Imaging of the olfactory system.

The olfactory system consists of the primary olfactory nerves in the nasal cavity, the olfactory bulbs and tracts, and numerous intracranial connections and pathways. Diseases affecting the sense of smell can be located both extracranially and intracranially. Many sinonasal inflammatory and neoplastic processes may affect olfaction. Intracranially congenital, traumatic, and neurodegenerative disorders are usually to blame for olfactory dysfunction. The breadth of diseases that affect the sense of smell is astounding, yet the imaging ramifications have barely been explored.

GnRH-like immunoreactivity in the peripheral olfactory system and forebrain of Atlantic salmon (Salmo salar): a reassessment at multiple life history stages.

Neurons with gonadotropin releasing hormone-like immunoreactivity (GnRH-ir) were identified within the peripheral olfactory system of Atlantic salmon (Salmo salar) at multiple life history stages. Within the forebrain, GnRH-ir somata were found in the preoptic area and in the caudomedial olfactory bulb in a position comparable to the ganglion of the nervus terminalis of other teleosts. Somata positive for GnRH were also found throughout the rostro-caudal extent of the olfactory nerve, and clustered within the medial component of the olfactory nerve as it arises from the olfactory epithelium. Transport of molecules from nose to brain: transneuronal anterograde and retrograde labeling in the rat olfactory system by wheat germ agglutinin-horseradish peroxidase applied to the nasal epithelium.

Transneuronal anterograde labeling with the conjugate wheat germ agglutinin-horseradish peroxidase (WGA-HRP) has been documented in the mammalian and immature avian visual system [6,14]. Transneuronal retrograde labeling was significant only in the chick [6]. The present study was performed to determine whether transneuronal labeling could be shown in the mammalian olfactory system, whether the phenomenon was robust in adults, and whether transneuronal retrograde transport could label several transmitter-specific centrifugal afferent projections to the olfactory bulb. In addition we wished to

TRPV1-STAT3 signaling as a driver of lethal pathology during pneumococcal nose-to-brain invasion.

Nose-to-brain infection represents a non-hematogenous route by which pathogens invade the central nervous system via neural pathways. The olfactory and trigeminal systems are implicated in this process, but the molecular mechanisms underlying lethal pathology remain incompletely understood. We hypothesized that trigeminal TRPV1 and downstream STAT3 signaling contribute to disease progression during pneumococcal nose-to-brain infection. A murine intranasal infection model using Streptococcus pneumoniae was established with methimazole-induced olfactory epithelial damage and/or resiniferatoxin-m

Tau propagation in the brain olfactory circuits is associated with smell perception changes in aging.

The direct access of olfactory afferents to memory-related cortical systems has inspired theories about the role of the olfactory pathways in the development of cortical neurodegeneration in Alzheimer's disease (AD). In this study, we used baseline olfactory identification measures with longitudinal flortaucipir and PiB PET, diffusion MRI of 89 cognitively normal older adults (73.82 ± 8.44 years; 56% females), and a transcriptomic data atlas to investigate the spatiotemporal spreading and genetic vulnerabilities of AD-related pathology aggregates in the olfactory system. We find that odor iden…

Origin and endpoint of the olfactory nerve fibers: as described by Santiago Ramón y Cajal.

In the late Nineteenth Century, Santiago Ramón y Cajal was able to reproduce an exceptional illustration of the Olfactory Nerve pathway and its myriad of cells, by using the Golgi Method. Dr. Cajal focused intense study on the histology of the nervous system and published a treatise on the olfactory nerve fibers and the distinct peripheral origin and central nervous system endpoint of this unique pathway. The original title of this work is "Origen y terminación de las fibras nerviosas olfatorias" published in 1890. As the original publication is in Spanish, here we provide an English translati

Receptors Involved in COVID-19-Related Anosmia: An Update on the Pathophysiology and the Mechanistic Aspects.

Olfactory perception is an important physiological function for human well-being and health. Loss of olfaction, or anosmia, caused by viral infections such as severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has received considerable attention, especially in persistent cases that take a long time to recover. This review discusses the integration of different components of the olfactory epithelium to serve as a structural and functional unit and explores how they are affected during viral infections, leading to the development of olfactory dysfunction. The review mainly focused on

Exuberant growth and synapse formation of olfactory sensory neuron axonal arborizations.

Neural connections in the adult nervous system are established with a high degree of precision. Several examples throughout the nervous system indicate that this precision is achieved by first establishing an initial exuberant immature pattern of connectivity that is then sculpted into the adult pattern via pruning. This often emerges as an activity-dependent process. In the olfactory system, sensory axons from neurons expressing the same odorant receptor project with high precision to specific glomerular structures in the olfactory bulb. This process undergoes maturation-dependent refinements

Short-axon cells in the olfactory bulb: dendrodendritic synaptic interactions.

1. In the rabbit olfactory bulb, analysis has been carried out of extracellular unitary responses in the glomerular layer to olfactory nerve volleys. 2. Units in the glomerular layer responded to single volleys with single, double, triple or longer repetitive spike discharges. The shortest initial latencies are consistent with monosynaptic excitation from the olfactory nerves; longer latencies may reflect longer nerve pathways or polysynaptic connexions in the glomerular layer. 3. Like mitral and tufted cells, some glomerular layer units gave evidence of activation by discrete nerve bundles. T

Evaluation of brain and neurophysiologic function in isolated congenital anosmia.

PURPOSE: Isolated congenital anosmia (ICA) is a relatively rare condition characterized by a complete lack of olfactory perception dating from birth or early childhood. Congenital deficits in sensory systems appear to have distinct effects on brain function and nervous system pathway, but little is known about ICA. The study aims to explore the neural basis of ICA through measuring brain activation in response to olfactory stimuli and cerebral processing in patients with ICA. METHODS: Retrospective review of medical records of 11 ICA patients from two smell and taste centers. All patients who

[Depressive disorders and olfactory dysfunction].

Depressive disorder is a highly prevalent psychiatric condition that imposes substantial psychological burden on patients and their families, as well as significant economic costs to society. A close relationship exists between depressive disorders and abnormalities in olfactory function, and depressive disorders are frequently accompanied by marked impairments in social functioning. Patients exhibit dysfunction across multiple dimensions of olfactory processing, including olfactory sensitivity, identification, discrimination, and memory, and these abnormalities are influenced by symptom sever

Nose-to-brain transport pathways of wheat germ agglutinin conjugated PEG-PLA nanoparticles.

PURPOSE: To investigate the possible pathways for transport of wheat germ agglutinin conjugated PEG-PLA nanoparticles (WGA-NP) into the brain after nasal administration. METHODS: The nose-to-brain pathways were investigated using WGA-NP containing 6-coumarin (as a fluorescent marker) and (125)I-labeled WGA-NP. Ex vivo imaging analysis was also employed to visualize the transport process. RESULTS: Nasal administration of WGA-NP to rats resulted in transcellular absorption across the olfactory epithelium and transfer to the olfactory bulb within 5 min. After entering the lamina propria, a prop

Synaptic actions on mitral and tufted cells elicited by olfactory nerve volleys in the rabbit.

1. A unitary study has been carried out of mitral and tufted cell responses to olfactory nerve volleys in the olfactory bulb of rabbits lightly anaesthetized with urethane-chloralose. 2. With volleys of different strengths, some mitral cells responded with a spike whose latency decreased considerably as the strength increased (elastic response); other cells responded at an invariant latency (inelastic response). The former may reflect diffuse olfactory nerve inputs to the dendritic tufts in the olfactory glomeruli, while tha latter may reflect input from discrete bundles of fibres. 3. The shor

Developmental, tract-tracing and immunohistochemical study of the peripheral olfactory system in a basal vertebrate: insights on Pax6 neurons migrating along the olfactory nerve.

The olfactory system represents an excellent model for studying different aspects of the development of the nervous system ranging from neur … toward an extrabulbar region. We also demonstrate that Pax6-ir cells coursing along the developing olfactory pathways in S. canicula are young migrating (HuC/D and DCX-ir) neurons of the migratory mass that do not form part of the terminal nerve pathway. Evidences that these Pax6 neurons originate in the olfactory epithelium are also reported. As Pax6 neurons in th …

Hyposmia following laryngectomy: experimental model.

Hyposmia following laryngectomy is a well-known clinical observation, yet the causes have been controversial for many years. In an attempt to resolve this issue, an animal model was constructed. Eighteen dogs were divided into three equal groups: control dogs, dogs that underwent tracheostomy, and dogs that underwent tracheostomy and denervation of the larynx, simulating total laryngectomy. Four to 6 months following these operations, biopsies from olfactory mucosa were taken. The results showed marked changes in the olfactory mucosa of the two test groups: cystic degeneration of secretory gla

Dynamics of propagating waves in the olfactory network of a terrestrial mollusk: an electrical and optical study.

1. The procerebral (PC) lobe of the terrestrial mollusk Limax maximus contains a highly interconnected network of local olfactory interneurons that receives ipsilateral axonal projections from superior and inferior noses. This network exhibits an approximately 0.7-Hz intrinsic oscillation in its local field potential (LFP). 2. Intracellular recordings show that the lobe contains at least two classes of neurons with activity phase locked to the oscillation. Neurons in one class produce periodic bursts of spikes, followed by a period of hyperpolarization and subsequently a depolarizing afterpote

Effects of parturition and maternal experience on noradrenaline and acetylcholine release in the olfactory bulb of sheep.

Acetylcholine (ACh) and noradrenaline (NA) release in the olfactory bulb (OB) of ewes was monitored using microdialysis. Both ACh and NA release increased at parturition in multiparous but not in primiparous ewes. However, vaginocervical stimulation performed 6 hr postpartum induced an increase of ACh and NA release in both primiparous and multiparous ewes, indicating that a maturation process had occurred. Finally, pharmacological challenges to the ACh and NA inputs revealed differential responsiveness between nulliparous and multiparous nongestant ewes. These results suggest that the first p

Retronasal or internasal olfaction can mediate odor-guided behaviors in newborn mice.

Studies of olfactory deprivation have most frequently used unilateral naris occlusion to effect deprivation. Recent psychophysical evidence suggests that adult rodents with either acute or chronic naris occlusion show little decrement in olfactory ability. In this study the effect of naris occlusion coupled with ipsilateral or contralateral olfactory bulb deafferentation on odor-guided behaviors was tested in neonatal mice. Animals that received bilateral bulbectomy or control manipulation were also included. In Experiment 1, olfactory lesions were produced by bulb aspiration on the second day
