Transition from embryonic to adult transcription pattern of serotonin N-acetyltransferase gene in avian pineal gland.

The study reports the change of transcription pattern of serotonin N-acetyltransferase gene and melatonin receptor genes during ontogenesis of the avian pineal gland. The RT-PCR technique was used to investigate the expression of the arylalkylamine N-acetyltransferase (AA-NAT) and melatonin receptor genes during development of the pineal glands isolated from Japanese quail (Coturnix coturnix japonica) embryos incubated from 3 days on until hatching (17 days), and in some organs (pineal, brain hemisphere, eye, leg, heart) of the 3-day-old quail embryo. It was shown that two phases of AA-NAT exp

Post-transcriptional regulation of the human liver/bone/kidney alkaline phosphatase gene.

Osteoblasts express high levels of liver/bone/kidney alkaline phosphatase (LBK AP), an enzyme critical for bone formation. Other tissues and cell types generally express much lower levels of LBK AP and correspondingly lower levels of mRNA. In light of our early observations that the human LBK AP promoter is expressed equally when transfected into a variety of different cells, we have carried out a detailed study of LBK AP gene expression in Saos-2 cells which are osteoblast-derived and express high levels of LBK AP mRNA, and in HepG2 hepatoblastoma cells which express LBK AP mRNA at levels whi

LncRNAs are Involved in the Process of Atherosclerosis at Diverse Levels.

Atherosclerosis is the most common cause of cardiovascular disease globally, associated with a high incidence of clinical events. Accumulating evidence has elucidated that long non-coding RNAs (lncRNAs) as a novel class of transcripts with critical roles in the pathophysiological processes of atherosclerosis. In this review, we summarize the recent progress of lncRNAs in the development of atherosclerosis. We mainly describe the diverse regulatory mechanisms of lncRNAs at the transcriptional and post-transcriptional levels. This study may provide helpful insights about lncRNAs as therapeutic t

Dual transcriptional and post-transcriptional regulation by the bHLH proteins Rtg1 and RtgX in Komagataella phaffii.

In Saccharomyces cerevisiae, the basic helix-loop-helix (bHLH) transcription factors Rtg1 and Rtg3 mediate retrograde signaling. In Candida albicans, the Rtg1-Rtg3 complex regulates galactose and sphingolipid metabolism and contributes to virulence. In the methylotrophic yeast Komagataella phaffii (formerly Pichia pastoris), Rtg1 (KpRtg1) controls glutamate and methanol metabolism by regulating the synthesis of GDH2 (glutamate dehydrogenase 2), PEPCK (phosphoenolpyruvate carboxykinase), and AOX1 (alcohol oxidase 1). The function of the putative Rtg3 ortholog, designated here as KpRtgX, has rem

Forkhead box class O family member proteins: The biology and pathophysiological roles in diabetes.

Forkhead box class O family member proteins (FoxOs) of transcription factors are essential regulators of cellular homeostasis, including glucose and lipid metabolism, oxidative stress response and redox signaling, cell cycle progression, and apoptosis. Altered FoxO1 expression and activity have been associated with glucose intolerance, dyslipidemia and complications of diabetes. In the liver, they direct carbons to glucose or lipid utilization, thus providing a unifying mechanism for the two abnormalities of the diabetic liver: excessive glucose production, and increased lipid synthesis and se

Mitochondrial disorders.

Mitochondria, the organelles devoted to energy production, have unique genetic features. They possess their own genome encoding several subunits of the respiratory chain, the majority of which are encoded by nuclear DNA, as well as factors involved in replication, transcription and translation of mitochondrial DNA. In the past few years, molecular lesions of mitochondrial DNA have been reported with increasing frequency as a source of human disorders. Several mutations of mitochondrial DNA, either as sporadic large scale rearrangements (deletions, duplications) or maternally-inherited point mu

Hypoxia-mediated regulation of mRNA metabolism: from transcription to stability.

BACKGROUND: Hypoxia plays a critical role in both physiological and pathological processes. Cells adapt to hypoxia through the extensive reprogramming of gene expression. While the transcriptional regulations of hypoxia-inducible factors (HIFs) are well-studied, recent advances have emphasized the importance of post-transcriptional regulation, including mRNA translation, modification, stability, and degradation. Summarizing the regulation of mRNA homeostasis under hypoxia will improve our understanding of how hypoxia contributes to diseases such as cancer and ischemic injury. MAIN BODY: This r

Cellular concentrations of glutamine synthetase in murine organs.

Glutamine synthetase (GS) is the only enzyme that can synthesize glutamine, but it also functions to detoxify glutamate and ammonia. Organs with high cellular concentrations of GS appear to function primarily to remove glutamate or ammonia, whereas those with a low cellular concentration appear to primarily produce glutamine. To validate this apparent dichotomy and to clarify its regulation, we determined the GS concentrations in 18 organs of the mouse. There was a >100-fold difference in GS mRNA, protein, and enzyme-activity levels among organs, whereas there was only a 20-fold difference in

Post-transcriptional regulation of zenk expression associated with zebra finch vocal development.

In the male zebra finch, highly variable juvenile song and stereotyped adult song induce mRNA expression of the immediate early gene zenk in telencephalon. However, the functional consequences of this behavior-driven gene expression remain unknown. Here we characterize the developmental expression of zenk mRNA and protein in two forebrain song regions (HVC, the higher vocal center, and RA, the robust nucleus of the archistriatum). In HVC, singing results in similar percentages of cells producing zenk mRNA and zenk protein at different stages of vocal development. Similarly, song behavior at al

Export of Precursor tRNAIle from the Nucleus to the Cytoplasm in Human Cells.

In the current concept, tRNA maturation in vertebrate cells, including splicing of introns, trimming of 5' leader and 3' trailer, and adding of CCA, is thought to occur exclusively in the nucleus. Here we provide evidence to challenge this concept. Unspliced intron-containing precursor tRNAIle was identified in Human Immunodeficiency Virus type 1 (HIV-1) virions, which are synthesized in the cytoplasm. Northern blot, confocal microscopy and quantitative RT-PCR further verified enrichment of this unspliced tRNAIle within the cytoplasm in human cells. In addition to containing an intron, the cyt

Genome-wide identification of RNA editing sites in chloroplast transcripts and multiple organellar RNA editing factors in tea plant (Camellia sinensis L.): Insights into the albinism mechanism of tea leaves.

RNA editing is a post-transcriptional modification process, the chloroplast genes of which are involved in the process of chloroplast development in plant. However, the RNA editing sites of chloroplast genes remains unknown. In this study, we identified 39 RNA editing sites in 18 chloroplast genes from chloroplast genome of C. sinensis. Furthermore, the feature, structures and specificity of RNA editing sites were systematic analyzed. The differential editing efficiency were examined at 11 RNA editing sites among C. sinensis var. sinensis 'Huabai 1', 'Baiye 1' and 'Longjing 43'. Meanwhile, we

BR-bodies link RNA homeostasis to stress tolerance and intracellular fitness in Brucella.

Bacterial gene expression depends on the coordinated regulation of RNA synthesis, processing, and decay. The conserved RNA degradosome scaffold, ribonuclease E (RNase E), assembles into phase-separated bacterial ribonucleoprotein bodies (BR-bodies) through its C-terminal intrinsically disordered region (IDR). This IDR also scaffolds recruitment of degradosome client proteins that carry out post-transcriptional gene regulation. Whether BR-bodies regulate virulence programs to support infection, however, is largely undefined. We show that RNase E of the intracellular pathogen Brucella ovis forms

Nuclear Protein Aggregates Disrupt RNA Processing and Alter Biomechanics in a Muscle Cell Model of OPMD.

Aggregation of RNA-binding proteins (RBPs) is a hallmark of several age-related neuromuscular diseases. However, our understanding of how these aggregates drive dysfunction is often limited by the use of non-disease-relevant models. Oculopharyngeal muscular dystrophy (OPMD) is caused by a short alanine expansion mutation in the PABPN1 gene, which leads to nuclear aggregation of the protein. To investigate how these aggregates impair muscle cell function, we developed a muscle cell model with inducible expression of the pathogenic PABPN1 (A16) variant and confirmed its relevance to OPMD. Using

Cross-Species Analysis of Transcriptomic Response to Alpha-Herpesvirus Infection in Human, Bovine and Equine Cells.

Comparative transcriptomics offers a powerful approach to elucidate host-virus interactions across related pathogens, yet systematic evaluations across species-matched cellular systems remain limited. We performed a cross-species RNA sequencing analysis of respective species' cells infected with three alphaherpesviruses-herpes simplex virus 1 (HSV-1), bovine alphaherpesvirus 1 (BHV-1), and equid alphaherpesvirus 1 (EHV-1)-to dissect conserved and virus-specific transcriptional responses. We show that certain orthologous genes and orthologous pathways are differentially regulated upon infection

'See-saw' expression of microRNA-198 and FSTL1 from a single transcript in wound healing.

Post-transcriptional switches are flexible effectors of dynamic changes in gene expression. Here we report a new post-transcriptional switch that dictates the spatiotemporal and mutually exclusive expression of two alternative gene products from a single transcript. Expression of primate-specific exonic microRNA-198 (miR-198), located in the 3'-untranslated region of follistatin-like 1 (FSTL1) messenger RNA, switches to expression of the linked open reading frame of FSTL1 upon wounding in a human ex vivo organ culture system. We show that binding of a KH-type splicing regulatory protein (KSRP,

DDX17 and viral infection.

DDX17 (DEAD-box RNA helicase 17) is an essential RNA helicase and regulatory ATPase in host cells, extensively involved in various cellular processes during viral infections, such as RNA splicing, transcriptional regulation, and post-transcriptional modification. DDX17 exhibits dual functionality in viral infections: it enhances the stability, packaging, and replication of viral RNA through interactions with viral ribonucleoprotein complexes, as evidenced in infections caused by influenza viruses and Hantaan virus (HTNV). Conversely, DDX17 can inhibit viral proliferation by disrupting viral RN

Posttranscriptional regulation of human endogenous retroviruses by RNA-binding motif protein 4, RBM4.

The human genome encodes for over 1,500 RNA-binding proteins (RBPs), which coordinate regulatory events on RNA transcripts. Most studies of RBPs have concentrated on their action on host protein-encoding mRNAs, which constitute a minority of the transcriptome. A widely neglected subset of our transcriptome derives from integrated retroviral elements, termed endogenous retroviruses (ERVs), that comprise ∼8% of the human genome. Some ERVs have been shown to be transcribed under physiological and pathological conditions, suggesting that sophisticated regulatory mechanisms to coordinate and preven

An approach for the systematic profiling of drug-induced remodeling of RNA-RBP (RNA-binding protein) interactions.

Nucleic acid-based therapeutics targeting RNA-related regulatory networks have received substantial attention in drug development, among which the interventions of RNA-RBP (RNA-binding protein) interactions or RNA-protein complex assemblies represent a promising strategy for treating various diseases. RBPs play essential roles in post-transcriptional regulation, and their interactions with RNA are closely associated with physiological homeostasis and pathological progression. Although RNA-RBP interactions are increasingly recognized as therapeutic targets, the lack of systematic methods for sc

[Research progress on the involvement of N6-methyladenosine modification in the regulation of immune response in sepsis].

Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection, its exact pathogenesis has not yet been fully elucidated, and the clinical mortality rate remains high. N6-methyladenosine (m6A) modification is a common form of post-transcriptional chemical modification in eukaryotes. By influencing RNA splicing, nuclear-cytoplasmic transport, and translation, it plays a role in regulating various pathophysiological processes. Recent studies have revealed that dynamic changes in m6A modifications are associated with damage caused by the immune response in seps
