amenable
