[Clinico-experimental long-term study on the combined effect of clofibrin and nicotinic acid on abnormal fatty acid patterns in lipid metabolism disorders in the aged].

Seventy-one patients (forty-two males and twenty-nine females aged forty-five to seventy-six) with disturbances of fat metabolism were treated for three years with clofibric acid and nicotinic acid derivatives. Regular gas chromatographic analyses of the composition of cholesterol ester and triglyceride fatty acids in the serum showed an increase of linoleic, linolenic, arachidonic, and eicosane-pentaenoic acids and a decrease of palmitic, palmitoleic, stearic, oleic, and eicosanetrienic acids during treatment. These changes were far more strongly marked than under monotherapy. Possible causes

[The difference of conformation of apoB-100 in lipoproteins of low and very low density. The modified lipoproteins and destructive inflammation in intima of arteries: a lecture].

The formation of ligand occurs in phylogenetically earlier lipoproteins of very low density and later lipoproteins of very low density when apoB- 100 takes active conformation in association with essential polyenoic fatty acids, in form of ethers with alcohol cholesterol, palmitic and oleic triglycerides. In lipoproteins of low density apoB-100-domain-ligand is formed, in lipoproteins of very low density apoE/B-100-ligand is formed. The ligand lipoproteins absorb cells using apoE/B-100 and apoB-100 receptor endocytosis. In cases of excess of palmitic triglycerides. and lipoproteins of very low

Inhibition of NAPDH Oxidase 2 (NOX2) Prevents Oxidative Stress and Mitochondrial Abnormalities Caused by Saturated Fat in Cardiomyocytes.

Obesity and high saturated fat intake increase the risk of heart failure and arrhythmias. The molecular mechanisms are poorly understood. We hypothesized that physiologic levels of saturated fat could increase mitochondrial reactive oxygen species (ROS) in cardiomyocytes, leading to abnormalities of calcium homeostasis and mitochondrial function. We investigated the effect of saturated fat on mitochondrial function and calcium homeostasis in isolated ventricular myocytes. The saturated fatty acid palmitate causes a decrease in mitochondrial respiration in cardiomyocytes. Palmitate, but not the

Characterization of esterase and alcohol dehydrogenase activity in skin. Metabolism of retinyl palmitate to retinol (vitamin A) during percutaneous absorption.

Retinyl palmitate, a widely used ingredient in cosmetic products, is promoted for its beneficial effects on the appearance of skin. Previous studies suggest that enzymes are available in skin to metabolize this ingredient during skin absorption. Esterase activity hydrolyzes retinyl palmitate to retinol (vitamin A), which is oxidized in many tissues to retinoic acid primarily by alcohol dehydrogenase. The activities of esterase and alcohol dehydrogenase were characterized in hairless guinea pig skin by using flow-through diffusion cells and radiolabeled model compounds (methyl salicylate and be

The role of fatty acids on ICSI outcomes: a prospective cohort study.

BACKGROUND: Our objective was to determine the effect of fatty acids (FAs) in serum and follicular fluid (FF) on fertilization and intracytoplasmic sperm injection (ICSI) outcomes. METHODS: One hundred five women aged 18-38 years undergoing ICSI were recruited in this prospective cohort study. oocyte and emberyo quality was morphologically assessed. FAs in serum and FF were analyzed, using gas chromatography-mass spectrometry (GC-MS). RESULTS: The mean number of mature oocytes was associated with serum levels of oleic acid (r = 0.58; P = 0.002). There were negative correlations between metap

A truncated FatB resulting from a single nucleotide insertion is responsible for reducing saturated fatty acids in maize seed oil.

We identified a G-nucleotide insertion in a maize FatB responsible for reducing saturated fatty acids through QTL mapping and map-based cloning and developed an allele-specific DNA marker for molecular breeding. Vegetable oils with reduced saturated fatty acids have signficant health benefits. SRS72NE, a Dow AgroSciences proprietory maize inbred line, was found to contain signficantly reduced levels of palmitic acid and total saturated fatty acids in seed oil when compared to other common inbreds. Using F2 and F3 populations derived from a cross between SRS72NE and a normal inbred SLN74, we ha

Falling amniotic fluid palmitates in diabetic pregnancies.

Palmitic acid levels were measured in samples of amniotic fluid obtained from 15 patients with diabetes. Seven patients had palmitate values which decreased at some time in pregnancy but only one of the infants developed respiratory distress. Eight of the 15 patients had final amniotic fluid palmitate values which were greater than 0.07 mmol/l and one infant developed respiratory distress. The other seven patients had final amniotic fluid palmitate values of 0.07 mmol/l or less and one of the infants developed respiratory distress. The significance of falling amniotic fluid palmitate values is

Differential Lipotoxic Effects of Palmitate and Oleate in Activated Human Hepatic Stellate Cells and Epithelial Hepatoma Cells.

BACKGROUND/AIMS: Nonalcoholic fatty liver disease (NAFLD) progression to fibrosis, cirrhosis and hepatocellular carcinoma, alters the cellular composition of this organ. During late-stage NAFLD, fibrotic and possibly cancerous cells can proliferate and, like normal hepatocytes, are exposed to high concentrations of fatty acids from both surrounding tissue and circulating lipid sources. We hypothesized that primary human activated hepatic stellate cells and epithelial hepatoma (HepG2) cells respond differently to lipotoxic conditions, and investigated the mechanisms involved. METHODS: Primary

Biosynthesis and palmitoylation of endothelial nitric oxide synthase: mutagenesis of palmitoylation sites, cysteines-15 and/or -26, argues against depalmitoylation-induced translocation of the enzyme.

The presence of myristoylated endothelial nitric oxide synthase (eNOS) in cytosolic fractions of bovine aortic endothelial cells (BAEC) suggests that N-myristoylation of eNOS is not sufficient for membrane localization. Therefore, we examined if posttranslational palmitoylation was another molecular signal for the membrane attachment of eNOS. Metabolic labeling showed incorporation of [3H]palmitic acid into the membrane-bound, but not the cytosolic, form of eNOS. Fatty acid analysis demonstrated the labeled fatty acid incorporated into eNOS was palmitate, linked via a hydroxylamine-labile thio

[The titration of double bonds in fatty acids of blood plasma in patients in testing of glucose tolerance].

The article deals with per oral glucose tolerance test applied to 20 patients with arterial hypertension. The blood plasma was analyzed to detect content of individual fatty acids, double bounds, glucose, insulin and metabolites of fatty acids. In patients with different resistance to insulin content of non-etherized fatty acids decreased approximatively up to 3 times. Without insulin resistance secretion of insulin in 2 hours after glucose load increased up to 3 times and content of individual fatty acids decreases in greater extent. Under insulin resistance secretion of insulin increases up

Isolation of myocardial membrane long-chain fatty acid-binding protein: homology with a rat membrane protein implicated in the binding or transport of long-chain fatty acids.

Abnormal myocardial long-chain fatty acid uptake is suspected of being involved in certain types of heart disease, but the mechanism by which the heart takes up long-chain fatty acids remains unclear. The sulfo-N-succinimidyl derivatives of long-chain fatty acids have been reported to undergo covalent binding to a membrane protein and to irreversibly inhibit the transport of long-chain fatty acids by rat adipocytes (Harmon et al., 1991). It has been suggested that the membrane protein bound by these derivatives is a candidate transporter for long-chain fatty acids in adipocytes. However, myoca

Systematic analysis of the insulinotropic and glucagonotropic potency of saturated and monounsaturated fatty acid mixtures in rat pancreatic islets.

BACKGROUND: Blood contains a mixture of different fatty acids (FFAs) with palmitate and oleate as major components whose molar ratio is dependent on dietary habits. Based on the theory of lipotoxicity for the development of type 2 diabetes such variances in the FFA composition might interfere with lipotoxic effects on the endocrine pancreas. METHODS: Using different ratios of FFA mixtures with palmitate and oleate, we have looked at FFA specific effects on the secretion of mature insulin and glucagon in isolated rat pancreatic islets. RESULTS: The insulinotropic potency of the oleate dominat

ANT-Mediated Inhibition of the Permeability Transition Pore Alleviates Palmitate-Induced Mitochondrial Dysfunction and Lipotoxicity.

Hyperlipidemia is a major risk factor for vascular lesions in diabetes mellitus and other metabolic disorders, although its basis remains poorly understood. One of the key pathogenetic events in this condition is mitochondrial dysfunction associated with the opening of the mitochondrial permeability transition (MPT) pore, a drop in the membrane potential, and ROS overproduction. Here, we investigated the effects of bongkrekic acid and carboxyatractyloside, a potent blocker and activator of the MPT pore opening, respectively, acting through direct interaction with the adenine nucleotide translo

IRON DEFICIENCY PROMOTES HELICOBACTER PYLORI-INDUCED GASTRIC CARCINOGENESIS BY ENABLING RECIPROCITY BETWEEN CARCINOGENIC SECONDARY BILE ACIDS AND PROTECTIVE LONG-CHAIN FATTY ACIDS.

Helicobacter pylori (Hp) is the strongest known risk factor for gastric adenocarcinoma. We previously demonstrated that iron deficiency, a condition of the exposome, augments Hp-induced carcinogenesis. To define mechanisms driving this phenotype, we performed targeted metabolomics in mice which revealed that Hp significantly increased levels of deoxycholic acid (DCA), a carcinogenic secondary bile acid, exclusively under iron-deficient conditions. Further, DCA directly promoted Hp-induced dysplasia. Because bile acids and fatty acids can exert opposing effects on disease, fatty acids were also

Free and bound tryptophan in the blood of depressives.

In in vitro studies, the influence of pH and the concentration of albumin on the binding of tryptophan to this protein is shown. Addition of palmitic acid to human plasma decreases specifically bound and increases free tryptophan. In separate experiments, free and bound tryptophan, tyrosine, free fatty acids (NEFA) and albumine were studied in healthy volunteers and depressives treated by sleep deprivation.

Hypoxia Potentiates Palmitate-induced Pro-inflammatory Activation of Primary Human Macrophages.

Pro-inflammatory cytokines secreted by adipose tissue macrophages (ATMs) contribute to chronic low-grade inflammation and obesity-induced insulin resistance. Recent studies have shown that adipose tissue hypoxia promotes an inflammatory phenotype in ATMs. However, our understanding of how hypoxia modulates the response of ATMs to free fatty acids within obese adipose tissue is limited. We examined the effects of hypoxia (1% O2) on the pro-inflammatory responses of human monocyte-derived macrophages to the saturated fatty acid palmitate. Compared with normoxia, hypoxia significantly increased p
