Cone Density Changes After Repeated Low-Level Red Light Treatment in Children With Myopia.

IMPORTANCE: Repeated low-level red light (RLRL) therapy has emerged as a potential intervention for controlling myopia progression in children. However, its long-term effects on retinal photoreceptors remain relatively unknown. OBJECTIVE: To evaluate changes associated with RLRL therapy on cone photoreceptor density in children with myopia using high-resolution adaptive optics scanning laser ophthalmoscopy (AOSLO). DESIGN, SETTING, AND PARTICIPANTS: This retrospective multicenter cohort study analyzed data collected from January to March 2024, focusing on Chinese children with myopia. All pa

Temporal requirement of the alternative-splicing factor Sfrs1 for the survival of retinal neurons.

Alternative splicing is the primary mechanism by which a limited number of protein-coding genes can generate proteome diversity. We have investigated the role of the alternative-splicing factor Sfrs1, an arginine/serine-rich (SR) protein family member, during mouse retinal development. Loss of Sfrs1 function during embryonic retinal development had a profound effect, leading to a small retina at birth. In addition, the retina underwent further degeneration in the postnatal period. Loss of Sfrs1 function resulted in the death of retinal neurons that were born during early to mid-embryonic devel

Choroideremia: analysis of the retina from a female symptomatic carrier.

PURPOSE: To define the retinal pathology in a 91 year-old affected matriarch of a three-generation choroideremia family with multiple manifesting carriers. METHODS: Tissue from three different retinal areas was processed for immunohistochemistry. The macular area was processed for transmission electron microscopy. Cryosections were studied by indirect immunofluorescence, using well-characterized antibodies to cone cytoplasm, rhodopsin and cone opsins. The affected donor eyes were compared to a postmortem matched normal eye. RESULTS: The retina displayed areas of severe degeneration, with no

Morphological characterization of the retinal degeneration in three strains of mice carrying the rd-3 mutation.

Retinal development in 3 strains of rd-3/rd-3 mutant mice, previously shown to have different rates of degeneration, was studied using light, electron, and immunofluorescence microscopy. The time course and phenotype of the degeneration as well as details on the mechanism of massive photoreceptor cell loss are compared with other known retinal degenerations in mice. Up until postnatal day (P) 10, the retinas of all three strains (RBF, 4Bnr, In-30) develop similarly to those of pigmented and nonpigmented controls. TUNEL-positive cells appear in the outer nuclear layer (ONL) by P14, and reach a

Kcnv2 E151X Mouse Captures Hallmarks of KCNV2-Associated Retinal Dystrophy.

BACKGROUND: KCNV2-associated retinopathy is a rare inherited retinal dystrophy caused by variants in the KCNV2 gene, leading to disrupted photoreceptor behaviour and progressive deterioration of vision. Patients have characteristic electroretinography abnormalities, including reduced cone response, delayed and reduced rod response to low light flashes and paradoxically large rod-driven response to bright flashes of light. To model this condition, we have generated a Kcnv2 E151X mouse line and assessed its structural and functional retinal features. METHODS: We have employed CRISPR/Cas 9 gene

Localization of Usher 1 proteins to the photoreceptor calyceal processes, which are absent from mice.

The mechanisms underlying retinal dystrophy in Usher syndrome type I (USH1) remain unknown because mutant mice lacking any of the USH1 proteins-myosin VIIa, harmonin, cadherin-23, protocadherin-15, sans-do not display retinal degeneration. We found here that, in macaque photoreceptor cells, all USH1 proteins colocalized at membrane interfaces (i) between the inner and outer segments in rods and (ii) between the microvillus-like calyceal processes and the outer segment basolateral region in rods and cones. This pattern, conserved in humans and frogs, was mediated by the formation of an USH1 pro

Cones and cone pathways remain functional in advanced retinal degeneration.

Most defects causing retinal degeneration in retinitis pigmentosa (RP) are rod-specific mutations, but the subsequent degeneration of cones, which produces loss of daylight vision and high-acuity perception, is the most debilitating feature of the disease. To understand better why cones degenerate and how cone vision might be restored, we have made the first single-cell recordings of light responses from degenerating cones and retinal interneurons after most rods have died and cones have lost their outer-segment disk membranes and synaptic pedicles. We show that degenerating cones have functio

Adaptive-optics scanning laser ophthalmoscopy study of non-arteritic anterior ischemic optic neuropathy.

OBJECTIVE: To investigate the changes in cone photoreceptors in patients with non-arteritic anterior ischemic optic neuropathy (NAION). METHODS: Adaptive-optics scanning laser ophthalmoscopy (AO-SLO) was performed in patients with NAION and healthy participants. Acute NAION was defined as a disease duration of <2 months, whereas chronic NAION was defined as a duration of >2 months. We analyzed four quadrants (superior, inferior, nasal, and temporal with respect to the central fovea of the macula) of the cone layer images at 2.4° × 2.4° collected at 3° eccentricity. Patients with NAION were fu

Loss of ABCA4 from photoreceptor discs is associated with glial transcriptomic changes in retinal organoids.

BACKGROUND: Loss-of-function mutations in the ABCA4 gene cause Stargardt disease (STGD1), the most common inherited macular dystrophy leading to progressive central vision loss. METHODS: Here, we generated human induced pluripotent stem cell-derived retinal organoids harboring a premature stop codon in exon-24 of ABCA4 to evaluate the impact of this mutation on mRNA and protein levels in a human model. RESULTS: Immunofluorescence analysis revealed the absence of ABCA4 protein in the mutant photoreceptor outer segment discs, while single-cell RNA sequencing detected no major transcriptional a

AMIGO1 Promotes Axon Growth and Territory Matching in the Retina.

Dendrite and axon arbor sizes are critical to neuronal function and vary widely between different neuron types. The relative dendrite and axon sizes of synaptic partners control signal convergence and divergence in neural circuits. The developmental mechanisms that determine cell-type-specific dendrite and axon size and match synaptic partners' arbor territories remain obscure. Here, we discover that retinal horizontal cells express the leucine-rich repeat domain cell adhesion molecule AMIGO1. Horizontal cells provide pathway-specific feedback to photoreceptors-horizontal cell axons to rods an

The human THRB thyroid hormone receptor gene and the puzzle of retinal disease phenotypes.

The THRB thyroid hormone receptor gene has attracted growing attention for its role in retinal development and disease. This gene has particularly critical roles in cone photoreceptors, the specialized cells that mediate color vision and high acuity vision. THRB controls the diversity of cone types that are required for color vision and influences cone survival in mammalian model species and human retinal organoid cultures. This central role of THRB in the cone life history prompts an expectation of equally critical roles in the human retina in vivo. Puzzlingly, overt retinal phenotypes have g

Rapid and Progressive Loss of Multiple Retinal Cell Types in Cathepsin D-Deficient Mice-An Animal Model of CLN10 Disease.

Vision loss is among the characteristic symptoms of neuronal ceroid lipofuscinosis (NCL), a fatal neurodegenerative lysosomal storage disorder. Here, we performed an in-depth analysis of retinal degeneration at the molecular and cellular levels in mice lacking the lysosomal aspartyl protease cathepsin D, an animal model of congenital CLN10 disease. We observed an early-onset accumulation of storage material as indicated by elevated levels of saposin D and subunit C of the mitochondrial ATP synthase. The accumulation of storage material was accompanied by reactive astrogliosis and microgliosis,

Midget ganglion cell anatomy and center structure: More complex than it looks.

It is usually assumed that the center of the midget ganglion cell in the primate retina derives input from a single cone in and near the fovea, providing a 'private line to the brain'. Thus, the physiological center should have the dimensions of a single cone sampling aperture. Centers measured physiologically are much larger, partly due to optical blur. With retinal eccentricity, at 10 degrees eccentricity cones are larger but optics do not change much. Yet physiological centers have much increased in size and are still much larger than expected from the anatomy. As retinal eccentricity incre

Gene therapy for retinitis pigmentosa and Leber congenital amaurosis caused by defects in AIPL1: effective rescue of mouse models of partial and complete Aipl1 deficiency using AAV2/2 and AAV2/8 vectors.

Defects in the photoreceptor-specific gene encoding aryl hydrocarbon receptor-interacting protein-like 1 (AIPL1) are clinically heterogeneous and present as Leber Congenital Amaurosis, the severest form of early-onset retinal dystrophy and milder forms of retinal dystrophies such as juvenile retinitis pigmentosa and dominant cone-rod dystrophy. Micro-abnormalities in the retina and choroid induced by anti-CTLA4 treatment.

Anti-Cytotoxic T-Lymphocyte Associated protein 4 agents, such as ipilimumab, are widely applied to various cancers. However, they cause immune-related adverse effects throughout the body, including the eye. This study examined whether ipilimumab induces retinal and choroidal abnormalities in rodents, and investigated potential underlying mechanisms. Female wild-type mice were injected with ipilimumab three times/week for 5 weeks. The mice underwent optical coherence tomography (OCT) on the first day of the 6th week. Retinal function and morphology were evaluated by light microscopy, immunohist

AAV-RPGR Gene Therapy Rescues Opsin Mislocalisation in a Human Retinal Organoid Model of RPGR-Associated X-Linked Retinitis Pigmentosa.

Variants within the Retinitis Pigmentosa GTPase regulator (RPGR) gene are the predominant cause of X-Linked Retinitis Pigmentosa (XLRP), a common and severe form of inherited retinal disease. XLRP is characterised by the progressive degeneration and loss of photoreceptors, leading to visual loss and, ultimately, bilateral blindness. Unfortunately, there are no effective approved treatments for RPGR-associated XLRP. We sought to investigate the efficacy of RPGRORF15 gene supplementation using a clinically relevant construct in human RPGR-deficient retinal organoids (ROs). Isogenic RPGR knockout

Synthesis and secretion of interstitial retinol-binding protein by the human retina.

Interstitial retinol-binding protein (IRBP) is a soluble glycoprotein present between the retina and pigmented epithelium, which may function to shuttle vitamin A derivatives between these tissues. While previous studies have shown that the retina is solely responsible for IRBP synthesis, the specific retinal cell(s) in which this occurs has not been established. Since the carbohydrate moiety of IRBP contains fucose, the authors have analyzed the sites of incorporation of 3H-fucose in the human retina in vitro, using autoradiography. Following a 30-min pulse incubation, all retinal layers exhi

Genetic variations strongly influence phenotypic outcome in the mouse retina.

Variation in genetic background can significantly influence the phenotypic outcome of both disease and non-disease associated traits. Additionally, differences in temporal and strain specific gene expression can also contribute to phenotypes in the mammalian retina. This is the first report of microarray based cross-strain analysis of gene expression in the retina investigating genetic background effects. Microarray analyses were performed on retinas from the following mouse strains: C57BL6/J, AKR/J, CAST/EiJ, and NOD.NON-H2(-nb1) at embryonic day 18.5 (E18.5) and postnatal day 30.5 (P30.5). O
