Characterization of the effects of binary metal mixtures on short-term uptake of Ag, Cu, and Ni by rainbow trout (Oncorhynchus mykiss).

Single metal Biotic Ligand Models (BLMs) have been developed for a number of metals and model organisms. While these BLMs improve our ability to regulate metals in the aquatic environment, in reality, organisms are often simultaneously exposed to metal mixtures. Recently, several attempts have been made to develop mixture BLMs (mBLMs). Some of these models assume competitive interactions between all metals, while others assume only metals with a similar mode of action (e.g., Na+ or Ca2+ antagonists) will competitively interact. To begin testing these assumptions in the mBLM framework, standard

Spatial Distribution of Gyrodactylus salmonis (Monogenea) on the Body of Captive Fingerling Oncorhynchus mykiss, Including Attachment Within the Olfactory Chamber.

Gyrodactylus salmonis is a common ectoparasite on the fins and body of North American salmonids in fresh water. In this study, the spatial distribution of G. salmonis on 60 captive hatchery-reared rainbow trout, Oncorhynchus mykiss , is reported. The highest parasite densities occurred on 5 × 5-mm(2) sections of the dorsal fin followed by the trunk, other fins, and the olfactory chamber, with the lowest densities on the head. The finding of infections within the olfactory chamber of 93% of the fish was unexpected. One possibility is that such infections represented spillover from high-density …

The adjuvant effect of low frequency ultrasound when applied with an inactivated Aeromonas salmonicida vaccine to rainbow trout (Oncorhynchus mykiss).

Vaccine adjuvants are classified according to their properties of either inducing the persistence of antigens within the animal after immunisation and/or activation of the animal's immune response. The adjuvant effect of low intensity low frequency sonophoresis (LFS) was tested in rainbow trout using an Aeromonas salmonicida bacterin vaccine administered by immersion vaccination using LFS at 37 kHz. The adjuvant effect obtained with LFS was compared with that of normal immersion or intraperitoneal injection vaccination. Quantitative PCR was used to measure bacterial DNA in vaccinated fish up t

Characterization of Weissella ceti infections in Brazilian rainbow trout, Oncorhynchus mykiss (Walbaum), farms and development of an oil-adjuvanted vaccine.

Weissella ceti is an emerging bacterial pathogen that affects rainbow trout, Oncorhynchus mykiss (Walbaum), farms. The aims of this study were to genotype W. ceti strains isolated from distinct geographical origins and to determine the efficacy of an oil-adjuvanted vaccine against the disease. Between 2010 and 2012, outbreaks were recorded in five Brazilian farms, and 34 W. ceti isolates were genetically characterized by repetitive extragenic palindromic PCR, enterobacterial repetitive intergenic consensus sequences PCR and pulsed-field gel electrophoresis. Two different W. ceti vaccines were …

The in vitro blood-O(2) affinity of triploid rainbow trout Oncorhynchus mykiss at different temperatures and CO(2) tensions.

Blood-O(2) affinity, Hill number and predicted arterial-O(2) saturation did not differ between diploid (2N) and triploid (3N) rainbow trout Oncorhynchus mykiss blood when compared under various temperature and CO(2) partial pressure combinations. These results suggest that reduced hypoxia and warm-water tolerance and aerobic capacity of 3N fishes are not due to altered blood-O(2) affinity. Further investigation into O(2) transport-diffusion capacity and intracellular pH within 3N fishes may further unravel the mechanisms behind reduced 3N tolerance of suboptimal environments.

Influence of acclimation and cross-acclimation of metals on acute Cd toxicity and Cd uptake and distribution in rainbow trout (Oncorhynchus mykiss).

The development of chronic metal toxicity models for fresh water fish is complicated by the physiological adjustments made by the animal during exposure which results in acclimation. This study examines the influence of a pre-exposure to a chronic sublethal waterborne metal on acclimation responses as well as the uptake and distribution of new metal into juvenile rainbow trout. In one series of tests, trout were exposed to either 20 or 60 microg/L Cu, or 150 microg/L Zn for a month in moderately hard water and then cross-acclimation responses to Cd were measured in 96 h LC(50) tests. Cu expose

Purification and Characterization of Novel Collagen Peptides from Oncorhynchus mykiss Inhibiting Platelet Aggregation, and the Underlying Mechanism via Molecular Docking.

Rainbow trout is produced in large quantities worldwide, generating substantial processing by-products, among which collagen-rich skin and bone require high-value utilization. Rainbow trout collagen peptides potentially exhibit antiplatelet activity based on structural similarity to reported antiplatelet fish collagen peptides, while their active peptide sequences and underlying mechanisms require investigation. In this study, rainbow trout skin collagen was hydrolyzed by neutral protease, yielding a hydrolysate with 70.52% inhibition of platelet aggregation at 4 mg/mL. The fraction with high

Fluoride exposure affects development, thyroid-related gene expression, and follicle histology in early life stages of rainbow trout (Oncorhynchus mykiss).

Endocrine-disrupting chemicals (EDCs) are attracting growing interest due to their effects on humans and on ecosystems. Among suspected EDCs, fluoride is a widespread pollutant in aquatic ecosystems, particularly as a result of fluoridation of drinking water. It has also been described to affect fish development. The aim of this study was to determine if chronic fluoride exposure could affect early life stages of rainbow trout (Oncorhynchus mykiss) especially their growth and thyroid hormone system. Trout embryos were exposed from 6 to 40 days post-fertilization, to 0, 1.5, 5, 17, 27.5, or 51.…

Ultrastructural Characteristics of the Juvenile Chum Salmon (Oncorhynchus keta) Cerebellum: Interneuron Composition, Neuro-Glial Interactions, Homeostatic Neurogenesis, and Synaptic Plasticity.

Currently, the problem of climate change on Earth is becoming increasingly urgent. These changes are the reason for the increasingly pronounced adaptive differences in different species of fish. A significant gap in ultrastructural data on the organization of the salmon cerebellum was the main motivation for this study's microscopic and ultrastructural analyses using transmission and scanning electron microscopy of the cerebellum of juvenile chum salmon Oncorhynchus keta. The study of the interneuron composition of the cerebellum showed the presence of stellate cells in the molecular layer, pr…

Ultrastructural examination of the host inflammatory response within gills of netpen reared chinook salmon (Oncorhynchus tshawytscha) with Microsporidial Gill Disease.

The sequence of host changes following the rupture of spore-laden xenomas of the microsporidian Loma salmonae during Microsporidial Gill Disease of Salmon was deduced from ultrastructural examination of the gills of naturally infected, moribund, chinook salmon from a commercial aquaculture site. The gills contained many stages of parasite development suggesting fish were chronically exposed to the parasite. Intact xenomas were generally found beneath the endothelium in arteries and arterioles and were encapsulated by a layer of collagen containing fibroblasts sometimes joined by desmosomes. Xe

Relationship between exposure duration, tissue residues, growth, and mortality in rainbow trout (Oncorhynchus mykiss) juveniles sub-chronically exposed to copper.

We conducted a 56-day sub-chronic test on the effects of Cu on rainbow trout (Oncorhynchus mykiss) fry at a nominal water hardness of 100 mg l(-1) (as CaCO(3)). Response measures were growth, whole body Cu concentrations, and mortality. Significant mortality was observed in fish exposed to 54.1 microg Cu l(-1) (47.8%) and 35.7 microg Cu l(-1) (11.7%). Growth was dose-dependent over the range of Cu treatments (0-54 microg Cu l(-1)), and was modeled as a function of Cu exposure concentration and exposure duration. Calculated inhibition concentrations (based on change in wet weight through a 56-d…

Molecular characterization and pathogenicity of Klebsiella pneumoniae species complex associated with mass mortality of Oncorhynchus mykiss in the Ladakh trans-himalayas.

Bacterial diseases pose a significant threat to the entire aquaculture system, both in cold and warm-water environments. The Himalayan region has been widely utilised for trout farming. However, this region is presently under tremendous stress due to the outbreak of several diseases, which are responsible for a lot of mortality and have resulted in significant economic loss. In the present study, Klebsiella pneumoniae belonging to the K. pneumoniae species complex was isolated from diseased rainbow trout Oncorhynchus mykiss cultured in the trans-Himalayan region of Ladakh. Infected fish were i…

Viability of probiotic C6-6 in fish feed and ability to inhibit columnaris-causing bacteria in rainbow trout Oncorhynchus mykiss.

Probiotics are increasingly recognized as sustainable alternatives to antibiotics in aquaculture. This study evaluated the viability and disease-mitigating potential of the candidate probiotic Lelliotia amnigena strain C6-6 in rainbow trout Oncorhynchus mykiss. Viability was assessed under varying storage temperatures (-15, 4, and 17°C), time intervals (up to 24 wk), and feed preparation methods. Results showed that colder storage, particularly at -15°C, preserved higher concentrations of C6-6, while viability rapidly declined at 17°C. The viability of C6-6 over time was enhanced in feeds prep…

Effects of nano-selenium on the histopathological changes and gene expression of rainbow trout (Oncorhynchus mykiss) spleen under heat stress.

Rainbow trout(Oncorhynchus mykiss) is a typical cold-water fish often threatened by high summer temperatures. Nano-selenium as a feed additive can improve the antioxidant capacity of the body and relieve stress. In this study, different levels of nano-selenium (0, 5 and 10 mg/kg) were added to the feed of rainbow trout to determine the changes in spleen structure and expression of related genes in rainbow trout at the proper temperature (18℃) and heat stress temperature (24℃). The results showed that the addition of 5 mg/kg nano-selenium had a more obvious protective effect on rainbow trout sp…

A nonfunctional copy of the salmonid sex-determining gene (sdY) is responsible for the "apparent" XY females in Chinook salmon, Oncorhynchus tshawytscha.

Many salmonids have a male heterogametic (XX/XY) sex determination system, and they are supposed to have a conserved master sex-determining gene (sdY) that interacts at the protein level with Foxl2 leading to the blockage of the synergistic induction of Foxl2 and Nr5a1 of the cyp19a1a promoter. However, this hypothesis of a conserved master sex-determining role of sdY in salmonids is challenged by a few exceptions, one of them being the presence of naturally occurring "apparent" XY Chinook salmon, Oncorhynchus tshawytscha, females. Here, we show that some XY Chinook salmon females have a sdY g…
