Klebsiella pneumoniae, Azotobacter vinelandii and Rhodobacter capsulatus were cultivated in media containing 99MoO4(2-) . The distribution of 99Mo in cells grown under conditions of repression and derepression of nitrogenase synthesis, was investigated by anion-exchange (DEAE-Sephacel) chromatography. Cells of K. pneumoniae took up MoO4(2-) only under conditions of derepression of nitrogenase thus serving the formation of the FeMo cofactor of the MoFe protein (Kp1) as the predominant Mo-containing species. In the case of A. vinelandii, under diazotrophic growth conditions, molybdenum was prefe

Calcium (Ca2+) and phosphorous (PO43-) significantly influence the form and effectiveness of nitrogen (N), however, the precise mechanisms governing the adsorption of ammonium nitrogen (NH4+-N) and nitrate nitrogen (NO3--N) are still lacking. This study employed batch adsorption experiments, charge distribution and multi-site complexation (CD-MUSIC) models and density functional theory (DFT) calculations to elucidate the mechanism by which Ca2+ and PO43- affect the adsorption of NH4+-N and NO3--N on the goethite (GT) surface. The results showed that the adsorption of NH4+-N on the GT exhibited

Iron is a common cofactor in biological processes such as respiration, photosynthesis, and nitrogen fixation. The genes isiC and isiD encode unknown proteins, and the growth of ΔisiC and ΔisiD mutants is inhibited under iron-deficient conditions. To study the regulatory mechanisms of IsiC and IsiD during iron starvation, we carried out transcriptome and metabolome sequencing. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the photosynthesis, nitrogen metabolism, and ABC transporter pathways play a vital role in regulating iron deficiency. Upon iron repletion, IsiC and …

The nitrogen-related phosphoenolpyruvate phosphotransferase system (PTS(Ntr) ) is involved in controlling ammonia assimilation and nitrogen fixation. The additional role of PTS(Ntr) as a regulatory link between nitrogen and carbon utilization in Escherichia coli is assumed to be closely related to molecular functions of IIA(Ntr) in potassium homeostasis. We have determined the crystal structure of IIA(Ntr) from Burkholderia pseudomallei (BpIIA(Ntr) ), which is a causative agent of melioidosis. The crystal structure of dimeric BpIIA(Ntr) determined at 3.0 Å revealed that its active sites are mu…

The reduction of nitrogen, acetylene, azide, and cyanide at various oxygen concentrations by nitrogenase from Azotobacter vinelandii was measured with a well-defined system. Oxygen inhibited the reduction of each substrate uncompetitively. The inhibition constants (K(i)) were 0.014, 0.023, 0.008, and 0.003 atm of oxygen for reduction of nitrogen, acetylene, azide, and cyanide, respectively. The system used included ATP-generating components, subcellular particles from A. vinelandii with high nitrogenase specific activity, and illuminated spinach chloroplasts plus carriers to supply electrons.

The filamentous heterocyst-forming cyanobacterium Anabaena sp. PCC 7120 is an important model organism for studying cell differentiation, nitrogen fixation, and photosynthesis. This cyanobacterium possesses a high number of membrane transporters. Not much is known about the roles of the membrane transporters, especially the ATP-binding cassette (ABC) transporters, in the multidrug resistance of this cyanobacterium. In the present work, we performed a mutational analysis of the genes alr4280/alr4281/alr4282 and alr3647/alr3648/alr3649 that code for the components of putative ABC exporters and a…

High NUE (nitrogen use efficiency) has great practical significance for sustainable crop production. Wheat is one of the main cultivated crops worldwide for human food and nutrition. However, wheat grain productivity is dependent upon cultivars with high NUE in addition to the application of nitrogen fertilizers. In order to understand the molecular mechanisms exhibiting a high NUE response, a comparative transcriptomics study was carried out through RNA-seq analysis to investigate the gene expression that regulates NUE, in root and shoot tissue of N-efficient (PBW677) and N-inefficient (703)

The factors that control the growth and nitrogen fixation rates of marine diazotrophs such as Trichodesmium have been intensively studied because of the role that these processes have in the global cycling of carbon and nitrogen, and in the sequestration of carbon to the deep sea. Because the phosphate concentrations of many ocean gyres are low, the bioavailability of the larger, chemically heterogeneous pool of dissolved organic phosphorus could markedly influence Trichodesmium physiology. Here we describe the induction, by phosphorus stress, of genes from the Trichodesmium erythraeum IMS101 …

Collagen hydrogels are widely used as biomimetic three-dimensional matrices whose mechanical behaviour depends strongly on fibrillar micro- and nanoarchitecture and crosslinking state. Electron microscopy offers high spatial resolution for structural analysis, but sample preparation can substantially distort highly hydrated collagen networks, thereby confounding interpretation. Here, we compared common SEM and TEM preparation workflows for their ability to preserve structurally subtle, mechanically validated differences in collagen type I hydrogels. Collagen hydrogels containing riboflavin wer

Acidic soils are widespread in global agroforestry systems and severely constrain crop production through proton stress, aluminum/manganese phytotoxicity, phosphorus fixation, and rhizosphere microbiome reassembly. Rhizosphere diazotrophs can theoretically contribute to plant nitrogen (N) nutrition and stress adaptation. However, their N-fixation efficiency and growth-promoting effects in acidic soils are often highly unstable, limiting the predictability of field applications. This narrative mechanistic review synthesizes and critically interprets evidence on the physicochemical filters gover

Nitric oxide (NO), a signaling molecule and free radical, regulates microalgal photosynthesis, yet its potential in enhancing carbon fixation alongside pollutant mitigation remains underexplored. This study investigated the effects of NO (0, 50, 100 ppm) in simulated flue gas (15% CO2) on Chlorella sp., examining intracellular/extracellular NO dynamics, antioxidant response, key enzyme activity (nitrate reductase), and growth-photosynthesis coupling. Under optimized aeration (25 mL min-1, 3 h), 50 ppm of NO optimally elevated intracellular (0.62-1.30 nmol L-1) and extracellular (2.56-3.90 nmol

Nitrogen (N2) fixation by nature, which is a crucial process for the supply of bio-available forms of nitrogen, is performed by nitrogenase. This enzyme uses a unique transition-metal-sulfur-carbon cluster as its active-site co-factor ([(R-homocitrate)MoFe7S9C], FeMoco)1,2, and the sulfur-surrounded iron (Fe) atoms have been postulated to capture and reduce N2 (refs. 3-6). Although there are a few examples of synthetic counterparts of the FeMoco, metal-sulfur cluster, which have shown binding of N2 (refs. 7-9), the reduction of N2 by any synthetic metal-sulfur cluster or by the extracted form

With the growing interest in sustainable agriculture, there has been a surge in exploration of multitrophic interactions between plants and microbes that can help plants adapt to changing environments and enhance their resilience to climate changes. One such beneficial microbe is Gluconacetobacter diazotrophicus, an aerobic, nitrogen-fixing endophyte currently being studied because of its ability to fix atmospheric nitrogen within plant tissues under aerobic conditions. This endophyte also promotes plant growth through processes like phytohormone production, nutrient solubilization, and improv

Although the metabolic pathways that allow the utilization of one-carbon compounds as sole sources of carbon and energy (methylotrophy) are well characterized, this understanding has been substantially refined and expanded in recent years. The paradigm-shifting discovery of the lanthanide-dependent methanol dehydrogenase, XoxF, established the biological relevance of rare-earth metals and revealed that methylotrophy required reassessment. We now know that XoxF is broadly distributed among bacteria and may actually constitute an ancestral form by which methylotrophy initially evolved, as well a

Exploring the natural diversity of phylogenetically distant nitrogenases is crucial for gaining new insights into the mechanism of atmospheric N2 fixation and unlocking biotechnological developments in sustainable ammonia production. Here, we investigated the N2-fixing system of Methanocaldococcus infernus, a deep-sea hyperthermophilic archaeon growing diazotrophically at 92 °C. This natively isolated nitrogenase has a melting temperature close to the water boiling point, with an extrapolated specific activity superior to mesophilic homologues. The crystal structures obtained at near-atomic re

This study assessed the effects of 22 years of sustainable (Smng) versus conventional (Cmng) management on cyanobacteria-enriched microbial communities in a Mediterranean olive orchard. Surface soils were selectively cultivated in nitrogen-free Bristol medium to obtain cultivable cyanobacteria-dominated, nitrogen-fixing assemblages for molecular and metabolomic analyses. Cyanobacteria-enriched assemblages derived from Smng soils showed greater phylogenetic diversity and distinct community clustering under N-free cultivation conditions. At the cyanobacterial genus level, Cmng-derived communitie
