The formation of amyloid fibrils by the intrinsically disordered protein α-synuclein is a hallmark of Parkinson disease. Corannulene dimers made up of corannulene monomers with different curvature and substituents were studied using M06-2X, B97D and ωB97XD functionals and 6-31+G* basis set. Corannulene molecules were substituted with five alternating Br, Cl, CH(3), C(2)H or CN units. The half-angle of an ac cone is shown to exhibit a universal (length-scale independent) asymptotic value of 12.6 degrees at high-permittivity ratio, which is quite distinct from the 49.3 degrees of dc Taylor cones. Our theory and experiments suggest that ac entrainment of low-mobility anions, generated by field-assisted ion dissociation, sustains a net charge at the ac cone. Mutual Coulombic repulsion between these free charges compensates the singular azimuthal capillary force and elongates the cone with an axial force that does not exist in dc cones.

BACKGROUND: Urate oxidase (EC 1.7.3.3 or UOX) catalyzes the conversion of uric acid and gaseous molecular oxygen to 5-hydroxyisourate and hydrogen peroxide, in the absence of cofactor or particular metal cation. The functional enzyme is a homo-tetramer with four active sites located at dimeric interfaces.
RESULTS: The catalytic mechanism was investigated through a ternary complex formed between the enzyme, uric acid, and cyanide that stabilizes an intermediate state of the reaction. When uric acid is replaced by a competitive inhibitor, no complex with cyanide is formed.
