Galectin-1 Modulates the Fusogenic Activity of Placental Endogenous Retroviral Envelopes.

Syncytin-1 and -2 are glycoproteins encoded by human endogenous retrovirus (hERV) that, through their fusogenic properties, are needed for the formation of the placental syncytiotrophoblast. Previous studies suggested that these proteins, in addition to the EnvP(b) envelope protein, are also involved in other cell fusion events. Since galectin-1 is a β-galactoside-binding protein associated with cytotrophoblast fusion during placental development, we previously tested its effect on Syncytin-mediated cell fusion and showed that this protein differently modulates the fusogenic potential of Syncy

Plasma placental growth factor as a susceptibility biomarker for longitudinal cognitive change.

INTRODUCTION: Current biomarkers used to assess cerebrovascular pathology largely reflect end-stage disease, and more sensitive biomarkers are needed. We examined the role of baseline placental growth factor (PlGF) as a susceptibility biomarker for the progression of white matter injury and longitudinal cognitive decline. METHODS: A total of 272 functionally intact older adults completed baseline blood draws, with plasma analyzed for PlGF (Meso Scale Discovery), and longitudinal neuroimaging and neuropsychological testing. A subset of 110 participants (n = 110) had banked plasma assayed on a

Does the thyroid-stimulating hormone measured concurrently with first trimester biochemical screening tests predict adverse pregnancy outcomes occurring after 20 weeks gestation?

CONTEXT: Maternal hypothyroidism in early pregnancy is associated with adverse outcomes, but not consistently across studies. First trimester screening for chromosomal anomalies is routine in many centers and provides an opportunity to test thyroid function. OBJECTIVE: To determine if thyroid function tests performed with first trimester screening predicts adverse pregnancy outcomes. DESIGN, PARTICIPANTS AND SETTING: A cohort study of 2411 women in Western Australia with singleton pregnancies attending first trimester screening between 9 and 14 weeks gestation. OUTCOME MEASURES: We evaluate

Vitamin K Biochemistry and Pharmacokinetics: The Basis of Late Vitamin K Deficiency Intracranial Bleeding in Early Infancy.

Vitamin K is a fat-soluble vitamin essential for the activation of vitamin K-dependent proteins involved in coagulation and other physiological processes. Neonates are particularly vulnerable to vitamin K deficiency due to limited placental transfer, low hepatic stores, immature liver function, and insufficient dietary intake, especially in exclusively breastfed infants. This review summarizes the biochemistry and pharmacokinetics of vitamin K, focusing on their role in the pathogenesis of late vitamin K deficiency bleeding (VKDB), including intracranial hemorrhage in early infancy. The limita

A brief overview of preeclampsia.

Preeclampsia (PE) is a leading cause of maternal mortality and morbidity worldwide. It occurs in women with first or multiple pregnancies and is characterized by new onset hypertension and proteinuria. Improper placentation is mainly responsible for the disease. If PE remains untreated, it moves towards more serious condition known as eclampsia. Hypertension, diabetes mellitus, proteinuria, obesity, family history, nulliparity, multiple pregnancies and thrombotic vascular disease contribute as the risk factors for PE. PE triggered metabolic stress causes vascular injury, thus contributing to t

MDIFN: a deep learning-based multi-dimensional interaction fusion network for placental MRI segmentation.

BACKGROUND: The placenta serves as a vital channel for maternal-fetal substance exchange and is a crucial organ for maintaining pregnancy. Placental diseases can severely affect maternal health and fetal growth. Given the limitations of single-dimension segmentation models and traditional fusion strategies in meeting the demands of automatic placental magnetic resonance imaging (MRI) segmentation, this study developed an automatic placental MRI segmentation method based on a multi-dimensional interaction fusion network (MDIFN). METHODS: This study included 267 placental MRI cases that met the

Identifying late-onset fetal growth restriction by measuring circulating placental RNA in the maternal blood at 28 weeks' gestation.

BACKGROUND: Late-onset fetal growth restriction (FGR) is often undetected prior to birth, which puts the fetus at increased risk of adverse perinatal outcomes including stillbirth. OBJECTIVE: Measuring RNA circulating in the maternal blood may provide a noninvasive insight into placental function. We examined whether measuring RNA in the maternal blood at 26-30 weeks' gestation can identify pregnancies at risk of late-onset FGR. We focused on RNA highly expressed in placenta, which we termed "placental-specific genes." STUDY DESIGN: This was a case-control study nested within a prospective c

Placental DNA methylation changes associated with maternal prepregnancy BMI and gestational weight gain.

BACKGROUND: Maternal obesity prior to or during pregnancy influences fetal growth, predisposing the offspring to increased risk for obesity across the life course. Placental epigenetic mechanisms may underlie these associations. We conducted an epigenome-wide association study to identify placental DNA methylation changes associated with maternal prepregnancy body mass index (BMI) and rate of gestational weight gain at first (GWG1), second (GWG2), and third trimester (GWG3). METHOD: Participants of the NICHD Fetal Growth Studies with genome-wide placental DNA methylation (n = 301) and gene ex

Amniotic fluid and umbilical artery levels of sex hormones and prostaglandins in human cocaine users.

Biochemical evaluation of amniotic fluid contents is often used to monitor fetal secretory and excretory functions. To determine whether cocaine use during pregnancy affects fetal endocrine secretions, amniotic fluid and umbilical artery samples were collected at term, and selected gonadal hormones and prostaglandins were assayed. Gestational age, birth weights, and placental weights were recorded. The data showed that birth weights were significantly decreased (P less than 0.001) in cocaine users, but gestational age and placental weights were unaffected. Amniotic fluid levels of androstenedi

Creatine kinase regulates energy metabolism and growth of trophoblasts.

STUDY QUESTION: Does the human placenta utilize the creatine phosphagen system for energy homeostasis during development? SUMMARY ANSWER: Components of the creatine (Cr)-creatine kinase (CK)-phosphocreatine (PCr) system are dynamically expressed by the trophoblast and mesenchymal compartments throughout gestation wherein creatine kinase is required for cellular ATP metabolism, cell cycle, and proliferation of trophoblast cells. WHAT IS KNOWN ALREADY: The Cr-CK-PCr system maintains ATP homeostasis in tissues with high energy demand and is required for proliferation, migration, and invasion of

[Not Available].

This case report describes an atypical progression of pregnancy in a woman who was infected with COVID-19 in her second trimester and initially presented with signs of absent foetal movements. The foetus was diagnosed with severe intrauterine growth restriction and abnormal foetal Doppler flows. The condition slowly improved over months, and the pregnancy was terminated with an acute caesarean section in gestational week 35. Placenta was tested negative for SARS-CoV-2-nucleocapsid. Four days old, the child was found COVID-19-positive, but remained asymptomatic. This case is a rare example cons

Machine learning analysis of MRI-derived texture features to predict placenta accreta spectrum in patients with placenta previa.

PURPOSE: To evaluate whether a machine learning (ML) analysis employing MRI-derived texture analysis (TA) features could be useful in assessing the presence of placenta accreta spectrum (PAS) in patients with placenta previa (PP). The hypothesis is that TA features may reflect histological abnormalities underlying PAS in patients with PP thus helping in differentiating positive from negative cases. MATERIALS AND METHODS: Pre-operative MRI examinations of 64 patients with PP of which 20 positive (12 accreta, 7 increta and 1 percreta) and 44 negative for PAS were retrospectively selected. Multi

TGFβ1 Inhibits Migration, Invasion, and Proliferation of HTR-8/Svneo Cells via tTG-Activated TLR4/NF-κB Signaling Pathway.

TGFβ1 plays a key role in placental development, but its regulation of trophoblast function isn't fully known. Meanwhile, the TLR4/NF-κB pathway is linked to trophoblast dysfunction, yet the mediator linking TGFβ1 to this pathway is unclear. Thus, this study examines if tTG mediates TGFβ1's regulation of trophoblast functions via TLR4/NF-κB and its relevance in preeclampsia (PE). To address this, HTR-8/Svneo cells were treated with TGFβ1, cell migration, invasion, and proliferation were assessed via Transwell and CCK-8 assays. The expression of tTG, TLR4/NF-κB pathway components, and pro-infla

High-Dose Vitamin D Supplementation Alters Tissue-Specific Vitamin D Metabolite Distribution in Mice During Pregnancy.

BACKGROUND: Pregnancy is a major modifier of vitamin D metabolism, which is suggested to serve an immunomodulatory role, support placental function, and aid fetal development. However, little is known about local tissue concentrations of vitamin D metabolites following supplementation during gestation. OBJECTIVES: We aimed to quantify intracellular concentrations of 25-hydroxyvitamin D3 (25[OH]D) and the bioactive 1,25-dihydroxyvitamin D3 (1,25[OH]2D) across tissues following vitamin D supplementation and to determine how this distribution is altered during gestation in mice. METHODS: Forty

[Diagnostic efficacy of biophysical tests and cerebral-umbilical index when assessing fetal oxygenation].

INTRODUCTION: Perinatal morbidity and mortality are the ultimate indicators of antenatal care today, whose responsible task is to assess the respiratory function of the placenta, fetal growth and placental maturation in order to provide conditions for the delivery of a living and viable newborn. The diagnostic procedures of antenatal care tested within this study were the biophysical tests of cardiotocography and the fetal biophysical profile, along with the colour doppler evaluation of the cerebral-umbilical ratio. The objective of this study was to determine the most effective diagnostic pro

Transient diabetes insipidus in pregnancy.

UNLABELLED: Gestational diabetes insipidus (DI) is a rare complication of pregnancy, usually developing in the third trimester and remitting spontaneously 4-6 weeks post-partum. It is mainly caused by excessive vasopressinase activity, an enzyme expressed by placental trophoblasts which metabolises arginine vasopressin (AVP). Its diagnosis is challenging, and the treatment requires desmopressin. A 38-year-old Chinese woman was referred in the 37th week of her first single-gestation due to polyuria, nocturia and polydipsia. She was known to have gestational diabetes mellitus diagnosed in the se

Mitochondrial-targeted MitoQ attenuates PFOS-induced vasoconstriction and cAMP suppression in human placental arteries†.

Perfluorooctanesulfonic acid (PFOS), a persistent environmental pollutant, is elevated in the plasma of preeclamptic women and may contribute to gestational hypertension. However, its direct effects on placental vascular function remain unclear. This study investigated PFOS' impact on vascular reactivity in human placental chorionic plate arteries (CPAs) and evaluated the therapeutic potential of the mitochondrial-targeted antioxidant mitoquinone (MitoQ). CPAs from normotensive pregnancies were treated with PFOS (10 μM) or vehicle for 24 h. Wire myography assessed contractile responses to KCl

Acute Placental Inflammation Is Associated with Reduced Progesterone Receptor Density in the Basal Decidua in Spontaneous Preterm Birth.

A functional progesterone withdrawal has been thought to contribute to the pathophysiology of spontaneous preterm birth (PTB). The density of the progesterone receptor (PR) in gestational tissues could play a role in functional progesterone withdrawal. We sought to understand the relationship between placental pathology and the density of PR in the basal decidua in the setting of spontaneous preterm delivery. This is a secondary analysis of a retrospective cohort study of 40 patients with spontaneous PTB < 37 weeks from a prior study at NorthShore University HealthSystem previously described.
