Hypokalemic periodic paralysis: in vitro investigation of muscle fiber membrane parameters.

To study the mechanism of attacks in familial hypokalemic paralysis, we recorded resting membrane potentials, action potentials, current-voltage relationships, and isometric forces in intercostal muscle fibers from three patients. In normal extracellular medium, the resting potential was reduced, but membrane conductance was not different from control. Excitability was reduced and the action potentials had no overshoot. On exposure to a 1-mM potassium solution, with or without insulin, the cells depolarized to about -50 mV, and became inexcitable. Over the tested membrane potential range from

Pathophysiologic approach in genetic hypokalemia: An update.

The pathophysiology of genetic hypokalemia is close to that of non-genetic hypokalemia. New molecular pathways physiologically involved in renal and extrarenal potassium homeostasis have been highlighted. A physiological approach to diagnosis is illustrated here, with 6 cases. Mechanisms generating and sustaining of hypokalemia are discussed. After excluding acute shift of extracellular potassium to the intracellular compartment, related to hypokalemic periodic paralysis, inappropriate kaliuresis (>40mmol/24h) concomitant to hypokalemia indicates renal potassium wasting. Clinical analysis dist

Electrolytes and water in the brain and cerebrospinal fluid of the foetal sheep and guinea-pig.

1. Samples of cisternal cerebrospinal fluid (c.s.f.) and blood plasma have been obtained nearly simultaneously from foetal sheep of different ages, the foetus having been exteriorized and maintained in a normal state with respect to its blood gases and arterial pH. The brains were removed from these foetuses and also from foetal guinea-pigs after exsanguination.2. A comprehensive study has been made of the concentrations of water, chloride, sodium, potassium, calcium and magnesium in blood plasma, c.s.f. and brain from foetal sheep and in brain from foetal guinea-pigs during development in ute

The effect of hydrochlorothiazide and amiloride administered together on muscle electrolytes in normal subjects.

Hydrochlorothiazide, 150 mg daily, and amiloride, 15 mg daily, have been administered together to 10 normal subjects during one week. Muscle biopsies were performed before and after the period of administration of the diuretics and the material was analysed for water and electrolytes. The body weight decreased by 1.9 kg. The serum sodium, chloride and potassium concentrations decreased significantly and standard bicarbonate and blood pH increased. In muscle tissue, extracellular water, chloride and sodium contents and intracellular sodium concentration decreased but the muscle potassium conten

Endogenous Na(+)-K+ (or NH4+)-2Cl- cotransport in Rana oocytes; anomalous effect of external NH4+ on pHi.

1. In Rana oocytes, measurements with chloride-sensitive microelectrodes show that the mean intracellular chloride activity (34.8 +/- 6.3 mM, n = 79) is three times higher than that expected for the passive distribution of chloride ions across the outer membrane (12.4 mM, mean membrane potential -43 +/- 8.8 mV, n = 79). 2. Reuptake of chloride into oocytes depleted by prolonged exposure to chloride-free saline takes place against the electrochemical gradient. 3. Chloride reuptake does not take place in sodium-free solution or in a sodium-substituted potassium-free solution. It is inhibited by

Water and electrolyte balance after ileoanal anastomosis.

Water and electrolyte balance was studied in 30 patients with ileoanal anastomosis and J pouch, 10 patients with conventional ileostomy, and nine nonoperated patients with quiescent ulcerative colitis. Serum electrolyte concentrations, daily urinary volume, and daily losses of sodium, potassium, and chloride were measured in all patients. Daily fecal weight and daily losses of sodium and potassium were analyzed in patients with ileoanal anastomosis or conventional ileostomy. Serum chloride in patients with ileoanal anastomosis was significantly lower (P less than 0.05) than in those with conve

Deafness disrupts chloride transporter function and inhibitory synaptic transmission.

Loss of sensory function leads to atrophy or death within the developing CNS, yet little is known about the physiology of remaining synapses. After bilateral deafening, gramicidin-perforated-patch recordings were obtained from gerbil inferior colliculus neurons in a brain slice preparation. Afferent-evoked IPSPs had a diminished ability to block current-evoked action potentials in deafened neurons. This change could be attributed, in part, to a loss of potassium-dependent chloride transport function, with little change in K-Cl cotransporter expression. Treatments that suppressed chloride cotra

Comparative sensitivity of three tropical cladoceran species (Diaphanosoma brachyurum, Ceriodaphnia rigaudii and Moinodaphnia macleayi) to six chemicals.

The toxicities of six chemicals (cadmium chloride, potassium dichromate, sodium dodecyl sulfate, potassium chloride, Tritox X-100 and copper (II) sulphate) was determined for three tropical freshwater cladoceran, Moinodaphnia macleayi, Ceriodaphnia rigaudii and Diaphanosoma brachyurum. The data was subsequently used to compare the relative sensitivity of the three species. Relative sensitivities were determined by comparing the 48-hour LC50 values using an independent t-test and interspecies correlation. Potassium dichromate was the most toxic compound tested, while potassium chloride was the …

The effect of removing area postrema on the sodium and potassium balances and consumptions in the rat.

Previous studies have indicated that rats lose weight and develop a taste preference for solutions of sodium chloride after area postrema has been removed; results in other species have suggested that area postrema may be concerned with regulating the excretion of sodium and potassium. The results reported here demonstrate that in rats the reduction of weight can be explained adequately by anorexia, that diminished excretion of sodium and potassium results from anorexia, and that the consequent slower rate of gain of weight causes diminution of sodium and potassium balances. The results also s

Renal function and urinary prostanoid excretions in salt-depleted women: comparative effects of enalapril and indomethacin treatments.

The acute effects on urinary prostanoid excretion and on renal function induced by pharmacological inhibition of either the angiotensin-converting enzyme or of the cyclooxygenase system, respectively, have been studied in healthy salt-depleted women. Two experimental groups were studied during salt depletion, SD1 (n=8) and SD2 (n=6). Salt depletion was obtained by combining a low sodium chloride dietary intake (< or =60 mmol per day) with natriuretic and potassium sparing treatment. Paired studies were performed in the absence and in the presence of enalapril (SD1 group) or indomethacin (SD2 g

Effect of acute metabolic alkalosis and acidosis on intestinal electrolyte transport in vivo.

The effects of acute metabolic alkalosis and acidosis on intestinal electrolyte transport were studied in adult Sprague-Dawley rats. Animals were made alkalotic or acidotic by gavage feeding of 1 M solutions of NaCl (pH = 7.42), NaHCO3 (pH = 7.52), NH4Cl (pH = 7.18), or 0.75 M (NH4)2SO4 (pH = 7.21). After 1-3 h, animals were anesthetized and prepared for in vivo perfusion of the jejunum, ileum, and colon. The jejunum exhibited increased net potassium absorption in alkalosis and decreased potassium absorption in acidosis. In the ileum, net sodium absorption and potassium secretion were decrease

Renal tubular hyperkalaemia in childhood.

Potassium output from the body is regulated by renal excretion, which takes place predominantly in the late distal and cortical collecting tubules. The accepted model for potassium secretion implies the accumulation of potassium into the cell by the activity of basolateral Na-K-ATPase and its exit through voltage-dependent conductive channels. The factors regulating renal potassium secretion are potassium intake, distal urinary flow, systemic acid-base equilibrium, aldosterone, antidiuretic hormone and, probably, epinephrine. Renal handling of potassium is best studied by the response to the a
