Intravaginal poly-(D, L-lactic-co-glycolic acid)-(polyethylene glycol) drug-delivery nanoparticles induce pro-inflammatory responses with Candida albicans infection in a mouse model.

In a recent study, using an in vitro model to study intravaginal nanoparticle exposure during yeast infections, we demonstrated that C. albicans exposure suppressed apoptotic gene expression and induced oxidative stress and pyroptosis in vaginal epithelial cells. The mucous-penetrating drug delivery nanoparticles made from poly-(D, L-lactic-co-glycolic acid)-(polyethylene glycol) induced cytotoxicity by activating apoptosis, endoplasmic reticulum (ER) stress, oxidative stress, and DNA damage repair responses alone and, in some cases with C. albicans. In the current study we evaluated the effec

Expression of cellular oncogenes in teratoma-derived cell lines.

The expression of ten proto-oncogenes was studied in cell lines derived from transplantable mouse teratomas. The cell lines represent different forms of early embryonic cell specialization. The analysis included two embryonal carcinoma (EC) lines (PCC3 and F9), and four differentiated cell lines derived from teratocarcinoma, namely trophoblastoma (3-TDM), parietal endoderm (PYS-2), visceral endoderm (PSA5-E) and skeletal myoblasts (Cl10). The expression of c-oncogenes was studied by analysing poly(A)+RNA for complementary sequences by dot blot and Northern blot hybridization. The results were

Functional characterization of specialized immune cells in a cnidarian reveals an ancestral antiviral program.

Examining early-branching animal phyla can help reconstruct the evolutionary origins of immune cells. Here, we characterize immune-related cell programs in embryos of the sea anemone Nematostella vectensis, a model of Cnidaria, which diverged ~600 million years ago from other animals. Using a transgenic Nematostella reporter line expressing mCherry under the RLRb antiviral promoter, we identify a morphologically and transcriptomically distinct cell population activated by the viral mimic poly(I:C). These cells upregulate immune effector and regulator genes and show increased phagocytic activit

Generation of the Early-Gestational Maternal Immune Activation Mouse Model to Assess Prenatal Inflammation on Neurodevelopment.

Neurodevelopmental disorders (NDDs) arise from complex interactions between genetic and environmental risk factors, including prenatal immune activation. Maternal immune activation (MIA) in mice is a widely used model to study environmental contributions to NDDs; variability in experimental outcomes limits reproducibility. Here, we present a standardized protocol for inducing MIA during early gestation at embryonic day 9.5 (E9.5) in mice, incorporating multiple quality control metrics to assess immune response and predict pregnancy outcomes. Following administration of polyinosinic:polycytidyl

SIFD-associated TRNT1 deficiency unveils importance of TSPO during macrophage antibacterial and antiviral responses.

INTRODUCTION: Mitochondria support cellular biosynthetic and bioenergetic demands and mediate cell signaling. Their dysfunction is implicated in a wide range of diseases, including congenital disorders. One such disorder, sideroblastic anemia with B-cell immunodeficiency, periodic fevers, and developmental delay (SIFD), is caused by mutations in the tRNA-nucleotidyltransferase enzyme TRNT1. While SIFD is known to affect immune function, the role of macrophages-key mediators between innate and adaptive immunity-remains underexplored. METHODS: To investigate the impact of TRNT1 deficiency on ma

GSTP1 is a negative regulator of MAVS in the antiviral signaling against SVCV infection.

Glutathione S-transferase P1 (GSTP1), the most ubiquitous member of the GST superfamily, plays vital roles in the detoxification, antioxidant defense, and modulation of inflammatory responses. However, limited studies have been conducted on the function of GSTP1 in antiviral innate immunity. In this study, we have cloned the homolog of GSTP1 in triploid hybrid crucian carp (3nGSTP1) and investigated its regulatory role in the interferon signaling pathway. The open reading frame of 3nGSTP1 is composed of 627 nucleotides, encoding 209 amino acids. In response to spring viremia of carp virus (SVC

Trial watch: TLR3 agonists in cancer therapy.

UNLABELLED: Toll-like receptor 3 (TLR3) is a pattern recognition receptor that senses exogenous (viral) as well as endogenous (mammalian) double-stranded RNA in endosomes. On activation, TLR3 initiates a signal transduction pathway that culminates with the secretion of pro-inflammatory cytokines including type I interferon (IFN). The latter is essential not only for innate immune responses to infection but also for the initiation of antigen-specific immunity against viruses and malignant cells. These aspects of TLR3 biology have supported the development of various agonists for use as stand-al

Poly ICLC in the treatment of postinfectious demyelinating encephalomyelitis.

Three weeks after a mild and presumably infectious illness, a 21-year-old man developed a CNS disorder characterized by involvement of the cerebellum, cerebrum, and brainstem. It progressed, sometimes stepwise, without remission, over five months to being bedfast with total spastic paraplegia, severe ataxia, and unintelligible dysarthric speech. The CSF showed increased levels of protein, IgG, and myelin basic protein, as well as five oligoclonal bands. Because of failure of the patient's condition to respond to prolonged prednisone therapy, poly ICLC was given intravenously weekly for 20 week

Heterologous expression of L-lactate dehydrogenase in Chlorella pyrenoidosa: A beginning for poly(L-lactic acid) biosynthesis from CO2 by eukaryotic microalgae.

Lactic acid is a monomer of poly(lactic acid), and the use of sugar-based raw materials in conventional lactic acid production poses a risk to food security. CO2 is an abundant carbon resource, and microalgae are crucial for the development of third-generation bio-manufacturing through CO2 utilization. In this study, the transformation efficiency of Chlorella pyrenoidosa (C. pyrenoidosa) was investigated during Agrobacterium-mediated transformation, and an efficient transformation system was established. L-lactate dehydrogenases (LLDHs) from Bos taurus, Lactobacillus plantarum, and Bacillus co

Intranasal exposure of poly (I:C) exacerbates OVA-induced allergic asthma by causing a major shift in the immune response.

Viral respiratory infections are the major cause of exacerbation of allergic asthma, often resulting in increased emergency visits and hospitalizations. However, the understanding of the immune pathways at the cellular/molecular level under the conditions is lacking. Therefore, the present work was designed to elucidate the complex interplay of immune response under the settings mimicking exacerbation of allergic asthma upon viral infection using mouse model of the condition. Mice were sensitized & challenged with Ovalbumin (OVA) to induce allergic asthma, and subsequently subjected to intrana

Pentraxin family members of largemouth bass (Micropterus salmoides): Cloning, characterization and expression responses to LPS, Poly (I:C) and Nocardia seriolae.

Pentraxins (PTXs) are highly conserved soluble pattern recognition molecules, and play important roles in the innate immunity and autoimmune diseases. In present study, ten PTXs including CRP1, SAP, NPTX1-like, NPTX2a, NPTX2b, NPTXR-like, NPTXRb, PTX3a, PTX3-like and PTX4 were identified from largemouth bass (Micropterus salmoides). These PTXs exhibited high sequence identities with their counterparts of other fish species, and contained conserved motifs and structures of the PTX family, except of PTX3-like. Phylogenetic analysis revealed that these largemouth bass PTXs were closely clustered

Developing a primer for communicating about penetrance, expressivity, and genotype-phenotype correlation of R117H and the poly-T tract in CFTR.

Variants in the cystic fibrosis transmembrane conductance regulator (CFTR) gene, which causes cystic fibrosis, have become increasingly well-characterized in recent decades. However, clinicians remain challenged by CFTR variants that result in variable expressivity and reduced penetrance, including the variant R117H (c.350G>A) and the intron 9 poly-thymidine (poly-T) and poly-thymidine-guanosine (TG) tracts, highlighting a knowledge gap that impacts the delivery of accurate information to patients and families. To address this gap, we developed an educational resource that provides descriptive

Rapid, non-invasive imaging of alphaviral brain infection: reducing animal numbers and morbidity to identify efficacy of potential vaccines and antivirals.

Rapid and accurate identification of disease progression are key factors in testing novel vaccines and antivirals against encephalitic alphaviruses. Typical efficacy studies utilize a large number of animals and severe morbidity or mortality as an endpoint. New technologies provide a means to reduce and refine the animal use as proposed in Hume's 3Rs (replacement, reduction, refinement) described by Russel and Burch. In vivo imaging systems (IVIS) and bioluminescent enzyme technologies accomplish the reduction of animal requirements while shortening the experimental time and improving the accu

Adsorption-Engineered Hydrocarbon Ionomers for Durable Proton-Exchange Membrane Fuel Cells.

Reducing reliance on perfluoroalkyl substances (PFAS) in proton-exchange membrane fuel cells requires hydrocarbon ionomers that combine high performance with long-term durability, a persistent challenge in catalyst-layer design. Here, we identify oxidation-driven ionomer-catalyst interfacial degradation as a dominant failure pathway in hydrocarbon ionomer-bonded cathodes and introduce an adsorption-engineering strategy to overcome this limitation. The comparison of a commercial sulfonated poly(phenylene) (Pemion) with structurally engineered sulfonated poly(fluorene)s demonstrated that electro

Preparation and antitumor characteristics of PLA/(PEG-PPG-PEG) nanoparticles loaded with camptothecin.

Camptothecin (CPT)-loaded nanoparticles were prepared using poly(dl-lactic acid) (PLA) and poly(ethylene glycol)-block-poly(propylene glycol)-block-poly(ethylene glycol) copolymer (PEG-PPG-PEG), and examined for particle characteristics, in vitro release, pharmacokinetics and efficacy. The preparative condition, in which the ratio of PLA/PEG-PPG-PEG/CPT was 35/35/4 (w/w/w) and organic solvent (dichloromethane) was evaporated from the emulsion at 18 degrees C, gave the nanoparticles with the diameter of approximately 230nm, fairly high drug content (ca. 1.6% (w/w)) and stable entrapment of the

Bioinspired multifunctional hydrogel actuator with rapid temperature/near-infrared light dual-stimuli responsiveness and strain-sensing capability.

In this work, a bilayer hydrogel with dual responsiveness to temperature and near-infrared (NIR) light, together with strain-sensing capability, was prepared via stepwise in situ free-radical polymerization. Upon heating or NIR irradiation, the poly(N-isopropylacrylamide-co-acrylamide) (PNAM)/Ti₃C₂Tₓ MXene active layer contracts, while the poly(acrylamide) (PAAm)/poly(vinyl alcohol) (PVA) passive layer provides flexible mechanical support and undergoes relatively limited deformation, resulting in directional bending of the bilayer hydrogel. Meanwhile, MXene serves as both the photothermal comp

Degradation of an ultrasonically welded device for surgical suture holding.

A biodegradable ultrasonically welded device has for the first time been developed for in-body sutures that eliminates the need for surgical knotting. The device comprises two parts that fit together, with a suture inserted between them. Ultrasonic welding is then used to secure the suture by welding the two parts together. The device was manufactured from three biodegradable polymers: Poly(L-lactide-co-D,L-lactide) [PLDLA]; Poly(L-lactide-co-glycolide) [PLGA]; Poly(L-lactide) [PLLA]. All devices were degraded through immersion in phosphate buffer solution at a temperature of 37°C ± 2°C. Knott

Sphingomyelin Phosphodiesterase Acid-Like 3b is Essential for Toll-Like Receptor 3 Signaling in Human Podocytes.

Recent studies have revealed the importance of cell membrane stability in normal cell function. Sphingomyelin phosphodiesterase acid-like 3b (SMPDL3b), a lipid modifying enzyme that converts sphingomyelin to ceramide in the cell membrane, is expressed in macrophages and regulates Toll-like receptor (TLR) 4 signaling by altering cell membrane fluidity. SMPDL3b is also expressed in human podocytes, which are involved in the pathogenesis of several glomerular diseases such as diabetic kidney disease, focal segmental glomerulosclerosis, and idiopathic nephrotic syndrome in children; however, the r

Conformational characteristics and conformation-dependent properties of poly(ε-caprolactone).

Structures and properties of isolated and crystalline poly(ε-caprolactone) (PCL) chains have been investigated. Ab initio molecular orbital (MO) calculations were conducted for methyl 6-acetoxyhexanoate (MAH), a model compound of PCL, to yield Gibbs free energies of all its existent conformers. Bond conformations of its seven bonds corresponding to the repeating unit of PCL were calculated therefrom; two C-C bonds close to the ester group show gauche preferences owing to intramolecular C-H⋯O attractions, and the other five prefer trans forms. 1H and 13C NMR vicinal coupling constants of 13C-la
