Programmed cell death and immunity in plants are finely orchestrated to promote antimicrobial defense while preventing autoimmunity. However … uld substantially exacerbate the cell death and enhance disease resistance, implying their roles in plant immunity. Our findings provide novel insights into the regulatory mechanisms of ECDR1 in cell death and defense pathways, will aid in understanding the functions of PI3K and PI4K in plant immunity.

The chloroplast, a key organelle for plant immunity, is frequently targeted by viral proteins to suppress host defense. Here, we demonstrate that NSvc4, the movement protein of rice stripe virus (Tenuivirus oryzaclavatae; genus Tenuivirus), functions as a chloroplast-localized virulence effector. We show that NSvc4 enters chloroplasts and directly associates with NbPsbQ, a subunit of the oxygen-evolving complex (OEC) of Photosystem II. This interaction competitively disrupts the binding of NbPsbQ to its native partners NbPsbO and NbPsbP, thereby dampening the accumulation of chloroplast-derive…

Plant mitogen-activated protein kinases (MAPKs) are involved in important processes, including stress signaling and development. In a functional yeast screen, we identified mutations that render Arabidopsis thaliana MAPKs constitutively active (CA). Importantly, CA-MAPKs maintain their specificity toward known activators and substrates. As a proof-of-concept, Arabidopsis MAPK4 (MPK4) function in plant immunity was investigated. In agreement with the phenotype of mpk4 mutants, CA-MPK4 plants were compromised in pathogen-induced salicylic acid accumulation and disease resistance. MPK4 activity w…

Plant immunity depends on massive expression of pathogenesis-related genes (PRs) whose transcription is de-repressed by pathogen-induced signals. Salicylic acid (SA) acts as a major signaling molecule in plant immunity and systemic acquired resistance triggered by bacterial or viral pathogens. SA signal results in the activation of the master immune regulator, Nonexpressor of pathogenesis-related genes 1 (NPR1), which is thought to be recruited by transcription factors such as TGAs to numerous downstream PRs. Despite its key role in SA-triggered immunity, the biochemical nature of the transcri…

Plant life is coordinated by several developmental and defense associated mechanisms and cell wall is actively important in these mechanisms. The plant cell wall also serves as an important component system that monitors plant growth and defense apart from only protecting the cell. However, resident sensors of the plant cell membrane and pattern recognition receptors (PRRs) also constitute major response mechanisms in plants while they are both as well developed for maintaining cell wall integrity (CWI). These PRRs are molecules such as ligands, peptides or glycans that are released for sensin

Plant-associated yeasts modulate host immunity to promote or prevent disease. Mechanisms of yeast perception by the plant innate immune system remain unknown, with progress hindered by lack of a model system with the model plant Arabidopsis thaliana (Arabidopsis). A yeast strain of Taphrina tormentillae, named M11, was previously isolated from wild Arabidopsis. Taphrina have been found on many non-host plants, and their complex ecology remains understudied. Here, the interaction of M11 with Arabidopsis was characterised. Infection of Arabidopsis with the birch pathogen T. betulina, used as a n

BIR1 is a receptor-like kinase that functions as a negative regulator of basal immunity and cell death in Arabidopsis. Using Arabidopsis thaliana and Tobacco rattle virus (TRV), we investigate the antiviral role of BIR1, the molecular mechanisms of BIR1 gene expression regulation during viral infections, and the effects of BIR1 overexpression on plant immunity and development. We found that SA acts as a signal molecule for BIR1 activation during infection. Inactivating mutations of BIR1 in the bir1-1 mutant cause strong antiviral resistance independently of constitutive cell death or SA defens…

Plants have evolved powerful immune systems to recognize pathogens and avoid invasions, but the genetic basis of plant susceptibility is less well-studied, especially to oomycetes, which cause disastrous diseases in many ornamental plants and food crops. In this research, we identified a negative regulator of plant immunity to the oomycete Phytophthora parasitica, AtRTP5 (Arabidopsis thaliana Resistant to Phytophthora 5), which encodes a WD40 repeat domain-containing protein. The AtRTP5 protein, which was tagged with green fluorescent protein (GFP), is localized in the nucleus and plasma membr…

Sucrose non-fermenting-1-related protein kinase-1 (SnRK1) belongs to a family of evolutionary conserved kinases with orthologs in all eukaryotes, ranging from yeasts (SnF1) to mammals (AMP-Activated kinase). These kinases sense energy deficits caused by nutrient limitation or stress and coordinate the required adaptations to maintain energy homeostasis and survival. In plants, SnRK1 is a global regulator of plant metabolism and is also involved in abiotic stress responses. Its role in the response to biotic stress, however, is only starting to be uncovered. Here we studied the effect of altere

Chitosan oligomers prepared from enzyme hydrolysis of chitosan have for many years been recognized as potent elicitors of plant innate immunity, but their efficacy is limited by the degree of polymerization and the degree of acetylation. In this study, we presented a new type of chitosan oligomers (COS), with the name of quaternized chitosan oligomers (QCOS) that were prepared by reacting COS with glycidyltrimethylammonium chloride and overcome these problems. First, QCOS clearly induced hydrogen peroxide accumulation and callose deposition in Arabidopsis seedlings. Second, we found that PAD3

Plant surface-localized pathogen recognition receptors (PRRs) perceive conserved microbial features, termed pathogen-associated molecular patterns (PAMPs), resulting in disease resistance. PAMP perception leads to calcium influx, MAPK activation, a burst of reactive oxygen species (ROS) mediated by RbohD, accumulation of the defense hormone salicylic acid (SA), and callose deposition. Lectin receptor-like kinases (LecRKs) belong to a specific PRR family and are important players in plant innate immunity. Here, we report that LecRK-IX.2 is a positive regulator of PRR-triggered immunity. Pathoge

While herbivorous insects often secrete effector proteins to suppress plant immunity, alternative adaptation strategies remain less explored. Here, we report a counterintuitive approach in the small brown planthopper, Laodelphax striatellus. SBPH downregulated a salivary gland-specific elicitor gene, LsG40, when transferred from the optimal host rice to the suboptimal host corn. When expressed in plants, including the model organism tobacco and the two host plants corn and rice, LsG40 acts as a defense elicitor triggering calcium-dependent reactive oxygen species burst, cell death, callose dep…

Plant roots are extremely heterogeneous and non-photosynthetic tissue that is drastically different from leaves. Pioneering studies have revealed the existence of pattern-triggered immune responses in roots; however, whether roots mount tissue-specific effector-triggered immunity (ETI) is a fundamental question in plant immunity. By utilizing inducible effector-expressing lines, we systematically characterize non-conventional physiological and molecular responses during ETI activation in roots. Root ETI seems to be weaker than leaf ETI, evidenced by restricted cell death in the transition zone…

The plant defense hormone, salicylic acid (SA), plays essential roles in immunity and systemic acquired resistance. Salicylic acid induced by the pathogen is perceived by the receptor nonexpressor of pathogenesis-related genes 1 (NPR1), which is recruited by TGA transcription factors to induce the expression of pathogenesis-related (PR) genes. However, the mechanism by which post-translational modifications affect TGA's transcriptional activity by salicylic acid signaling/pathogen infection is not well-established. Here, we report that the loss-of-function mutant of brassinosteroid insensitive

Nonspecific lipid transfer proteins (LTPs) are small, cysteine-rich proteins that play numerous functional roles in plant growth and development, including cutin wax formation, pollen tube adhesion, cell expansion, seed development, germination, and adaptation to changing environmental conditions. LTPs contain eight conserved cysteine residues and a hydrophobic cavity that provides a wide variety of lipid-binding specificities. As members of the pathogenesis-related protein 14 family (PR14), many LTPs inhibit fungal or bacterial growth, and act as positive regulators in plant disease resistanc

The pepper plant, a member of the Solanaceae family, is widely cultivated and valued as an important spice in many dishes worldwide. Viruses cause significant damage to both the quality and quantity of pepper production, with cucumber mosaic virus (CMV) being particularly detrimental. Chitosan oligosaccharide (COS), a natural oligosaccharide derived from chitosan degradation, act as a plant immunity inducer. The present study aimed to verify the role of COS in inducing CMV resistance in capsicum and to explore its potential defense-related signaling pathways. The optimal condition for inducing…

In a recent issue of Science, Gao et al.1 discover a link between pattern-triggered immunity (PTI) initiated by plant cell surface receptors and effector-triggered immunity (ETI) initiated upon detection of pathogen effectors by intracellular nucleotide-binding leucine-rich repeat (NLR) immune receptors. PTI activates alternative splicing that results in mRNAs for the potato NLR Rpi-vnt1.1 that lack a repressive N-terminal prodomain.
