[Risk factors associated with neural tube defects: exposure during the first trimester of gestation].

The neural tube defects (NTD) are a group of malformations of multifactorial etiology. Their high incidence in Mexico and the etiologic heterogeneity observed in several studies, prompted the present investigation with the main objective of looking for risk factors associated to NTD. We analyzed maternal exposure during the first trimester of pregnancy to different environmental factors, such as acute or chronic illnesses, immunizations, smoking, alcoholism, maternal or paternal occupation and exposure to chemicals. The sample include 360 patients with anencephaly, 249 with spina bifida and 44

Short-term parental exposure to diazepam disrupts development, behavior, and transcriptional profiles in zebrafish offspring.

Diazepam (DZP) is a pervasive emerging contaminant known to induce significant sublethal adverse effects in aquatic species. However, the intergenerational impacts of parental DZP exposure on fish offspring remain poorly understood. In this study, we exposed zebrafish (F0) to DZP at 8 μg/L for 7 days and investigated variation in the development, behavior, and transcriptional profiles of their offspring (F1) reared in DZP-free medium. The results suggest that DZP accumulates in F0 ovaries and is vertically transmitted to F1 embryos, resulting in-ovo exposure. The parental DZP exposure signific

Paternal Folic Acid Supplementation Alleviated Hepatic Steatosis in Male Offspring Through Sperm DNA Methylation Modification.

In recent years, studies have demonstrated a link between paternal preconception nutrient exposure and the phenotype of the offspring. However, there is a paucity of direct scientific evidence on the effects and the underlying mechanism of paternal folic acid supplementation (PFAS) on offspring phenotype. In this study, male mice were fed different doses of folic acid for 10 weeks, and then mated with females to produce offspring. In our findings, male offspring of PFAS fathers exhibit lower body weight and reduced hepatic lipid accumulation. Moreover, liver lipidomics analysis indicates the b

Monitoring spontaneous and induced human mutation by RAPD-PCR: a response to Weinberg et al. (2001).

Weinberg et al. (2001, Proc. R. Soc. Lond. B 268, 1001-1005) have recently reported a major increase in mutation rate in the children of Chernobyl liquidators as a result of their fathers' exposure to ionizing radiation. If correct, this would provide dramatic evidence for radiation-induced mutation in humans, and would raise major concerns over the genetic effects of radiation. However, mutants were mainly detected using random amplified polymorphic DNA-PCR, an unreliable technology. These mutants were not validated and had no obvious molecular basis. They may, instead, have arisen as PCR art

Age-Dependent Impact of Fluoroscopic Radiation on the Gender of Off-Spring: An International Survey of Cardiologists.

Background: Fluoroscopic radiation has been implicated in reducing the sex ratio (M:F) by potentially damaging the Y chromosome. We examined the effects of exposure to fluoroscopic radiation on gender of offspring of cardiologists across the world. Methods: An internet based survey was e-mailed worldwide to 8000 physicians who practice invasive electrophysiology and/or interventional cardiology. Survey questions included age, race, sub-specialty, hours of exposure to radiation, number of children, gender of off-spring, miscarriages and mutations and exposure to radiation prior to conception of

Spontaneous Abortions of Known Karyotype Related to Occupational and Environmental Factors: A Case-Referent Study.

The aim of the study was to examine the roles of occupational and environmental factors in relation to spontaneous abortions with known karyotypes. A case-referent study was performed among 793 women hospitalized for spontaneous abortion and 808 referents attending ultrasound screening at 17-18 weeks of pregnancy. Chromosomal analysis were obtained for 371 abortuses and 679 referent newborns. Significantly elevated odds ratios (ORs) for abortions with abnormal chromosomes were found for maternal psychological strain at work (OR:1.8), exposure to anesthetic gases (OR:6.9), video display unit wo

Intergenerational sex-specific transcriptomic alterations in gonads of mice after parental lead exposure.

Lead (Pb) exposure is known to influence gene expression across multiple tissues, potentially affecting germline biology and offspring devel … ing alterations in immune‑related and enzymatic pathways. In contrast, males with both maternal and paternal exposure showed the fewest DEGs, though a greater proportion were located on the Y chromosome. These sex‑specific transcriptional patterns, particularly those observed in the offspring of exposed sires, suggest that Pb‑related molecular effects may arise ind …

Parental phenols exposure and spontaneous abortion in Chinese population residing in the middle and lower reaches of the Yangtze River.

Widespread use of phenols has led to ubiquitous exposure to phenols. In experimental animals, phenols increased resorptions, reduced live litter size and fetal body weights. However, there are limited epidemiological evidences of the relationships between exposure to phenols and pregnancy outcomes. We evaluated the associations between parental urinary levels of various phenols and spontaneous abortion in a Chinese population residing in the middle and lower reaches of the Yangtze River. A case-control study was conducted that included 70 case couples with medically unexplained spontaneous abo

Parental occupational exposure to potential endocrine disrupting chemicals and risk of hypospadias in infants.

OBJECTIVES: Hypospadias is a urogenital birth defect affecting infant boys. Periconceptual parental occupational exposure to endocrine disrupting chemicals (EDCs) with oestrogenic or anti-androgenic properties may adversely affect male genital development in utero. The aim of this study was to investigate the association between both maternal and paternal occupational exposures to EDCs and hypospadias. METHODS: This registry-based case-control study considered 1202 cases of hypospadias in children born in Western Australia between 1980 and 2000 and 2583 male controls randomly selected from bi

Associations of maternal weight status with the risk of offspring atopic dermatitis and wheezing by 1 year of age.

BACKGROUND: Maternal pre-pregnancy body mass index (BMI) and gestational weight gain (GWG) have been linked to offspring allergic disorders. However, associations observed in previous studies were inconsistent and might be confounded by unmeasured familial factors. We aimed to examine the associations of maternal weight with offspring allergic disorders by using paternal BMI as a negative control exposure. METHODS: We included the data of 10,522 children from the Born in Guangzhou Cohort Study, 2012-2017. Data on maternal weight were obtained from questionnaires and obstetric records, and pat

How do lifestyle and environmental factors influence the sperm epigenome? Effects on sperm fertilising ability, embryo development, and offspring health.

Recent studies support the influence of paternal lifestyle and diet before conception on the health of the offspring via epigenetic inherita … ability to fertilise oocytes, possibly via a differentially regulated MAKP81IP3 signalling pathway. Paternal exposure to toxic endocrine-disrupting chemicals (EDCs) is also linked to the transgenerational transmission of increased predisposition to disease, infertility, testicular disorders, obesity, and polycystic ovarian syndrome (PCOS) in females through epigene …

Risk of attention deficit hyperactivity and autism spectrum disorders among the children of parents with autoimmune diseases: a nationwide birth cohort study.

Studies have suggested that maternal autoimmune diseases are associated with an increased risk of attention deficit hyperactivity disorder (ADHD) and autism spectrum disorder (ASD). However, research on the association of paternal autoimmune diseases with ADHD and ASD risk has remained inconclusive. Using the Taiwan National Health Insurance Research Database, we selected 708,517 family triads (father-mother-child) between 2001 and 2008 and followed them until the end of 2011. Parental autoimmune diseases as well as ADHD and ASD in children were identified during the study period. Increased AD

Progesterone and estrogen responsiveness to father-toddler interaction.

OBJECTIVES: We assessed the responsiveness of salivary progesterone (P4) and estradiol (E2) to father-child interaction, including testing for differences in short-term hormonal change based on paternal characteristics. We also predicted that P4 exposure during the study period would relate positively to post-interaction paternal mood. METHODS: We conducted an in-home intervention study in which fathers (n = 44) played with their toddlers. Subjects provided saliva samples before interacting with their children, with additional collections 40 and 70 min later. RESULTS: E2 did not significantl

Sperm aneuploidy in fathers of children with paternally and maternally inherited Klinefelter syndrome.

BACKGROUND: It is unclear whether frequency of sperm aneuploidy is associated with risk of fathering children with trisomy. METHODS: We recruited 36 families with a boy with Klinefelter syndrome (KS), interviewed the fathers about their exposures and medical history, received a semen sample from each father, and collected blood samples from the mother, father and child. We applied a multicolour fluorescent in-situ hybridization assay to compare the frequencies of sperm carrying XY aneuploidy and disomies X, Y and 21 in fathers of maternally and paternally inherited KS cases. RESULTS: Inherit

Parental Alcohol Exposures Associate with Lasting Mitochondrial Dysfunction and Accelerated Aging in a Mouse Model.

Although detrimental changes in mitochondrial morphology and function are widely described symptoms of fetal alcohol exposure, no studies have followed these mitochondrial deficits into adult life or determined if they predispose individuals with fetal alcohol spectrum disorders (FASDs) to accelerate biological aging. Here, we used a multiplex preclinical mouse model to compare markers of cellular senescence and age-related outcomes induced by maternal, paternal, and dual-parental alcohol exposures. We find that even in middle life (postnatal day 300), the adult offspring of alcohol-exposed pa

Nonconceptus Mechanisms of Prenatal Alcohol Exposure That Disrupt Embryo-Fetal Development: An Integrative View.

PURPOSE: Prenatal alcohol exposure (PAE) is a leading cause of persistent neurodevelopmental disability, with additional adverse consequence … vailability. This altered metabolism could also reprogram placental and fetal epigenetics, as could paternal exposure to alcohol. Both epigenetic effects and inflammation can impair placental function and modulate the placenta-brain axis that modulates brain development. The review discusses limitations in the current understanding of these mechanisms and highlight …
