Steroid hormone-deprived sex reversal in cyp11a1 mutant XX tilapia experiences an ovary-like stage at molecular level.

Fish sex is largely influenced by steroid hormones, especially sex hormones. Here, we established a steroid hormone-free genetic model by mutation of cyp11a1 in Nile tilapia, which was confirmed by EIA assay. Gonadal phenotype and transcriptome analyses showed that the XX mutants displayed sex reversal from female to male but with defective spermatogenesis. Despite the sex reversal, the aromatase encoding gene cyp19a1a was continuously expressed in the gonads of the XX mutants, which might be caused by androgen deficiency. Whole-mount fluorescence in situ hybridization and transcriptome analys

Neural crest-derived neurons invade the ovary but not the testis during mouse gonad development.

Testes and ovaries undergo sex-specific morphogenetic changes and adopt strikingly different morphologies, despite the fact that both arise from a common precursor, the bipotential gonad. Previous studies showed that recruitment of vasculature is critical for testis patterning. However, vasculature is not recruited into the early ovary. Peripheral innervation is involved in patterning development of many organs but has been given little attention in gonad development. In this study, we show that while innervation in the male reproductive complex is restricted to the epididymis and vas deferens

Equine disorders of sexual development in 17 mares including XX, SRY-negative, XY, SRY-negative and XY, SRY-positive genotypes.

We described the clinical, cytogenetic and molecular findings of 17 clinical equine cases presented for abnormal sexual development and infertility. Six horses with an enlarged clitoris had an XX, SRY-negative genotype, which displayed male-like behavior (adult individuals). Bilateral ovotestes were noted in 2 of those cases, while another case showed increased levels of circulating testosterone. Six horses with a female phenotype, including normal external genitalia, had an XY, SRY-negative genotype. These individuals had small gonads and an underdeveloped internal reproductive tract. Four ho

Regulation of the amh gene promoter activity by SF-1 in sex-reversing Cheilinus undulatus and its adaptive evolution.

BACKGROUND: The humphead wrasse (Cheilinus undulatus) is a protogynous hermaphroditic fish and a top predator in coral reef ecosystems. However, overfishing has led to a significant decline in its population, making it challenging to study its adaptive evolution and sex reversal mechanisms. RESULTS: In this study, comparative genomics analyses revealed adaptive evolutionary signals associated with feeding capacity, digestive function, and color pattern transformation in C. undulatus. Notably, positive selection signals were detected in the key masculinization pathway gene amh in C. undulatus,

Male sex determination maintains proteostasis and extends lifespan of daf-18/PTEN deficient C. elegans.

Although females typically have a survival advantage, those with PTEN functional abnormalities face a higher risk of developing tumors than males. However, the differences in how each sex responds to PTEN dysfunction have rarely been studied. We use Caenorhabditis elegans to investigate how male and hermaphrodite worms respond to dysfunction of the PTEN homolog daf-18. Our study reveals that male worms can counterbalance the negative effects of daf-18 deficiency, resulting in longer adult lifespan. The survival advantage depends on the loss of DAF-18 protein phosphatase activity, while its lip

Identification of BiP as a temperature sensor mediating temperature-induced germline sex reversal in C. elegans.

Sex determination in animals is not only determined by karyotype but can also be modulated by environmental cues like temperature via unclear transduction mechanisms. Moreover, in contrast to earlier views that sex may exclusively be determined by either karyotype or temperature, recent observations suggest that these factors rather co-regulate sex, posing another mechanistic mystery. Here, we discovered that certain wild-isolated and mutant C. elegans strains displayed genotypic germline sex determination (GGSD), but with a temperature-override mechanism. Further, we found that BiP, an ER cha

The DSM diagnostic criteria for gender identity disorder in adolescents and adults.

Apart from some general issues related to the Gender Identity Disorder (GID) diagnosis, such as whether it should stay in the DSM-V or not, a number of problems specifically relate to the current criteria of the GID diagnosis for adolescents and adults. These problems concern the confusion caused by similarities and differences of the terms transsexualism and GID, the inability of the current criteria to capture the whole spectrum of gender variance phenomena, the potential risk of unnecessary physically invasive examinations to rule out intersex conditions (disorders of sex development), the

First evidence of higher female recombination in a species with temperature-dependent sex determination: the saltwater crocodile.

The first evidence of genetic linkage and sex-specific recombination in the order Crocodylia is reported. This study was conducted using a resource pedigree of saltwater crocodiles consisting of 16 known-breeding pairs (32 adults) and 101 juveniles. A total of 21 microsatellite loci were available for analysis. Ten of the 21 loci showed linkage with 4 linkage groups: 3 pairwise (Cj131/Cj127, CUD68/Cj101, and Cj107/Cp10) and 1 four-locus (Cj122, CUD78, Cj16, and Cj104) being found. Linkage analysis on the 21 loci revealed evidence of sex-specific differences in recombination rates. All 5 nonzer

Developmental alterations as a result of in ovo exposure to the pesticide metabolite p,p'-DDE in Alligator mississippiensis.

The pesticide metabolite p,p'-DDE (1,1-dichloro-2,2-bis (p-chlorophenyl) ethylene), has been implicated in numerous reproductive and developmental abnormalities. Previous work has shown this ubiquitous contaminant to behave in an estrogenic or antiandrogenic manner, depending on the species and endpoints examined. In the current study, we examined the effects of embryonic exposure to p,p'-DDE in the alligator, a species that exhibits temperature-dependent sex determination. We compared sex ratios at an intermediate and all male producing temperature, as well as plasma testosterone (T) and gona

Contaminant-induced endocrine disruption in wildlife.

Environmental contaminants have posed a threat to the health of wildlife since the onset of the industrial age. Over the last four decades, much concern has focused on the lethal, carcinogenic and/or extreme teratogenic manifestations of environmental pollution. During the last decade, evidence suggests that the disruption of normal endocrine signalling by environmental contaminants is a mechanism that must also be examined. Man-made chemicals, known as xenochemicals, released into the environment can act as hormone agonists or antagonists and thereby disrupt hormone synthesis, action and meta

The male-determining activity on the Y chromosome of the housefly (Musca domestica L.) consists of separable elements.

In the common housefly, the presence or absence of a male-determining factor, M, is responsible for sex determination. In different strains, M has been found on the Y, on the X, or on any of the five autosomes. By analyzing a Y-autosomal translocation and a ring-shaped, truncated Y chromosome, we could show that M on the Y consists of at least two regions with M activity: One of them can be assigned to the short arm of the Y chromosome (MYS), which is largely C-banding negative, the other region lies on the C-banding positive long arm of the Y, including the centromeric part (MYL). Each region

Sex determination factor, a novel male-linked gene in the sea cucumber Apostichopus japonicus: Molecular characterization, expression patterns and effects of gene knockdown.

Apostichopus japonicus is a highly significant marine aquaculture species. Research findings have indicated that male sea cucumbers demonstrate a more rapid growth rate compared to females, underscoring the potential advantages of establishing an all-male population. In this study, we identified a specific protein-coding gene (ORFan) within a 4565 bp male fragment and named it sex determination factor (sdf). The sdf transcript exhibited ubiquitous expression in various adult male tissues, along with dynamic expression patterns in the testis across different developmental stages. Notably, knock

Screening a new European hake (Merluccius merluccius) chromosome-level genome assembly suggests an XX/XY sex-determining system driven by the SRY-box transcription factor 3 (sox3).

Sex determination is exceptionally diverse and shows a high evolutionary rate in fish. European hake (Merluccius merluccius) is a species of great commercial value distributed throughout European coasts, which displays a significant sexual growth dimorphism. We present a chromosome-level genome of M. merluccius, composed of 215 contigs using long- and short-read sequencing, further scaffolded into the species' 21 chromosomes using Hi-C technique (715 Mb). RNA-seq on several tissues from pooled individuals improved annotation (26,625 protein-coding genes and 11,083 ncRNAs). Five males and 5 fem

A transgenic tool to assess Anopheles mating competitiveness in the field.

BACKGROUND: Malaria parasites, transmitted by the bite of an anopheline mosquito, pose an immense public health burden on many tropical and subtropical regions. The most important malaria vectors in sub-Saharan Africa are mosquitoes of the Anopheles gambiae complex including An. gambiae (sensu stricto). Given the increasing rates of insecticide resistance in these mosquitoes, alternative control strategies based on the release of genetically modified males are being evaluated to stop transmission by these disease vectors. These strategies rely on the mating competitiveness of release males, ho

Autosomal gsdf acts as a male sex initiator in the fish medaka.

Sex is pivotal for reproduction, healthcare and evolution. In the fish medaka, the Y-chromosomal dmy (also dmrt1bY) serves the sex determiner, which activates dmrt1 for male sex maintenance. However, how dmy makes the male decision via initiating testicular differentiation has remained unknown. Here we report that autosomal gsdf serves a male sex initiator. Gene addition and deletion revealed that gsdf was necessary and sufficient for maleness via initiating testicular differentiation. We show that gsdf transcription is activated directly by dmy. These results establish the autosomal gsdf as t

Spontaneous polyploidy, gynogenesis and androgenesis in second generation (F2 ) koi Cyprinus carpio × goldfish Carassius auratus hybrids.

The objective of this study was to characterize the genetics of second generation (F2 ) koi Cyprinus carpio × goldfish Carassius auratus hybrids. Spermatozoa produced by a novel, fertile F1 male were found to be diploid by flow-cytometric analysis. Backcross (F1 female × C. carpio male and C. carpio female × F1 male) juveniles were triploid, confirming that female and male F1 hybrids both produced diploid gametes. The vast majority of surviving F2 juveniles was diploid and small proportions were aneuploid (2·1n-2·3n and 3·1n-3·9n), triploid (3n) and tetraploid (4n). Microsatellite genotyping s

Integrating developmental biology and the fossil record of reptiles.

Numerous new discoveries and new research techniques have influenced our understanding of reptile development from a palaeontological perspective. They suggest for example that transition from mineralized to leathery eggshells and from oviparity to viviparity appeared much more often in the evolution of reptiles than was previously thought. Most marine reptiles evolved from viviparous terrestrial ancestors and had probably genetic sex determination. Fossil forms often display developmental traits absent or rare among modern ones such as polydactyly, hyperphalangy, the presence of ribcage armou

Role of platelet-derived growth factors in the testis.

Normal development and function of the testis are controlled by endocrine and paracrine signaling pathways. Platelet-derived growth factors (PDGFs) are growth factors that mediate epithelial-mesenchymal interactions in various tissues during normal and abnormal processes such as embryo development, wound healing, tissue fibrosis, vascular disorders, and cancer. PDGFs and their receptors (PDGFRs) have emerged as key players in the regulation of embryonic and postnatal development of the male gonad. Cells that express PDGFs and PDGFRs are found in the testis of mammals, birds, and reptiles, and

Single-cell exploration of gonadal somatic cell lineage specification during human sex determination.

Gonad development is an exciting model to study cell fate commitment. A better understanding of sex determination requires the identification of all involved cell types and their dynamic expression programs. Here, we present an atlas of 128,000 single cells from human gonads between 5 and 12 post-conceptional weeks. A focused analysis of somatic cells uncovered a population of bipotential progenitors derived from the coelomic epithelium of both testes and ovaries, which may have the capacity to commit to either a steroidogenic or a supporting fate. Moreover, our analyses suggest that early sup
