AIM: To use connectivity mapping, a bioinformatics approach, to identify compounds that could induce odontogenic differentiation of dental pulp cells (DPCs) and to experimentally validate this effect. A subsidiary aim was to investigate the anti-inflammatory effect of any identified compound.
METHODOLOGY: The Gene Expression Omnibus (GEO) database was searched for microarray data sets assessing odontogenic differentiation of human DPCs. An odontogenic gene expression signature was generated by differential expression analysis. The statistical significant connectivity map (ssCMap) method was u

We have shown that materials other than hydrogels commonly used in tissue engineering can be effective in enabling differentiation of dental pulp stem cells (DPSC). Here we demonstrate that a hydrophobic elastomer, polyisoprene (PI), a component of Gutta-percha, normally used to obturate the tooth canal, can also be used to initiate differentiation of the pulp. We showed that PI substrates without additional coating promote cell adhesion and differentiation, while their moduli can be easily adjusted either by varying the coating thickness or incorporation of inorganic particles. DPSC plated on

Odontogenic ameloblast-associated (ODAM) belongs to the secretory calcium-binding phosphoprotein (SCPP) gene cluster. It is expressed by the epithelial ameloblasts during the accrued mineralisation of enamel and by cells of the junctional epithelium (JE), a specialised portion of the gingiva that plays a critical role in periodontal health. In both cases, ODAM localises at the interface between the cells and the tooth surface. It is also present among the cells of the JE, and is distinctively highly expressed in many epithelial tumours. ODAM has been proposed to be a matricellular protein impl

In this report we provide data on dental eruption and tooth germ maturation at birth in a large sample constituting the broadest array of non-human primates studied to date. Over 100 perinatal primates, obtained from natural captive deaths, were screened for characteristics indicating premature birth, and were subsequently studied using a combination of histology and micro-CT. Results reveal one probable unifying characteristic of living primates: relatively advanced maturation of deciduous teeth and M1 at birth. Beyond this, there is great diversity in the status of tooth eruption and maturat

Mammalian tooth development is a precise and complicated procedure. Several signaling pathways, such as nuclear factor (NF)-κB and WNT, are key regulators of tooth development. Any disturbance of these signaling pathways can potentially affect or block normal tooth development, and presently, there are more than 150 syndromes and 80 genes known to be related to tooth agenesis. Clarifying the interaction and crosstalk among these genes will provide important information regarding the mechanisms underlying missing teeth. In the current review, we summarize recently published findings on genes re

Large cystic lesions of the maxillary sinus may initially manifest with malocclusion, cosmetic deformity, or both. Their origin may be odontogenic or nonodontogenic. Proper diagnosis of these cysts immeasurably aids in adequate (nonmutilating) treatment. A recent case of a calcifying and keratinizing odontogenic cyst that initially manifested as a facial mass exemplifies the need for adequate clinical and histological diagnosis to ensure appropriate treatment. Fewer than 35 cases have been reported since the original description in 1962.

Permanent tooth structures can respond to environmental disruptions even after the initiation of hard tissue deposition at the cusp tips, affecting dental morphology. In bioarcheological studies, linear enamel hypoplasia (LEH) provides evidence of both the presence and timing of developmental stressors that can occur during first molar odontogenesis. Yet, previous studies used LEH from the entirety of the anterior tooth surface to relate disruptions to changes in permanent tooth morphology, which could conflate causality by misrepresenting the timing of events. This study assessed associations

BACKGROUND: Tooth development hinged on reciprocal interactions between enamel and dentin, shaping tooth structures. Centrioles influenced cellular direction, critical for stem cell differentiation. Aberrant centrioles contributed to conditions like Huntington's disease and cancers. Centriole-related gene mutations, like Pericentrin and Centrosomal P4.1-associated protein (CPAP), led to tooth abnormalities, microcephaly. Our study explored the role of centrioles in ameloblasts during molar growth, shedding light on tooth development mechanisms.
METHODS: Tissue sections underwent immunofluores

We used fluorochrome labeling to study spatiotemporal variation of dentin apposition (DAR) and extension (DER) rates during crown and root formation of mandibular first molars from wild boar and domestic pigs. DAR was reconstructed along the course of dentinal tubules in four zones of the crown and in the upper root area. In all five zones, mean DAR increased during the first 30% to 40% of apposition, reaching highest values (22-23 μm/day) in the upper-lateral crown zone. Lowest values were recorded near the dentin-pulp interface (DPI). Typically, DARs in contemporaneously formed dentin areas

OBJECTIVE: Regenerative dentistry (RD) is an innovative strategy for treating necrotic teeth and regenerating damaged dental tissue. Biocompatible materials are pivotal for the advancement of RD, and the rising interest in environmental sustainability drives exploration of sustainable materials for dentistry. Bacterial nanocellulose (BNC) has emerged as a promising eco-friendly option and this study aims to assess BNC's suitability as scaffolds for regenerative dentistry applications.
METHODS: Different in vitro methods have been utilized to characterize the properties of BNC scaffolds in reg

INTRODUCTION: Delayed tooth eruption is often seen in patients with palatally displaced canines (PDC), but there is controversy over whether tooth formation is also delayed. This study attempts to elucidate the answer by addressing methodological shortcomings in previous studies.
METHODS: Archived records were searched, and 100 patients (aged 8-16 years) with unilateral PDC were identified. Records of 50 women and 50 men were matched for age and sex with unaffected controls. The exclusion criteria included hypodontia. Dental age (DA) was calculated using the population-specific Maltese databa

OBJECTIVES: Chronic rhinosinusitis (CRS) frequently stems from a dental origin, although odontogenic sinusitis (OS) remains underdiagnosed amongst different professionals. This study aimed to explore how often odontogenic causes are considered when diagnosing CRS.
MATERIALS AND METHODS: Patient records from 374 new CRS patients treated at a tertiary-level ear, nose, and throat (ENT) clinic were selected. Entries and radiological reports were assessed exploring how often dentition was mentioned and OS was suspected, how often radiologists reported maxillary teeth, and how commonly typical OS m

In past decades, morphologic, molecular, and cellular mechanisms that govern tooth development have been extensively studied. These studies demonstrated that the same signaling pathways regulate development of the primary and successional teeth. Mutations of these pathways lead to abnormalities in tooth development and number, including aberrant tooth shape, tooth agenesis, and formation of extra teeth. Here, we summarize the current knowledge on the development of the primary and successional teeth in animal models and describe some of the common tooth abnormalities in humans.

The periodontal ligament (PDL) connects the tooth root and alveolar bone. It is an aligned fibrous network that is interposed between, and anchored to, both mineralized surfaces. Periodontal disease is common and reduces the ability of the PDL to act as a shock absorber, a barrier for pathogens and a sensor of mastication. Although disease progression can be stopped, current therapies do not primarily focus on tissue regeneration. Functional regeneration of PDL may be achieved using innovative techniques, such as tissue engineering. However, the complex fibrillar architecture of the PDL, essen

Practitioners face several challenges during the root canal treatment of teeth with wide or immature apices, one of which is working length determination. There is relatively little data regarding the value of radiography and electronic apex locator (EAL) use when root formation is incomplete, and supplementary measurement techniques may be helpful. This review considers length determination for canals with wide or completely open apices in permanent and primary teeth. The Ovid Medline, Web of Science and Scopus databases were searched individually and in combinations to August 2012 using the

OBJECTIVE: The incisors of the mammalian dental arch develop from tissues arising from separated facial prominences. These primordial craniofacial structures undergo complex morphogenetic processes as they merge and fuse in a time and space dependent fashion. However, local contributions of precursor facial prominences to the incisors that develop subsequently remain unknown. The purpose of this study was to characterize the development of all three deciduous upper rostral teeth in the pig (Sus scrofa f. domestica) for the identification of the likely facial prominence contributions to the inc
