Expression of the surface protein Cnm has been directly implicated in the ability of certain strains of Streptococcus mutans to bind to collagen and to invade human coronary artery endothelial cells (HCAEC) and in the killing of Galleria mellonella. Sequencing analysis of Cnm(+) strains revealed that cnm is located between the core genes SMU.2067 and SMU.2069. Reverse transcription-PCR (RT-PCR) analysis showed that cnm is cotranscribed with SMU.2067, encoding a putative glycosyltransferase referred to here as PgfS (protein glycosyltransferase of streptococci). Notably, Cnm contains a threonine…

The pH-inducible acid tolerance response (ATR) is believed to play a major role in acid adaptation and virulence of Streptococcus mutans. To study this phenomenon in S. mutans JH1005, differential display PCR was used to identify and clone 13 cDNA products that had increased expression in response to pH 5.0 compared to that of pH 7.5-grown cells. One of these products, confirmed to be pH inducible by RNA dot blot and reverse transcription-PCR analyses, had 67% identity to a uvrA-UV repair excinuclease gene in Bacillus subtilis. Further sequence analysis of the uvrA homologue using the S. mutan…

Plaque-forming ability and cariogenic virulence of Streptococcus mutans AHT mutants having varying in vitro adherence abilities were studied in hamsters reared on a high sucrose diet over a period of 64 days. The parent wild-type strain produced heavy plaque and extensive carious lesions on molars. However, mutants M1, M2, and M13, which have lost the ability to form adhesive deposits on glass when grown in sucrose broth, and mutant M9, which adheres firmly to glass but in an extremely small amount, could not be recovered from the mouths of animals even after repeated inoculations at weekly in…

The relationship of the salivary concentration of Streptococcus mutans and host age to the colonization of this organism on erupting teeth was studied in humans. Plaques were obtained from fissures and buccal surfaces of erupting permanent first and second molars of children 6 to 7 and 11 to 14 years old, respectively. In subjects of both age groups with salivary S. mutans concentrations below 5 x 10(2) colony-forming units per ml, the organism was detected on only a few of the tooth surfaces; at concentrations of 5 x 10(2) to 4.9 x 10(4) colony-forming units per ml more than half of the surfa…

BACKGROUND: Biofilms contribute to the dental calculus formation, which in turn deteriorates oral and systemic health. The use of antimicrobial photodynamic therapy (PDT) may be limited by problems with photosensitizer penetration. In addition, toxicity issues severely limit the potential of PDT. In this study, the selected photosensitizer, curcumin (Cur), was loaded into ethosome (Eth) nanocarriers to enhance the effect of Cur for potential PDT against the mature Streptococcus mutans biofilm.
METHODS: The Cur-Eth formulation was characterized and evaluated for its PDT efficacy in destroying …

Dental caries is a multi-factorial oral infectious disease worldwide. Among the 600 different types of bacteria inhabiting the oral cavity, Streptococcus mutans is considered the most prolific colonizer contributing to dental caries. Although fluorides and broad-spectrum antimicrobials such as chlorhexidine remain the primary approaches for caries treatment, natural flavonoids can serve as helpful alternatives. Although flavonoids are effective against different biofilm-forming pathogens, in the present review, attempts have been made to emphasize the antibiofilm properties of only such flavon…

Streptococcus mutans is a major cariogenic pathogen whose proliferation, acidogenesis, and pathogenicity are strongly impacted by dietary sugars, resulting in dental biofilm-associated illnesses. This study evaluated the effects of sucrose, glucose, and alternative sweeteners (allulose, erythritol, stevia, sucralose, and xylitol) on S. mutans biofilm formation, adhesion, autoaggregation, acidogenicity, metabolic activity, and virulence-related gene expression across different dental surfaces. Sucrose significantly enhanced acid production and biofilm formation, with 3% sucrose resulting in the…

Microbes use quorum-sensing systems to respond to ecological and environmental changes. In the oral microbiome, the pathogenic bacterium Streptococcus mutans uses quorum-sensing to control the production of bacteriocins. These antimicrobial peptides kill off ecological competitors and allow S. mutans to dominate the microenvironment of dental plaques and form dental caries. One class of bacteriocins produced by S. mutans, the lantibiotic mutacins, are particularly effective at killing due to their broad spectrum of activity. Despite years of study, the regulatory mechanisms governing productio…

Streptococcus mutans is a major cause of dental caries worldwide. Targeted therapeutic strategies to eradicate S. mutans include oral phage rinses. In this study, we investigated how phage resistance develops in S. mutans. As a model phage, we used ɸAPCM01, which is known to infect a serotype e strain. We isolated and sequenced the genomes of 15 spontaneous resistant mutants and found that 10 had acquired novel CRISPR spacers targeting the phage, with a total of 18 new spacers identified. Additionally, eight strains contained mutations in rhamnose-glucose polysaccharide (RGP) biosynthetic gene…

BACKGROUND: Dental caries is a widespread global health issue strongly associated with Streptococcus mutans. Bacteriophage-derived lytic enzymes such as ClyR hold considerable promise as antibacterial potential, but the molecular mechanisms underlying their activity against S. mutans remain unclear.
OBJECTIVE: This study aimed to determine the role of water-insoluble exopolysaccharides (EPS) in mediating the antibacterial activity of ClyR against S. mutans.
DESIGN: We compared the antibacterial effects of ClyR on S. mutans UA159 and its ΔgtfB mutant, which is characterized by reduced synthes…

BACKGROUND: Streptococcus mutans harbors collagen-binding protein genes, namely cnm and cbm, which are implicated in its virulence and pathogenicity in both oral and extraoral infections. Although both genes were initially identified in S. mutans isolated from Japanese populations, their geographical prevalence, distribution, and genetic relatedness within Japan remain largely unexplored. This study investigates the prevalence of S. mutans strains carrying cnm and cbm genes across Japan, correlates these findings with clinical data, and analyzes the genetic relatedness of cnm-positive and cnm-…

S. mutans is a key pathogen in dental caries initiation and progression. It promotes oral biofilm dysbiosis and biofilm acidification. Sodium resinate is a salt of pine-oil-derived resin which has antimicrobial properties. Pine-oil-derived resin consists of terpenes, diterpenes, and abietic acids. The aim of this study was to determine the effects of pine (Pinus sylvestris) oil resinate (RS) on growth and acid production of cariogenic S. mutans strains in planktonic form and biofilm. The S. mutans type strain NCTC10449 and clinical isolate CI2366 were grown on 96-well plates for testing of RS

Early childhood dental caries (ECC) is the most common chronic disease among children, especially among low socioeconomic populations. Streptococcus mutans is most frequently associated with initiation of ECC. Although many studies report children with multiple S. mutans strains (i.e., genotypes) have greater odds of developing ECC, studies investigating intraspecies interactions in dental caries are lacking. This study investigates the impact of intraspecies interactions on cariogenic and fitness traits of clinical S. mutans isolates using in vitro and in vivo approaches. Association analysis…
