Clinicians typically avoid antibiotics use during immunotherapy due to concerns about reduced efficacy. However, cancer patients requiring antibiotics postoperatively or for infections urgently need options that provide antimicrobial coverage while potentially enhancing, rather than impairing, immunotherapy. Restoring ferroptosis susceptibility represents a promising strategy to overcome immunotherapy resistance, yet the role of antibiotics in modulating ferroptosis and interacting with immunotherapy remains unexplored. In this study, we screened 96 FDA-approved antibiotics across seven pharma

Di-(2-ethylhexyl) phthalate (DEHP) is a ubiquitous environmental endocrine disruptor that leaches from polyvinyl chloride products and metabolizes to mono-(2-ethylhexyl) phthalate (MEHP), causing spermatogenic dysfunction in males. However, the molecular mechanism linking DEHP/MEHP to Sertoli cell damage remains unclear. This study aimed to clarify the role of p38/NCOA4-dependent ferritinophagy in DEHP-induced ferroptosis and validate potential therapeutic targets. Prepubertal male KM mice were exposed to DEHP (250, 500, 1000 mg/kg/day) via oral gavage for 28 days (cyclophosphamide as positive

Ferroptosis contributes to intervertebral disc degeneration (IVDD), yet its upstream regulatory mechanisms in nucleus pulposus cells (NPCs) remain unclear. Here, we identify a novel UCHL3-HMGB1-NCOA4 signaling axis that drives NPC ferroptosis. Transcriptomic and clinical analyses reveal that UCHL3 is upregulated in degenerative discs, with its expression correlating positively with both IVDD severity and ferroptosis markers. Mechanistically, UCHL3 deubiquitinates and stabilizes HMGB1 by cleaving Lys48-linked polyubiquitin chains, thereby promoting the accumulation of cytoplasmic HMGB1. This st

The pathophysiology of osteoarthritis (OA), a common degenerative joint disease, is complicated. The significance of autophagy irregularities and iron metabolism issues in OA has garnered a lot of interest in recent years, with "Ferritinophagy" mediated by NCOA4 acting as a crucial link between the two and potentially a novel therapeutic target. The purpose of this research is to determine whether resveratrol, a naturally occurring substance, protects cartilage via controlling the ferritinophagy network. Using differential analysis, weighted gene co-expression network analysis, and other machi

BACKGROUND: Acetaminophen (APAP) overdose is a leading cause of drug-induced liver injury (DILI) and can progress to acute liver failure (ALF). Hepatocyte death is widely recognized as the central event in APAP-induced liver injury; however, the underlying mechanisms are complex and not yet fully elucidated. Ferroptosis, a recently identified form of programmed cell death characterized by glutathione (GSH) depletion and disruption of cellular redox homeostasis, shares key features with APAP-induced hepatotoxicity. This study aimed to investigate the role of ferroptosis in APAP-induced liver in

Ischemic stroke (IS) constitutes the majority of stroke cases. Ferroptosis, a non-apoptotic form of programmed cell death, is an essential mechanism of IS. This study aimed to investigate a ferroptosis-related molecular mechanism in IS progression. IS model in vitro was induced by oxygen-glucose deprivation/reoxygenation (OGD/R) in SH-SY5Y cells. RT-qPCR and Western blotting were performed for detection of NOP2/Sun RNA methyltransferase family member 2 (NSUN2), nuclear receptor coactivator 4 (NCOA4), and Krueppel-like factor 6 (KLF6). Cell functions were assessed by MTT assay, flow cytometry,

Iron is an essential nutrient for mitochondrial metabolic processes, including mitochondrial respiration. Ferritin complexes store excess iron and protect cells from iron toxicity. Therefore, iron stored in the ferritin complex might be utilized under iron-depleted conditions. In this study, we show that the inhibition of lysosome-dependent protein degradation by bafilomycin A1 and the knockdown of NCOA4, an autophagic receptor for ferritin, reduced mitochondrial respiration, respiratory chain complex assembly, and membrane potential under iron-sufficient conditions. However, autophagy did not

BACKGROUND AND OBJECTIVES: GREB1-rearranged uterine tumours encompass a group of uterine mesenchymal tumours with varied histologic appearances. The fusion partners to GREB1 include NCOA1-3, SS18 and NR4A3. Given that some GREB1-rearranged uterine tumours exhibit histologic features of uterine tumours resembling ovarian sex cord tumour (UTROSCT), there is a general belief that GREB1-rearranged uterine mesenchymal tumours are part of the UTROSCT family.
METHODS: In this study, we applied global DNA methylation and copy number analyses to a series of 10 GREB1-rearranged uterine tumours and 21 c

BACKGROUND: Ferroptosis in intestinal epithelial cells (IECs) acts as a crucial mechanism driving intestinal mucosal injuries and inflammatory reactions in colitis. Nuclear receptor coactivator 4 (NCOA4)-mediated ferritinophagy, a primary pathway for intracellular iron storage, takes part in regulating ferroptosis. Nevertheless, it is yet unknown whether ferritinophagy plays a role in ferroptosis of IECs in colitis. Hence, this experiment used dextran sulfate sodium salt (DSS) to establish an inflammation model with NCM460 cells and an animal model of colitis to discover the role of ferritinop

The liver represents a primary target organ frequently compromised during the progression of sepsis. Ferroptosis is a distinct mode of regulated cell death driven by iron-dependent lipid peroxidation, which has been identified as being involved in the pathogenesis of sepsis-induced hepatic dysfunction. However, the regulation of ferroptosis in hepatocytes by m6A-modified RNAs under sepsis remains unclear. In this study, we demonstrated that the m6A modification level was markedly increased in septic liver injury, complying with upregulation of the m6A methyltransferase METTL4 and the reader YT

Heart failure (HF) remains a critical global health challenge, with accumulating evidence implicating iron metabolism dysregulation through ferritinophagy activation in disease progression. Our study focused on sterol O-acyltransferase 1 (SOAT1), an established regulator of hypoxia-induced cardiomyocyte dysfunction, investigating its role in HF pathogenesis through ferritinophagy and ferroptosis regulation. Here, mouse HF models were generated via transverse aortic constriction (TAC), while primary cardiomyocyte injury was induced by isoproterenol (ISO) exposure. We demonstrated that SOAT1 kno

Spindle cell mesenchymal neoplasms are a diverse and often challenging diagnostic group. While morphological impression is sufficient for some diagnoses, increasingly immunohistochemical and even molecular data is required to render an accurate diagnosis, which can lead to the characterization of new entities. We describe five cases of novel mesenchymal neoplasms with rearrangements in the NCOA2 and NCOA3 genes partnered with either CTCF or CRTC1. Three tumors occurred in the head and neck (palate, auditory canal), while the other two were in visceral organs (lung, urinary bladder). All cases

ESR1 and GREB1 fusions have recently been described in uterine tumor resembling ovarian sex cord tumor (UTROSCT). Thus far, recurrences have been documented in a subset of those harboring GREB1 fusions, but not in those with ESR1 rearrangements. Here we describe the clinicopathologic features of 3 recurrent UTROSCTs with striking rhabdoid morphology (an unusual feature of these tumors overall) and ESR1-NCOA2 fusions. The patients were 32, 37, and 54 years at initial diagnosis and first recurrence occurred at 7, 9, and 32 years. The primary tumors (available in two cases) were centered in the m

Cells rely on autophagy to clear misfolded proteins and damaged organelles to maintain cellular homeostasis. In this study we use the new autophagy inhibitor PIK-III to screen for autophagy substrates. PIK-III is a selective inhibitor of VPS34 that binds a unique hydrophobic pocket not present in related kinases such as PI(3)Kα. PIK-III acutely inhibits autophagy and de novo lipidation of LC3, and leads to the stabilization of autophagy substrates. By performing ubiquitin-affinity proteomics on PIK-III-treated cells we identified substrates including NCOA4, which accumulates in ATG7-deficient

The mRNA of the nuclear coactivator peroxisome proliferator-activated receptor-gamma coactivator-1alpha (PGC-1alpha) increases during prolonged exercise and is influenced by carbohydrate availability. It is unknown if the increases in mRNA reflect the PGC-1alpha protein or if glycogen stores are an important regulator. Seven male subjects [23 +/- 1.3 yr old, maximum oxygen uptake (Vo(2 max)) 48.4 +/- 0.8 ml.kg(-1).min(-1)] exercised to exhaustion ( approximately 2 h) at 65% Vo(2 max) followed by ingestion of either a high-carbohydrate (HC) or low-carbohydrate (LC) diet (7 or 2.9 g.kg(-1).day(-

RATIONALE: Peroxisome proliferator-activated receptors (PPARs) (alpha, gamma, and delta/beta) are nuclear hormone receptors and ligand-activated transcription factors that serve as key determinants of myocardial fatty acid metabolism. Long-term cardiomyocyte-restricted PPARdelta deficiency in mice leads to depressed myocardial fatty acid oxidation, bioenergetics, and premature death with lipotoxic cardiomyopathy.
OBJECTIVE: To explore the essential role of PPARdelta in the adult heart.
METHODS AND RESULTS: We investigated the consequences of inducible short-term PPARdelta knockout in the adu

Spinal and bulbar muscular atrophy (SBMA) is a motor neuron disease caused by polyglutamine expansion mutation in the androgen receptor (AR). We investigated whether the mutant protein alters mitochondrial function. We found that constitutive and doxycycline-induced expression of the mutant AR in MN-1 and PC12 cells, respectively, are associated with depolarization of the mitochondrial membrane. This was mitigated by cyclosporine A, which inhibits opening of the mitochondrial permeability transition pore. We also found that the expression of the mutant protein in the presence of ligand results
