Fibroblast-macrophage crosstalk is well-established in vitro, and fibroblast-derived colony-stimulating factor 1 (CSF1) supports macrophages in select tissues. However, whether macrophages regulate fibroblasts in vivo remains unknown. Leveraging genetic mouse models, single-cell multi-omics, flow cytometry and imaging, we show that fibroblast depletion or loss of fibroblast-derived growth factors impacts skin macrophage populations in the dermis and hypodermis. Conditional deletion of Csf1 in Dpt+ fibroblasts progressively decreases CD64+ and CD11c+ macrophages, impairing skin wound healing. R

INTRODUCTION: Tenosynovial giant cell tumor (TGCT) is a rare, benign neoplasm driven by overexpression of colony-stimulating factor 1 (CSF-1), which recruits CSF-1 receptor (CSF-1 R)-bearing macrophages and other inflammatory cells to promote tumor formation. While surgery remains the first-line treatment for TGCT, many patients experience unresectable or recurrent disease with high morbidity, highlighting the need for effective systemic treatments.
AREAS COVERED: This review summarizes the molecular pathogenesis, diagnostic approach, and evolving systemic treatment landscape for TGCT, with a

We have previously shown that total T cells derived from lymph nodes (LN) involved by Hodgkin's disease (HD) secrete higher levels of colony-stimulating activity than total T cells present within benign hyperplastic (BH) LN and B-non-Hodgkin's lymphoma (B-NHL) LN, suggesting that T cells with particular properties accumulate in HD LN. To further characterize this T-cell population, we have quantified production of both granulocyte-macrophage colony-stimulating factor (GM-CSF) and macrophage colony-stimulating factor (M-CSF) production in a total of 98 T-cell clones (TCC) derived from CD25+ act

Endometriosis (EM) is a prevalent gynecological disorder linked to pelvic pain, menstrual disturbances, and infertility. Despite its clinical burden, the molecular mechanisms underlying EM remain elusive, necessitating the discovery of novel biomarkers and therapeutic targets. This study explored the role of colony-stimulating factor 1 receptor (CSF1R) in EM through an integrative approach combining bioinformatics and experimental validation. Differentially expressed genes (DEGs) were identified from the GSE11691 dataset using the limma package. Weighted Gene Co-expression Network Analysis (WG

Immune thrombocytopenia (ITP) is an autoimmune bleeding disorder characterized by platelet destruction and defective megakaryopoiesis. However, the mechanisms underlying megakaryocyte (MK) dysfunction in ITP remain unclear. To address this, we performed single-cell RNA sequencing (scRNA-seq) on bone marrow cells from a newly diagnosed ITP patient. ScRNA-seq analysis revealed a marked upregulation of colony-stimulating factor 1 receptor (CSF1R) in MK compared with healthy control. This finding was independently validated by flow cytometry in additional clinical samples. In vitro, MK differentia

A 66-year-old male with mantle cell lymphoma was treated with chemotherapy before consolidation with autologous stem cell transplantation. Despite three series of treatment with chemotherapy and stimulation with granulocyte colony-stimulating factor (filgrastim), it was not possible to mobilize hematopoietic stem cells for leukapheresis. After permission from the Danish Medicines Agency, a new stem cell factor, plerixafor, which is a direct antagonist of a surface receptor on stem cells was used. After five days of pre-treatment with filgrastim and two days of treatment with plerixafor, it was

Antiestrogens, such as tamoxifen, are widely used for endocrine treatment of estrogen receptor-positive breast cancer. However, as breast cancer progresses, development of tamoxifen resistance is inevitable. The mechanisms underlying this resistance are not well understood. To identify genes involved in tamoxifen resistance, we have developed a rapid screening method. To alter the tamoxifen-sensitive phenotype of human ZR-75-1 breast cancer cells into a tamoxifen-resistant phenotype, the cells were infected with retroviral cDNA libraries derived from human placenta, human brain, and mouse embr

Chronic infection with Schistosoma mansoni induces in humans and mice a Th2-dominant immune response in which eosinophils and IgE are conspicuously elevated. Human eosinophils express IgE receptors that participate in an IgE-dependent eosinophil-mediated ADCC reaction against Schistosomula larvae in vitro. To investigate the expression of IgE receptors on murine eosinophils, they were purified (>95% pure by Giemsa-stained cytospin preparations) from liver granulomas of Schistosoma-infected mice. Flow cytometric analysis showed the absence of the low-affinity IgE receptor Fc-epsilon RII (CD23)

BACKGROUND: Tenosynovial giant cell tumor (TGCT) is a locally aggressive neoplasm caused by dysregulation of the colony-stimulating factor 1 (CSF1) gene. Patients report substantial pain, stiffness, and declining physical function; those whose disease is not amenable to surgery require systemic therapy. Vimseltinib is an oral, switch-control kinase inhibitor of the CSF1 receptor (CSF1R). Here, the authors report safety and efficacy of vimseltinib in patients with TGCT based on prior treatment. Cohort A included patients who did not receive prior specific anti-CSF1/CSF1R agents (n = 46; prior i

BACKGROUND: Tenosynovial giant cell tumor refers to a group of rarely occurring tumors that are formed in the joints, which are characterized by pain, swelling, and limitation of movement of the joint. Surgery is the main treatment strategy, but the tumor is likely to recur, especially in pigmented villonodular synovitis, which is the diffuse-type giant cell tumor. Pexidartinib was approved in August 2019 by the Food and Drug Administration (FDA) with a brand name TURALIO as the first systemic approved therapy for patients having Tenosynovial Giant Cell Tumors (TGCT).
OBJECTIVE: In this revie

The granulocyte colony-stimulating factor (G-CSF), being a member of the hematopoietic growth factor family, is also critically involved in controlling proliferation and differentiation of neural stem cells. Treatment with G-CSF has been shown to result in substantial neuroprotective and neuroregenerative effects in various experimental models of acute and chronic diseases of the central nervous system. Although G-CSF has been tested in a clinical study for treatment of acute ischemic stroke, there is only fragmentary data on the distribution of this cytokine and its receptor in the human brai

Behçet's disease (BD) is a systemic inflammatory disorder characterized by recurrent episodes of acute inflammation consisting mainly of neutrophil infiltration around blood vessels in affected tissues. BD probably occurs due to neutrophil-based innate immune responses orchestrated by a complex interplay among gamma-delta T lymphocytes, natural killer T cells, monocytes and Th17 lymphocytes in which type-I interferon is possibly a key element for inflammatory downregulation. However, strong evidence is still scarce. This article compiles the literature in an attempt to summarize the possible m

Colony-stimulating factor 1 receptor (CSF1R) is a key regulator of macrophage-driven liver inflammation. Here, we report a series of CSF1R inhibitors discovered through a structure-guided optimization strategy for the acute-phase treatment of acetaminophen-induced liver injury. For instance, compound C52 exhibited potent CSF1R inhibition, a kinome-wide selective profile, and low cellular cytotoxicity. C52 rapidly suppressed M-CSF-induced phosphorylation events and downstream signaling in macrophages. In an acute liver injury model, therapeutic administration of C52 during the early inflammator

BACKGROUND: The colony-stimulating factor 1 receptor (CSF1R) is a receptor tyrosine kinase essential for microglial development and homeostasis. While dysregulated CSF1R signaling has been implicated in Alzheimer's disease (AD), the biological function of its soluble ectodomain (sCSF1R)-generated by a disintegrin and metalloproteinase 17 (ADAM17)-mediated cleavage-remains poorly understood in neurodegeneration.
METHODS: We quantified sCSF1R levels in the brain and cerebrospinal fluid (CSF) of 5×FAD transgenic mice and wild-type controls using ELISA and immunoblotting, and reanalyzed publicly

Colony stimulating factor 1 receptor (CSF1R) plays key roles in regulating development and function of the monocyte/macrophage lineage, including microglia and osteoclasts. Mono-allelic mutations of CSF1R are known to cause hereditary diffuse leukoencephalopathy with spheroids (HDLS), an adult-onset progressive neurodegenerative disorder. Here, we report seven affected individuals from three unrelated families who had bi-allelic CSF1R mutations. In addition to early-onset HDLS-like neurological disorders, they had brain malformations and skeletal dysplasia compatible to dysosteosclerosis (DOS)
