Parvovirus B19 is a common infection in adults and children. There are reports of secondary parvovirus infection in immunocompromised persons, but no reports of symptomatic secondary infection in healthy persons. We describe a healthy 39-year-old woman who presented with fever, rash, and arthralgia. Her symptoms were thought most compatible with parvovirus B19 infection, but she reported prior positive parvovirus antibody 2 years earlier during prenatal care. Tests were therefore also sent for HIV, streptococcal infection, hepatitis C, and Lyme disease. Testing revealed both elevated IgG and I…

Following its identification by Yvonne Cossart in 1975, human Parvovirus B19 has been recognized as the causative agent of a wide range of diseases. In childhood, the most common disease is a typical exanthema called "fifth disease". In adults, viral infection may be responsible for fetal loss and for aplastic anaemia in immuno-compromised patients. Because persistent viral infection may induce an autoimmune response, Parvovirus B19 is emerging as an environmental factor linked to the pathogenesis of autoimmunity. As a result of its expanding disease spectrum, Parvovirus B19 is the subject of …

UNLABELLED: Severe hemorrhage complications are rare in idiopathic thrombocytopenic purpura. This pathology is often considered as benign.
CASE REPORT: We report the case of a four-year-old boy presenting a parvovirus B19 idiopathic thrombocytopenic purpura. Despite early and repeated use of intravenous immunoglobulin, the evolution was characterized by the secondary apparition of a cerebral hemorrhage. It was lethal seven days after the initial diagnosis.
CONCLUSION: Parvovirus B19 should be investigated as an etiologic agent of idiopathic thrombocytopenic purpura, using PCR. The unpredicti…

This paper reports the study of outbreaks of an acute exanthematous disease among children of three schools in the municipality of Braga (Portugal). Laboratory tests were performed for five cases, showing that the disease was not due to infection by measles or rubella virus, and infection with parvovirus B19 was confirmed. There were 41 cases in children: 12 in the kindergarten, 17 in the secondary school and 12 in the primary school. There was only one case in a staff member, who worked in the kindergarten. Eight cases were identified among household contacts; two of them were brothers, one f…

Human parvovirus B19 has a strong tissue tropism for erythroid progenitor cells and is a causative agent for anemia. However, it remains unclear whether patients with hematological disorders are at a higher risk of B19 infection. In the present study, we evaluated the prevalence of B19 infection in 278 patients with hematological disorders by indirect antibody capture enzyme-linked immunosorbent assay. Virus inoculation into cell culture (of TF-6, WRL-68, and HL-60) was carried out using serum from patients with positive IgM anti-B19 and was then examined by nested polymerase chain reaction, d…

BACKGROUND: Human parvovirus B19 is responsible for a variety of clinical syndromes, such as erythema infectiosum, non-immune hydrops fetalis, transient aplastic anemia, and arthropathies. HPV is also suspected of playing a role in the pathogenesis of various chronic inflammatory and autoimmune diseases, such as systemic lupus erythematosus, rheumatoid arthritis, Kawasaki disease and multiple sclerosis.
OBJECTIVES: To study the age distribution and clinical presentation of patients hospitalized for human parvovirus B19 infection.
METHOD: We reviewed the case records of all pediatric patients…

OBJECTIVE: We investigated the clinical aspects of adult human parvovirus (HPV) B19 infection.
PATIENTS AND METHODS: We retrospectively reviewed the charts of 21 consecutive patients [4 males, aged 32 to 43 years (average 38.0 years), and 17 females, aged 15 to 43 (average 34.2)] with adult HPV B19 infection who visited our outpatient department between July 1997 and June 1998. All diagnoses of adult HPV B19 infection were based on positive anti-HPV B19 IgM antibody in serum and/or positive HPV B19 DNA in peripheral blood.
RESULTS: The predominant signs and symptoms of the patients were: fev…

Infection with parvovirus B19 is common in children and typically causes mild illness. We report here the case of a 5-year-old girl who died suddenly, 2 weeks after the clinical diagnosis of a parvoviral infection (erythema infectiosum). Microscopic examination of the heart showed severe myocarditis with extensive T-cell and macrophage infiltration. Cultures, serology, and molecular analyses of serum for enteroviridae, adenovirus, influenza, varicella zoster, cytomegalovirus, and herpes simplex viruses were negative. Quantitative polymerase chain reaction (PCR) analysis for parvovirus B19 in p…

OBJECTIVE: To determine whether there is a relationship between parvovirus B19 infection and spontaneous abortion.
METHODS: 105 embryo tissues from spontaneous abortions of unknown cause during 1994-1995 were studied using nested polymerase chain reaction (PCR) assay for parvovirus B19 DNA. As controls 40 fetal tissues were also examined. Among which, 20 were collected from induced abortions, and the others were stillbirths. The positive specimens underwent further study for TORCH infections.
RESULTS: 26 (24.5%) were B19 DNA positive in the case group and 2 (5.0%) in the control group. The d…

Nonradioactive slot blot hybridization assays were established for the detection of porcine parvovirus (PPV), using either a digoxigenin-labeled DNA probe or a biotinylated RNA probe. All probes were prepared from a 3.3-kb Pst1-EcoR1 DNA fragment of the NADL8 isolate of PPV. The sensitivity and specificity of the probes in a slot blot system were evaluated in comparison with a 32P-radiolabeled RNA probe. Using an anti-digoxigenin alkaline phosphatase detection system, at least 1 ng of viral replicative form (RF) DNA, or the equivalent of 100 plaque forming units (PFU) of infectious virus, coul…

The autonomous parvoviruses cause a broad spectrum of acute and chronic infections of animals and man. The discrimination of sites of viral replication from sites of viral sequestration is an important goal in elucidating the pathogenesis of these diseases. It is possible to employ strand-specific RNA hybridization probes in such analyses because a 'plus' sense probe will react with single stranded virion DNA and duplex replicative form DNA, but a 'minus' sense probe will react preferentially with obligate replicative intermediates (duplex replicative form DNA and mRNA). Strand-specific RNA hy…

Canine parvovirus type 2 (CPV-2) is a highly contagious pathogen responsible for severe gastroenteritis in carnivores, particularly in dogs. Continuous antigenic evolution has resulted in the emergence of CPV-2a, CPV-2b, and CPV-2c variants, defined by amino acid substitutions in the VP2 capsid protein. In this study, genetically altered CPV-2 viruses carrying common VP2 mutations (S297A, V300G, D305Y, Y324I, N426D, N426E, and T440A) were generated to evaluate their effects on viral behavior in vitro. Mutations were individually introduced into an infectious backbone (strain 447), classified a…

Two feral cats (from the same colony) were presented to the veterinary clinic for weakness, weight loss, and anorexia. The cats were part of a study on feline hepatotropic viruses (collection A, 43 animals). On metaviromic investigation, parvoviral reads were identified in the sera of the two cats. The feline parvovirus genome was 5.3 kb long with an organization similar to other members of the Erythroparvovirus genus. In the ORF1 (nonstructural proteins) and ORF2 (VP1/VP2 precursor) the feline virus displayed 43.1% and 49.1% nucleotide identity to human parvovirus B19, and 48.9% and 56.6% to …

Parvovirus B19, a member of the Parvoviridae family, is a human pathogenic virus. It can be transmitted by respiratory secretions, hand-to-mouth contact, blood transfusion, or transplacental transmission. Most patients are asymptomatic or present with mild symptoms such as erythema infectiosum, especially in children. In rare cases, moderate-to-severe symptoms may occur, affecting blood cells and other systems, resulting in anemia, thrombocytopenia, and neutropenia. Non-immune pregnant women are at risk for fetal infection by parvovirus B19, with greater complications if transmission occurs in…
