Genome-Wide Association Mapping of Ear Traits in Early-Maturing Maize Under Contrasting Planting Densities.

Developing early-maturing maize varieties with improved tolerance to high-density planting is essential for enhancing yield in China. In this study, a unique panel of 498 cold-adapted early-maturing inbred lines were evaluated for ear length (EL) and ear diameter (ED) across two locations under contrasting planting densities in one growing season. Phenotypic analysis revealed abundant phenotypic variation in these ear traits among the tested lines, and there were significant correlations between environments. A total of 406,897 high-quality single nucleotide polymorphism markers (SNPs) were ob

Metabolic analysis of MYB30 that regulates iron deficiency stress in Arabidopsis.

INTRODUCTION: Iron is an essential microelement for animals, humans, and plants. Notably, approximately one-third of the world's soils are alkaline, leading to iron deficiency. Therefore, understanding the mechanism of iron absorption and transport in plants is crucial for improving iron bioavailability in crops. METHODS: In this research, reverse genetics was used to identify the transcription factor MYB30 as a positive regulator of the plant response to iron deficiency. RESULTS AND DISCUSSION: Phenotype analysis demonstrated that MYB30 mutant plants were sensitive to iron deficiency, exhib

Physiological and metabolic reprogramming under stress combination in plants.

In nature, plants are rarely challenged by a single, isolated, environmental stress factor. Instead, they continuously experience a dynamic range of abiotic and biotic constraints, generating conditions of multifactorial stress combination (MFSC). Driven by anthropogenic activities, many stressors are increasing in their frequency, intensity, and duration, exposing plants to complex conditions that go well beyond two interacting factors. A recent conceptual advance is the recognition that plant responses to MFSC cannot be predicted by simply summing up plant responses of the different individu

Where does Aedes albopictus (Diptera: Culicidae) really breed in a Mediterranean residential area? Results from a field study in Valencia, Eastern Spain.

Since its introduction in Spain in 2004, Aedes albopictus has rapidly spread across the country. Its aggressive biting behaviour causes nuisance, limiting outdoor activities. Also, its role as a vector of several arboviruses implies a major public health risk, with several cases of autochthonous dengue having been reported nationwide over the past few years. Control strategies usually focus on interventions in breeding sites. As such, accurate knowledge of the main larval habitats becomes a major priority in infested areas. A detailed identification of breeding sites of Ae. albopictus was carr

The MYB transcription factor APL is a rational target for base editing to engineer flowering time.

Achieving optimal flowering time is a pivotal factor in precision plant breeding. Here, in maize, we characterized an EMS-induced additive delayed-flowering mutant, edf1. edf1 and its heterozygotes are late-flowering under both long- and short-day photoperiods in the field, and harbor a splice-donor mutation in ALTERED PHLOEM DEVELOPMENT (APL). This mutation leads to the accumulation of the minor APL-β transcript, a splice variant expressed across a large natural maize population. Expression of the major splice variant APL-α is associated with flowering initiation; APL knockdown delays floweri…

The revelation of genomic breed composition using target capture sequencing: a case of Taxodium.

Taxodium plants have good flood tolerance and thus were introduced into China from North America in the early 1900s. The subsequent decades of cross-breeding experiments within Taxodium have produced many new hybrid cultivars in China while also creating confusion in the genetic background of Taxodium plants. In the present study, target capture sequencing-derived SNP markers were used to reveal the genomic composition of different species and cultivars of Taxodium. The results unraveled the phylogenetic relationship within Taxodium and suggested the possibility of recent interspecific natural

Lepidopteran pests and phytopathogenic fungi: The overlooked interface of insect‒microbe interactions.

Lepidopteran pests and phytopathogenic fungi are major biotic threats to crop health and productivity, yet their interactions remain poorly integrated into current frameworks of insect-microbe and plant disease ecology. Lepidopteran pests and fungal pathogens often share common ecological niches on host plants, particularly damaged tissues and microbe-rich interfaces. Chewing damage, frass deposition and movement by lepidopteran pests can create opportunities for fungal dispersal and invasion. This review synthesizes current knowledge on lepidopteran pest-phytopathogenic fungus-plant (LPP) tri

Analysis of Phytohormone Signal Transduction in Sophora alopecuroides under Salt Stress.

Salt stress seriously restricts crop yield and quality, leading to an urgent need to understand its effects on plants and the mechanism of plant responses. Although phytohormones are crucial for plant responses to salt stress, the role of phytohormone signal transduction in the salt stress responses of stress-resistant species such as Sophora alopecuroides has not been reported. Herein, we combined transcriptome and metabolome analyses to evaluate expression changes of key genes and metabolites associated with plant hormone signal transduction in S. alopecuroides roots under salt stress for 0

Durum Wheat Anther Culture: a Complete Protocol.

Durum wheat is one of the most recalcitrant species among cereals for anther culture, and is characterized by a low embryogenesis rate and a high percentage of albinism (95%). This chapter describes key features of durum wheat anther culture protocol. This includes growing donor plants under the most appropriate conditions, selecting spikes with microspores at the mid- to late-uninucleate stage of development, establishing aseptic conditions for dissection, and culturing anthers in appropriate media and conditions. A modified W14 medium is described that performs well in durum wheat anther cul

[Identification and expression analysis of TCP family members in tobacco (Nicotiana tabacum L.)].

TCP family as plant specific transcription factor, plays an important role in different aspects of plant development. In order to screen TCP family members in tobacco, the homologous sequences of tobacco and Arabidopsis TCP family were identified by genome-wide homologous alignment. The physicochemical properties, phylogenetic relationships and cis-acting elements were analyzed by bioinformatics. The homologous genes of AtTCP3/AtTCP4 were screened, and RT-qPCR was used to detect the changes of gene expression upon 20% PEG6000 treatment. The results show that tobacco contains 63 TCP family memb

A Glu209Lys substitution in DRG1/TaACT7, which disturbs F-actin organization, reduces plant height and grain length in bread wheat.

Plant height and grain size are two important agronomic traits that are closely related to crop yield. Numerous dwarf and grain-shape mutants have been studied to identify genes that can be used to increase crop yield and improve breeding programs. In this study, we characterized a dominant mutant, dwarf and round grain 1 (drg1-D), in bread wheat (Triticum aestivum L.). drg1-D plants exhibit multiple phenotypic changes, including dwarfism, round grains, and insensitivity to brassinosteroids (BR). Cell structure observation in drg1-D mutant plants showed that the reduced organ size is due to ir

TaACTIN7-D regulates plant height and grain shape in bread wheat.

Exploitation of new gene resources and genetic networks contributing to the control of crop yield-related traits, such as plant height, grain size, and shape, may enable us to breed modern high-yielding wheat varieties through molecular methods. In this study, via ethylmethanesulfonate mutagenesis, we identify a wheat mutant plant, mu-597, that shows semi-dwarf plant architecture and round grain shape. Through bulked segregant RNA-seq and map-based cloning, the causal gene for the semi-dwarf phenotype of mu-597 is located. We find that a single-base mutation in the coding region of TaACTIN7-D

Comparative analysis of salicylic acid levels and gene expression in resistant, tolerant, and susceptible cassava varieties following whitefly-mediated SLCMV infection.

Sri Lankan cassava mosaic virus (SLCMV), the primary pathogen responsible for cassava mosaic disease in cassava plantations, is transmitted via infected cutting stems and the whitefly vector, Bemisia tabaci. To obtain better insights into the defense mechanism of cassava against SLCMV, whiteflies were used to induce SLCMV infection for activating the salicylic acid (SA) signaling pathway, which triggers the innate immune system. The study aimed to investigate the specific interactions between viruliferous whiteflies and SA accumulation in resistant (C33), tolerant (Kasetsart 50; KU50), and sus

Identification of microRNAs involved in the Phosphate starvation response in Oil Palm (Elaeis guineensis Jacq.).

BACKGROUND: Plant microRNA, often known as miRNA, is a novel form of gene expression regulator that is known to play a significant role in phosphate starvation. The identification of microRNAs involved in the response to phosphate starvation in oil palms is beneficial for breeding programs. METHOD: The main nursery stage seedlings of two oil palm progenies were treated with three different fertiliser namely: complete fertiliser with urea, P2O5, K2O, and MgO based on the standard procedure as a control (C); fertiliser with urea, K2O, MgO without P2O5 (P0); and no fertiliser (F0) for 24 weeks.

Heterotic grouping of provitamin A-enriched maize inbred lines for increased provitamin A content in hybrids.

BACKGROUND: The establishment of heterotic groups of inbred lines is crucial for hybrid maize breeding programs. Currently, there is no information on the heterotic patterns of the Provitamin A (PVA) inbred lines developed in the maize improvement program of the International Institute of Tropical Agriculture (IITA) to form productive PVA enriched hybrids for areas affected by vitamin A deficiency. This study assessed the feasibility of classifying PVA-enriched inbred lines into heterotic groups based on PVA content without compromising grain yield in hybrids. Sixty PVA inbred lines were cross

Incorporating male sterility increases hybrid maize yield in low input African farming systems.

Maize is a staple crop in sub-Saharan Africa, but yields remain sub-optimal. Improved breeding and seed systems are vital to increase productivity. We describe a hybrid seed production technology that will benefit seed companies and farmers. This technology improves efficiency and integrity of seed production by removing the need for detasseling. The resulting hybrids segregate 1:1 for pollen production, conserving resources for grain production and conferring a 200 kg ha-1 benefit across a range of yield levels. This represents a 10% increase for farmers operating at national average yield le

The chemical composition of barley grain (Hordeum vulgare L.) landraces from the Canary Islands.

The proximate composition, total phenolics, antioxidant activity, minerals, and trace elements were determined in 42 barley landraces in order to highlight their nutritional potential and promote their cultivation. Two-row barley landraces had a higher average content of starch, protein, total phenolic compounds, and iron, when compared with six-row ones that presented higher mean ash and fiber concentrations. Additionally, the six-row barley landraces with strongly pigmented grains had lower zinc and manganese and higher protein mean concentrations than the whitish or lightly pigmented barley

Overexpression of a Novel Apple NAC Transcription Factor Gene, MdNAC1, Confers the Dwarf Phenotype in Transgenic Apple (Malus domestica).

Plant height is an important trait for fruit trees. The dwarf characteristic is commonly associated with highly efficient fruit production, a major objective when breeding for apple (Malus domestica). We studied the function of MdNAC1, a novel NAC transcription factor (TF) gene in apple related to plant dwarfing. Localized primarily to the nucleus, MdNAC1 has transcriptional activity in yeast cells. Overexpression of the gene results in a dwarf phenotype in transgenic apple plants. Their reduction in size is manifested by shorter, thinner stems and roots, and a smaller leaf area. The transgeni

Creation of the first ultra-low gluten barley (Hordeum vulgare L.) for coeliac and gluten-intolerant populations.

Coeliac disease is a well-defined condition that is estimated to affect approximately 1% of the population worldwide. Noncoeliac gluten sensitivity is a condition that is less well defined, but is estimated to affect up to 10% of the population, and is often self-diagnosed. At present, the only remedy for both conditions is a lifelong gluten-free diet. A gluten-free diet is often expensive, high in fat and low in fibre, which in themselves can lead to adverse health outcomes. Thus, there is an opportunity to use novel plant breeding strategies to develop alternative gluten-free grains. In this…
