Identification and Molecular Characterisation of a Novel Mu-Like Bacteriophage, SfMu, of Shigella flexneri.

S. flexneri is the leading cause of bacillary dysentery in the developing countries. Several temperate phages originating from this host have been characterised. However, all S. flexneri phages known to date are lambdoid phages, which have the ability to confer the O-antigen modification of their host. In this study, we report the isolation and characterisation of a novel Mu-like phage from a serotype 4a strain of S. flexneri. The genome of phage SfMu is composed of 37,146 bp and is predicted to contain 55 open reading frames (orfs). Comparative genome analysis of phage SfMu with Mu and other

Recovery and characterization of environmental variants of Shigella flexneri from surface water in Bangladesh.

Little is known about the distribution, survival, and transmission of Shigella in environmental surface waters. To gain more insight into the environmental biology of Shigella we isolated five bacterial strains serotyped as Shigella flexneri 2b from a freshwater lake in Bangladesh using a modified nutrient broth supplemented with nucleic acid bases. The biochemical properties of the isolates, including inability to ferment lactose and a negative lysine decarboxylase test, indicated common physiological characteristics with Shigella, but differed significantly from that of standard clinical str…

[Structure of the oligosaccharide region (core) of the lipopolysaccharides of Shigella flexneri types 2a and 5b].

The full structure of the lipopolysaccharide core of bacteria Shigella flexneri types 2a and 5b, the causative agents of bacillary dysentery (shigellosis), was established by chemical methods, high-resolution electrospray ionization mass spectrometry, and two-dimensional NMR spectroscopy. The structure of the O-antigen repeating unit and the configuration and position of the linkage between the O-antigen and the core were determined in the lipopolysaccharide of S. flexneri type 2a.

IpaD localizes to the tip of the type III secretion system needle of Shigella flexneri.

Shigella flexneri, the causative agent of shigellosis, is a gram-negative bacterial pathogen that initiates infection by invading cells within the colonic epithelium. Contact with host cell surfaces induces a rapid burst of protein secretion via the Shigella type III secretion system (TTSS). The first proteins secreted are IpaD, IpaB, and IpaC, with IpaB and IpaC being inserted into the host cell membrane to form a pore for translocating late effectors into the target cell cytoplasm. The resulting pathogen-host cross talk results in localized actin polymerization, membrane ruffling, and, ultim…

A DNA adenine methylase mutant of Shigella flexneri shows no significant attenuation of virulence.

Mutants of Salmonella defective in DNA adenine methylase (dam) have been reported to be attenuated for virulence and to provide protective immunity when used as vaccine strains. To determine whether these observations could be extended to Shigella, a dam mutant of Shigella flexneri 2a was characterized and examined for the role of dam in pathogenesis. The Shigella dam mutant showed some unique characteristics; however, it retained virulence in vivo as well as in vitro. The mutant invaded cultured L2 monolayer cells as efficiently as the wild-type parent, but its intracellular growth was suppre…

dksA is required for intercellular spread of Shigella flexneri via an RpoS-independent mechanism.

Pathogenesis of Shigella flexneri is dependent on the ability of the bacterium to invade and spread within epithelial cells. In this study, we identified dksA as a gene necessary for intercellular spread in, but not invasion of, cultured cells. The S. flexneri dksA mutant exhibited sensitivity to acid and oxidative stress, in part due to an effect of DksA on production of RpoS. However, an S. flexneri rpoS mutant formed plaques on tissue culture monolayers, thus excluding DksA regulation of RpoS as the mechanism responsible for the inability of the dksA mutant to spread intercellularly. Intrac…

[The biological characteristics of Shigella flexneri strains circulating in Saint Petersburg in 1992-1994].

S.flexneri 2a strains isolated in 1992-1994 from 371 patients in sporadic cases, from 90 patients and their contacts in cases of group diseases and from 71 lethal cases of Flexner's dysentery, were studied in comparison with strains isolated in 1980s. The study revealed the biological heterogeneity of S.flexneri 2a strains. The circulation of 3 enzymatic variants and 11 variants differing in their sensitivity to colicins from Frederick's collection was established. The overwhelming majority of strains circulating in 1990s were resistant to 6-7 antibiotics, while the strains isolated in 1980s w

MxiD, an outer membrane protein necessary for the secretion of the Shigella flexneri lpa invasins.

The invasive phenotype of Shigella flexneri is conferred by a 220 kb virulence plasmid, pWR100, that encodes both the lpa proteins, which are involved in the entry process, and factors which are required for the export and correct localization of the lpa proteins. We have characterized the mxiD gene, whose expression, like that of the ipa operon, is regulated by temperature. After inactivation of mxiD, the mutant strain was unable to invade HeLa cells and to provoke keratoconjunctivitis in guinea-pigs. Analysis of culture supernatants indicated that wild-type S. flexneri secretes about nine po…

Role of antigen form in development of mucosal immunoglobin A response to Shigella flexneri Antigens.

One major stumbling block in the development of an effective means to immunize against shigellosis and other enteric diseases has been the lack of a means to assess sequential mucosal immune responses to different potential immunogens. In the present study, we compared the abilities of live invasive organisms, noninvasive organisms, and nonviable antigen preparations of shigella to elicit mucosal immune responses. Whereas previous studies have found that effective immunity was produced best by vaccination with live invasive strains of shigella, in the present study, live noninvasive strains th

[Distribution of lysotypes and study of antibiotic sensitivity of Shigella flexneri 2 isolated in Iran].

152 strains of Shigella flexneri 2 isolated from the stools of patients having sporadic bacillary dysenteria have been studied for their phage types as well as for the presence of transferable resistance factor. The distribution of the phage types has shown a high predominance of lysotype 12 (67%). Proportion of Shigella flexneri 2 strains resistant to one or several antibiotics has been highest in the years 1968 and 1973. The most frequent transferable resistance pattern was found to be (Tc, Cm, Sm, Su) and the most successful transfer method was the celophane technique. Resistance loss varie…

Persistence of an Escherichia coli-Shigella flexneri hybrid in the intestinal tract of Macaca mulatta.

A live oral vaccine prepared from an Escherichia coli-Shigella flexneri 4b hybrid was administered by gavage to Macaca mulatta. In the three main studies involving 160 monkeys, three doses were used, generally consisting of 5 x 10(9) to 60 x 10(9) cells per dose, with an interval between doses of 6, 7, or 14 days. Rectal swab cultures at the time of the last vaccine dose, and 14 to 58 days later, revealed that the hybrid persisted in the intestinal tract for at least 7 days in 16%, and for at least 21 days in 8%, of the monkeys. Our findings are comparable to those of Formal et al. for sheddin…

Multiple drug-resistance in Shigella flexneri isolated from a patient with human immunodeficiency virus.

Persistent shigellosis, due to Shigella flexneri resistant to multiple antibiotics, developed in a 40-yr-old homosexual man with human immunodeficiency virus infection. The Shigella strain demonstrated resistance to ampicillin, tetracycline, and trimethoprim-sulfamethoxazole. Although Shigella flexneri isolates resistant to trimethoprim-sulfamethoxazole are uncommon in the United States, laboratories should monitor resistance patterns through routine in vitro susceptibility testing.

Differences in tetracycline resistance determinant carriage among Shigella flexneri and Shigella sonnei are not related to different plasmid Inc-type carriage.

OBJECTIVES: The aim of this study was to establish the prevalence of the most common molecular mechanisms involved in tetracycline resistance as well as their relationship with plasmid incompatibility (Inc) groups in a collection of Shigella spp. causing traveller's diarrhoea. METHODS: Tetracycline susceptibility was established in 187 Shigella spp. (74 Shigella flexneri and 113 Shigella sonnei), of which 153 isolates were recovered as a confirmed cause of traveller's diarrhoea. The prevalence of the tet(A), tet(B) and tet(G) genes was analysed by PCR. Eighteen plasmid Inc groups was determin…

Shigella antigen-specific B memory cells are associated with decreased disease severity in subjects challenged with wild-type Shigella flexneri 2a.

The role of Shigella-specific B memory (BM) in protection has not been evaluated in human challenge studies. We utilized cryopreserved pre- and post-challenge peripheral blood mononuclear cells and sera from wild-type Shigella flexneri 2a (wt-2457T) challenges. Challenged volunteers were either naïve or subjects who had previously ingested wt-2457T or been immunized with hybrid Escherichia coli-Shigella live oral candidate vaccine (EcSf2a-2). BM and antibody titers were measured against lipopolysaccharide (LPS) and recombinant invasion plasmid antigen B (IpaB); results were correlated with dis…

Inhibitory Effect of Terminalia chebula Retz. on MDR Shigella flexneri Invasion of HeLa Cells.

INTRODUCTION: Shigella flexneri causes bacillary dysentery by invading the colon epithe-lium, leading to inflammation and tissue damage. The emergence of multidrug-resistant (MDR) Shi-gella strains and the lack of effective vaccines have highlighted the need for alternative treatments. Natural plant compounds have been explored as potential antimicrobial agents. This study evaluates the antibacterial activity of ethanolic (70%-96%) and methanolic (70%-99.9%) extracts of Termi-nalia chebula Retz. against MDR S. flexneri and their effect on bacterial invasion of HeLa cells in vitro. We assessed …

Cysteine-Dependent Conformational Heterogeneity of Shigella flexneri Autotransporter IcsA and Implications of Its Function.

Shigella IcsA is a versatile surface virulence factor required for early and late pathogenesis stages extracellularly and intracellularly. Despite IcsA serving as a model Type V secretion system (T5SS) autotransporter to study host-pathogen interactions, its detailed molecular architecture is poorly understood. Recently, IcsA was found to switch to a different conformation for its adhesin activity upon sensing the host stimuli by Shigella Type III secretion system (T3SS). Here, we reported that the single cysteine residue (C130) near the N terminus of the IcsA passenger had a role in IcsA adhe
