Allelopathic Effects and Cytotoxic Mechanism of Juniperus sabina Essential Oil via Inhibition of Cell Division in Model Plant Species.

Juniperus sabina is a dominant species in the desert and mountain ecosystems. There have been reports evaluating its chemical composition and biochemical values, however, investigation on its allelopathic effect which might facilitate its dominance remains largely unexplored, with the major responsible allelochemicals undetermined. In this study, we compared the allelopathic effects of the aqueous extract, ethanol extract, as well as the essential oil (EO) of J. sabina on seed germination and seedling growth of two receiver species, the dicot plant lettuce (Lactuca sativa) and the monocot plan

Human Tyrosinase Displayed on the Surface of Chinese Hamster Ovary Cells for Ligand Fishing of Tyrosinase Inhibitors from Medicinal Plants.

Ligand fishing is a promising strategy for the screening of active ingredients from complex natural products. In this work, human tyrosinase (hTYR) was displayed on the surface of Chinese hamster ovary (CHO) cells for the first time; it was then used as bait to develop a new method for ligand fishing. The localization of hTYR on the CHO cell surface was verified by an enzyme activity test and fluorescence microscopy. The displayed tyrosinase (CHO@hTYR) maintained relatively stable enzymatic activity (82.59 ± 2.70%) within 7 days. Furthermore, it can be reused for fishing five times. Guided by

Pepper heat shock protein 70a interacts with the type III effector AvrBsT and triggers plant cell death and immunity.

Heat shock proteins (HSPs) function as molecular chaperones and are essential for the maintenance and/or restoration of protein homeostasis. The genus Xanthomonas type III effector protein AvrBsT induces hypersensitive cell death in pepper (Capsicum annuum). Here, we report the identification of the pepper CaHSP70a as an AvrBsT-interacting protein. Bimolecular fluorescence complementation and coimmunoprecipitation assays confirm the specific interaction between CaHSP70a and AvrBsT in planta. The CaHSP70a peptide-binding domain is essential for its interaction with AvrBsT. Heat stress (37°C) an

Liquid-liquid extraction for recovery of paclitaxel from plant cell culture: solvent evaluation and use of extractants for partitioning and selectivity.

A major challenge in the production of metabolites by plant cells is the separation and purification of a desired product from a number of impurities. An important application of plant cell culture is the biosynthesis of the anticancer agent paclitaxel. Liquid-liquid extraction plays a critical role in the recovery of paclitaxel and other valuable plant-derived products from culture broth. In this study, the extraction of paclitaxel and a major unwanted by-product, cephalomannine, from plant cell culture broth into organic solvents is quantified. Potential solvent mixtures show varying affinit

Culture medium and plant growth regulator regimes influence the transcriptome and cannabisin accumulation in Cannabis sativa cell suspension cultures.

Plant cell suspension cultures are a valuable biotechnological platform for the controlled production of high-value phytochemicals. Yet, the molecular mechanisms by which culture medium composition impacts cellular transcriptional programs remain poorly characterised, especially in non-model plant species. Here, we investigated how two culture media, Murashige & Skoog (MS)- and Linsmaier & Skoog (LS)-based, influence growth, gene expression and secondary metabolite production in Cannabis sativa cell suspension cultures derived from a non-commercial genotype, Line 43/178. Cultures were analysed

A highly selective fluorescent probe based on rhodamine derivatives for HClO detection in water, food, plants, live cells and zebrafish.

Hypochlorous acid (HClO) exerts diverse biological functions and has broad applications in daily life. Herein, a novel turn-on fluorescent probe, DCOH-OCl, was developed for HClO with rhodamine derivatives as the fluorescent group and N,N-dimethylthiocarbamate as the reactive group. The probe exhibited excellent performance, including fast response (30 s), wide pH range (3-10), high selectivity, and strong anti-interference capability. DCOH-OCl allowed quantitative determination of HClO in environmental and food samples and fluorescence imaging of HClO in living cells, zebrafish, and various p

The function of the Arabidopsis receptor kinase THESEUS1 in plant cell wall integrity maintenance: from evolutionary origin to future perspectives.

Plants actively monitor the state of their cell walls and adapt their structure and composition as needed. THESEUS1 (THE1), a receptor kinase from the Catharanthus roseus RECEPTOR-LIKE KINASE 1-LIKE (CrRLK1L) family, was the first receptor kinase described to be involved in this maintenance of cell wall integrity in Arabidopsis thaliana. It contributes to the regulation of cell wall stiffness, participates in lateral root development, and modulates production of the stress hormones abscisic acid and jasmonic acid. Besides, it is required for responses to cellulose biosynthesis inhibition, such

Using a cellulose-complementary oligosaccharide as a tool to probe exposed cellulosic surfaces in cotton fibres and growing plant cell walls.

Cellulosic microfibrils in plant cell walls are largely ensheathed and probably tethered by hydrogen-bonded hemicelluloses. Ensheathing may vary developmentally as hemicelluloses are peeled to enable cell expansion. We characterised a simple method to quantify ensheathed versus naked cellulosic surfaces based on the ability to adsorb a radiolabelled 'cellulose-complementary oligosaccharide', [3H]cellopentaitol. Filter-paper (cellulose) adsorbed 40% and >80% of aqueous 5 nM [3H]cellopentaitol within ∼1 and ∼20 h respectively. When [3H]cellopentaitol was rapidly dried onto filter-paper, ∼50% of

Plant cell culture extract of Cirsium eriophorum with skin pore refiner activity by modulating sebum production and inflammatory response.

Facial pore enlargement is considered a significant esthetic and health concern in skincare cosmetics. The pores fulfill the critical function of keeping the skin surface hydrated and protected against microbial infections. The hyperseborrhea, the stress factors, and the hormonal triggers can cause pore size enlargement, causing higher susceptibility of the skin to microbe aggressions and inflammatory reactions. Thus, reducing excessive sebum production and keeping functional pores are two of the most requested activities in skincare cosmetics. A Cirsium eriophorum cell culture extract was inv

PLANT NATRIURETIC PEPTIDE A and Its Putative Receptor PNP-R2 Antagonize Salicylic Acid-Mediated Signaling and Cell Death.

The plant stress hormone salicylic acid (SA) participates in local and systemic acquired resistance, which eventually leads to whole-plant resistance to bacterial pathogens. However, if SA-mediated signaling is not appropriately controlled, plants incur defense-associated fitness costs such as growth inhibition and cell death. Despite its importance, to date only a few components counteracting the SA-primed stress responses have been identified in Arabidopsis (Arabidopsis thaliana). These include other plant hormones such as jasmonic acid and abscisic acid, and proteins such as LESION SIMULATI

Rice SDSFL1 plays a critical role in the regulation of plant structure through the control of different phytohormones and altered cell structure.

Semi-dwarfism is one of the most important agronomic traits for many cereal crops. In the present study, a mutant with semi-dwarf and short flag leaf 1, sdsfl1, was identified and characterized. The sdsfl1 mutant demonstrated some distinguished structural alterations, including shorter plant height and flag leaf length, increased tiller numbers and flag leaf width, and decreased panicle length compared with those of wild type (WT). Genetic analysis suggested that the mutant traits were completely controlled by a single recessive gene. The SDSFL1 gene was mapped to the long arm of chromosome 3

Comparative analysis of whole plant, flower and root extracts of Chamomilla recutita L. and characteristic pure compounds reveals differential anti-inflammatory effects on human T cells.

INTRODUCTION: Chronic inflammation is a hallmark of chronic wounds and inflammatory skin diseases. Due to a hyperactive and prolonged inflammation triggered by proinflammatory immune cells, transitioning to the repair and healing phase is halted. T cells may exacerbate the proinflammatory milieu by secreting proinflammatory cytokines. Chamomilla recutita L. (chamomile) has been suggested for use in several inflammatory diseases, implying a capability to modulate T cells. Here, we have characterized and compared the effects of differently prepared chamomile extracts and characteristic pure comp

Flavone polyphenols dominate in Thymus schimperi Ronniger: LC-ESI-MS/MS characterization and study of anti-proliferative effects of plant extract on AGS and HepG2 cancer cells.

Thymus schimperi is a highly localized and a rare plant endemic to Ethiopia. An optimized and validated high-performance liquid chromatography-electrospray ionization-tandem mass spectrometry (HPLC-ESI-MS/MS) method was applied to characterize 23 polyphenolic compounds found in ethyl acetate extracts of the plant. From those, flavones dominated and luteolin was the major component contributing 21.83% of the total composition (or 46.05±0.59g/kg of fresh sample weight). Validation data showed a determination coefficient (R2)≥0.997. Limits of detection (LOD) and quantification (LOQ) were 0.03-0.9
