Pigmentation is a polygenic trait encompassing some of the most visible phenotypic variation observed in humans. Here we present direct estimates of selection acting on functional alleles in three key genes known to be involved in human pigmentation pathways--HERC2, SLC45A2, and TYR--using allele frequency estimates from Eneolithic, Bronze Age, and modern Eastern European samples and forward simulations. A 52-year-old woman with bullous pemphigoid developed a bizarre figurate erythematous eruption and generalized pigmentation and velvety skin thickening. Later on, these cutaneous signs were found to be associated with an underlying adenocarcinoma of the stomach. The complex of myxomas, spotty skin pigmentation and endocrine overactivity, or Carney complex (CNC), is a familial multiple endocrine neoplasia and lentiginosis syndrome.
