Spinosin, a C-glycoside flavonoid from semen Zizhiphi Spinozae, potentiated pentobarbital-induced sleep via the serotonergic system.

Semen Zizhiphi Spinozae has been used extensively for the treatment of insomnia. This study investigated the effect and possible mechanism of action of spinosin (also known as 2''-beta-o-glucopyranosyl swertisin), a major constituent of semen Zizhiphi Spinozae, on sleep in mice. The present results showed that spinosin significantly and dose-dependently augmented pentobarbital (45 mg/kg, i.p.)-induced sleep, reflected by increased sleep time and reduced sleep latency assessed with the loss-of-righting reflex, and these effects were potentiated by the 5-hydroxytryptamine (serotonin) precursor 5

Behavioral and neurochemical effects induced by subchronic combined exposure to toluene at 40 ppm and noise at 80 dB-A in rats.

We investigated whether exposure to noise, in addition to its well-known potentiating effect on toluene-induced ototoxicity, may also exacerbate behavioral disturbances and brain neurochemical alterations produced by subchronic exposure to low toluene concentration. To test this hypothesis, we evaluated whether subchronic combined exposure (16 weeks, 104 h per week) to noise at 80 dB-A and toluene at 40 ppm potentiates the recently reported neurotoxic effects of subchronic exposure to 40 ppm toluene. Locomotor and rearing activities, sensitization to narcosis induced by acute toluene at high c

Short-term immobilization of mice by methohexitone.

The feasibility of short-term immobilization for <5 min of female mice by methohexitone sodium was studied. In C3H/Neu mice, methohexitone at a dose <40 mg/kg did not result in chemical restraint, doses >50 mg/kg caused considerable lethality. A dose of 44 mg/kg, applied intraperitoneally at a concentration of 6.46 mg/ml, is suitable for immobilization without complications. This concentration was chosen in order to achieve an injection volume of about 0.15 ml for a mouse with an average body weight of 22 g, corresponding to about 1 mg/mouse. Complete immobilization, defined as absence of the

Developmental delays associated with prenatal alcohol exposure are reversed by thyroid hormone treatment.

Exposure to alcohol in utero has been associated with hypothyroidism and a variety of developmental defects characteristic of thyroid dysfunction. The present work examined whether these abnormalities could be reversed in infant rats treated with thyroid hormones. Subjects were offspring of dams which were on the following diet regimen during gestation: (1) free access to liquid diet containing ethanol (alcohol pups); (2) an equal volume of isocaloric liquid diet (pair-fed pups); or (3) ad libitum control diet (control pups). Neonates from each group were foster-nursed by control dams, and rec

Interactions of cocaine with barbital, pentobarbital and ethanol.

This study deals with the interactions of cocaine with barbital, pentobarbital and ethanol in nontolerant and tolerant male Sprague-Dawley rats. Cocaine hydrochloride (50 mg) pellets implanted s.c. in rats prior to the i.p. injections of sodium barbital (150 mg/kg dose once daily for 4 days) potentiated the hypothermic response 2 hr after the barbital injection, when maximum hypothermia occurred. The s.c. implantation of the same type of pellets prior to the i.p. injections of sodium pentobarbital (75 mg/kg dose once daily for 5 days) potentiated the pentobarbital hypnosis as measured by the d

Interactions of paraldehyde with ethanol and chloral hydrate.

The interaction of the depressant and toxic actions of paraldehyde with those of ethanol and chloral hydrate were investigated in two strains of male mice by determination of the ED50 for anesthetic action (righting reflex loss) and the 24 hr LD50, respectively. In all instances less than simple additive synergism was observed. Onset and duration times for the anesthetic actions of paraldehyde and ethanol were observed to be almost identical. Investigation of the time of maximum toxic effect revealed paraldehyde itself and mixtures containing 40% or more paraldehyde to be more acutely toxic th

Potential neuromuscular toxicity of N'-(2,4-xylyl)-N-methyl formamidine HCl in rodents.

The mammalian neuromuscular toxicity of N'-(2,4-xylyl)-N-methyl formamidine hydrochloride (U-40481A) was evaluated by programmed screening. The LD50 for mice, determined 48 h after single, subcutaneous (s.c.) injections, was 107 mg/kg body wt. Acute toxicity signs included abnormal gait, hindlimb hyperextension, transient hyperactivity followed by a protracted phase of hypoactivity, ataxia, progressive respiratory difficulty, cyanosis, loss of righting reflex, and death. Relatively low doses of U-40481A (1, 5, 10, and 20 mg/kg, s.c.) markedly impaired the ability of trained mice to ride a rota

[Effects of oxythiamine and pyrithiamine on rat liver. --Biochemical and morphological changes in the thiamine deficient rat (author's transl)].

We studied biochemical changes in the rat liver in a thiamine (T) deficient state as induced by oxythiamine (OT), pyrithiamine (PT) and thiamine deficient diet (TDD) and simultaneously observed morphological changes under light and electron microscope. Severe loss of weight was observed in the OT treated rats fed on TDD (OTD) and such was frequently accompanied by a complete loss of righting reflex. Biochemical changes commonly found in T deficient groups were decrease in serum total protein, alkaline phosphatase activity and liver lipids, and increase in serum total cholesterol and transamina

Influences of the sympathetic and parasympathetic nerve transection on cardiovascular reflexes induced by volleys in the IXth nerve fibers of rat.

The effect of vagotomy and sympathectomy on cardio-acceleratory and arterial hypertensive reflexes evoked by electrical stimulation of the rat glossopharyngeal (IXth) nerve was studied in relation to changes in baseline heart rate and arterial blood pressure. Uni- and bi-lateral transection of the cervical vagal trunk brought about augmentation of baseline heart rate, accompanied by a depression in reflex tachycardia; the amount of depression being inversely related to that of an increase in baseline heart rate. The latter increased more after a right vagotomy than after a left vagotomy. No ap

Cerebellar transcranial direct current stimulation modulates the effect of cerebellar transcranial magnetic stimulation on the excitability of spinal reflex.

Cerebellar transcranial magnetic stimulation (C-TMS) facilitates the ipsilateral soleus H-reflex, which reflects the excitability of the spinal motoneuron pool. This study aimed to investigate whether this facilitation of the spinal motoneuron pool excitability by C-TMS is affected by cerebellar transcranial direct current stimulation (ctDCS) in a polarity-specific manner. Eleven healthy adults participated in this study. The H-reflex was measured from the right soleus muscle by electrical stimulation of the right tibial nerve. The conditioning TMS over the right cerebellum was delivered 110 m

White matter lesions associated with the reemergence of grasp reflexes in patients with idiopathic normal pressure hydrocephalus.

BACKGROUND: The white matter damage inducing the reemergence of grasp reflexes and their potential lateralization remains unstudied. Idiopathic normal pressure hydrocephalus (iNPH), a subcortical dementia, is an ideal model for these investigations. We aim to understand the contributions of white matter to the inhibition of grasp reflexes in patients with iNPH. METHODS: A total of 48 patients (mean age at admission: 77.8 ± 5.2 years; 56% male) with probable iNPH were retrospectively enrolled in this study. The intensity of grasp reflexes was assessed using a four-category classification. A vo

Mild and Mild-to-Moderate Traumatic Brain Injury-Induced Significant Progressive and Enduring Multiple Comorbidities.

Traumatic brain injury (TBI) can produce life-long disabilities, including anxiety, cognitive, balance, and motor deficits. The experimental model of closed head TBI (cTBI) induced by weight drop/impact acceleration is known to produce hallmark TBI injuries. However, comprehensive long-term characterization of comorbidities induced by graded mild-to- mild/moderate intensities using this experimental cTBI model has not been reported. The present study used two intensities of weight drop (1.0 m and 1.25 m/450 g) to produce cTBI in a rat model to investigate initial and long-term disability of fo

Effects of prenatal exposure to Tityus bahiensis scorpion venom on rat offspring development.

Scorpion envenoming is a public health problem. In Brazil, the scorpion Tityus serrulatus is considered the most dangerous, but a large number of exposures also occur with Tityus bahiensis. There are quite a few studies in literature about the toxic effects of this venom but it is not known if the venom causes malformations or behavioral defects to the offspring of mothers exposed to the venom during pregnancy. The objective of this work was to determine, in rats, the possible toxic effects of T. bahiensis venom on offspring when injected into rats during different periods of fetal development

Blink reflex, H-reflex and nerve-conduction alterations in leprosy patients.

Peripheral nerve lesions are the most important cause of disability in leprosy patients. Electrophysiological studies are used in the diagnosis and prognosis of neuropathy. Nerve conduction is the most frequently used electrophysiological test method to detect neuropathy, although it evaluates only a part of the peripheral nervous system. Blink reflex and H-reflex are electrophysiological tests which evaluate facial and trigeminal nerve function. This study determined the frequencies of blink reflex, H-reflex and motor and sensory nerve conduction alterations in twenty five heterogeneous, clin

The auricular cough reflex: lateralization, prevalence, procedural determinants, and the role of examiner handedness.

BACKGROUND: The auricular cough reflex (Arnold's reflex) is a vagally mediated somato-visceral response triggered by stimulation of the external auditory canal (EAC). While classically considered rare, its clinical relevance in otologic procedures remains underexplored. OBJECTIVE: This study aimed to evaluate the prevalence, lateralization, and potential risk factors of the Arnold reflex during routine otologic examinations, with particular attention to examiner handedness, hearing aid use, and EAC inflammation. METHODS: In this prospective observational study, 404 consecutive patients under

High-fat diet consumption negatively influences closed-head traumatic brain injury in a pediatric rodent model.

Traumatic brain injury (TBI) is one of the most common causes of emergency room visits in children, and it is a leading cause of death in juveniles in the United States. Similarly, a high proportion of this population consumes diets that are high in saturated fats, and millions of children are overweight or obese. The goal of the present study was to assess the relationship between diet and TBI on cognitive and cerebrovascular outcomes in juvenile rats. In the current study, groups of juvenile male Long Evans rats were subjected to either mild TBI via the Closed-Head Injury Model of Engineered

Innervation of reconstructed bladder above the level of spinal cord injury for inducing micturition by contractions of the abdomen-to-bladder reflex arc.

OBJECTIVE: To establish an artificial bladder reflex arc in dogs via an abdominal reflex pathway above the level of spinal cord injury to reinnervate the neurogenic bladder and restore controllable micturition. METHODS: Ten beagles were used in the experiment. We anastomosed the proximal end of the right T12 ventral root and distal end of the right S2 ventral root by performing autogenous nerve grafting to build an abdomen-to-bladder reflex, whereas the right T12 dorsal root was kept intact. The early function of the reflex arc was evaluated by performing electrophysiological studies as well

Restoring walking after spinal cord injury: operant conditioning of spinal reflexes can help.

People with incomplete spinal cord injury (SCI) frequently suffer motor disabilities due to spasticity and poor muscle control, even after conventional therapy. Abnormal spinal reflex activity often contributes to these problems. Operant conditioning of spinal reflexes, which can target plasticity to specific reflex pathways, can enhance recovery. In rats in which a right lateral column lesion had weakened right stance and produced an asymmetrical gait, up-conditioning of the right soleus H-reflex, which increased muscle spindle afferent excitation of soleus, strengthened right stance and elim

Protective Effects of Prenatal Bacopa monnieri Extract Against Valproic Acid-Induced Autism-like Behavioral and Neurohistological Alterations in Mice.

Autism spectrum disorder (ASD) is a neurodevelopmental condition characterized by impaired social communication and repetitive behaviors. Oxidative stress is increasingly recognized as a key contributor to the neuro-degeneration and behavioral abnormalities associated with ASD. Bacopa monnieri (BM), a medicinal herb with potent antioxidant and neuro-protective properties, has shown promise in mitigating oxidative damage. This study evaluated the preventive effects of BM extract on behavioral and neuro-histological alterations in a valproic acid (VPA)-induced mouse model of ASD. Pregnant mice r
