A Brake on Toxicity: Iron Suppresses Organic-Induced Superoxide Generation in Macrophages.

Iron (Fe) is the predominant metal in brake wear particles (BWPs) and urban particulate matter, yet its role in modulating oxidative stress toxicity remains poorly understood. Here, we show that BWPs induce slight cellular superoxide (•O2-) at low doses (<10 μg mL-1) but suppress it dose-dependently at higher doses. In contrast, highway PM2.5 neither significantly induces nor suppresses cellular •O2- generation. Using Fe alone or mixed with isoprene secondary organic aerosols or 9,10-phenanthrenequinone, typical organic compounds known to induce cellular •O2- via nicotinamide adenine dinucleot

A Curcumin-Based Turn-on Fluorescent Probe for Superoxide Anion in Living Cells.

Superoxide (O2•-) plays a pivotal role in diverse physiological and pathological processes, a thorough understanding of its molecular mechanisms relies on dependable detection techniques. In this study, we designed and synthesized DFCD, a novel curcumin-based fluorescent probe with a switching mechanism, for the highly selective and sensitive detection of O2•-. By employing diphenylphosphinyl chloride as the reactive group, DFCD operates through a photoinduced electron transfer (PET) mechanism and exhibits significant fluorescence enhancement at 574 nm upon reaction with superoxide anion. The

Near-Infrared Chemiluminescence Probe for Superoxide Anion Imaging in Alzheimer's Disease.

Alzheimer's disease (AD) has a complex pathogenesis in which superoxide anion (O2•-), a key reactive oxygen species (ROS), plays a critical role. Monitoring O2•- is essential for understanding AD mechanisms and improving early diagnosis. Chemiluminescence imaging provides high contrast without external excitation, making it ideal for in vivo O2•- detection. However, few probes can specifically image O2•- in the AD brain. To address this, we designed a chemiluminescence probe, DIP-O, for in vivo O2•- imaging in AD. In vitro tests showed that DIP-O emits in the near-infrared region (708 nm) with

NIR-II Ratiometric Imaging of SOD Activity with a Superoxide-Responsive Nanoprobe.

Superoxide dismutase (SOD) maintains redox homeostasis, and its activity reflects oxidative stress in aging, inflammation, and diseases. However, real-time monitoring of SOD activity in vivo remains challenging. Here we constructed a superoxide anion radical (O2•-)-responsive NIR-II ratiometric probe based on a "substrate-monitoring and activity-back-calculation" strategy. The probe consists of an O2•--responsive unit (NKO, emission at 950 nm) and a stable internal reference unit (PbS quantum dots, emission at 1665 nm) conjugated via polyethylene glycol, affording robust aqueous dispersibility

Red-emitting chemosensor detects superoxide anion in models of myocardial hypertrophy.

Cardiac hypertrophy is a compensatory pathological alteration of the heart in response to prolonged stress, for which suitable diagnostic methods are lacking. Superoxide anion (O2˙-) levels are considered to be elevated during the progression of this disease. A red-emitting fluorescent chemosensor DO was rationally designed and synthesized. It enabled sensitive and accurate detection of O2˙- in solution, with a low limit of detection (LOD) of 93 nM. Chemosensor DO detected the increase in O2˙- concentration induced by angiotensin II (Ang II) in vitro and showed that losartan antagonizes Ang II

The Important Role of Superoxide Dismutase 2 in Controlling Poxvirus Proliferation and Pathogenicity.

Superoxide dismutase 2 (SOD2), a key mitochondrial antioxidant enzyme, is essential for maintaining cellular redox homeostasis by scavenging superoxide radicals. While viruses often induce oxidative stress, the specific role of SOD2 in antiviral defense remains unclear. Here, we report that vaccinia virus (VACV) infection triggers mitochondrial and cellular reactive oxygen species (ROS) and selectively upregulates SOD2, but not SOD1. Genetic knockout of SOD2 exacerbated mitochondrial ROS (mtROS) accumulation and significantly enhanced VACV replication and spread, resulting in larger viral plaq

Oxidative denaturation of Cu/Zn-superoxide dismutase associated with neurodegenerative diseases.

Misfolding of mutant Cu/Zn-superoxide dismutase (SOD1) is a well-established pathological feature of familial amyotrophic lateral sclerosis (ALS). While amino acid substitutions in mutant SOD1 destabilize its structure and promote misfolding, oxidation has also been implicated in the pathological alterations of wild-type SOD1, particularly in neurodegenerative diseases including sporadic ALS. However, the impact of oxidation on SOD1 folding remains to be fully elucidated. Here, we demonstrate that Cys111 is primarily oxidized to sulfonic acid upon exposure of apo-SOD1 to hydrogen peroxide, as

Targeting Superoxide to Prevent Diuretic Resistance and Reduce Heart Failure Mortality.

Diuretic resistance is a servomechanism involving superoxide that results in the need for escalating doses of diuretics in the treatment of heart failure. Without intervention, this typically is associated with increased heart failure mortality. Recent heart failure studies involving the use of SGLT2 inhibitors and GLP-1 agonists have shown reductions in the need for loop diuretics along with reductions in hospitalizations for heart failure and cardiovascular mortality. Multiple mechanisms have been suggested to explain the benefits of both these and related drugs, but this article is the firs

Environmental implications of superoxide radicals: From natural processes to engineering applications.

The roles of superoxide radical (O2•-) in the domains of physiological, physical, and material chemistry are becoming increasingly recognized. Although extensive efforts have been directed to understand O2•- functions in diverse aquatic systems, there is a lack of systematic and in-depth review for its kinetics and mechanisms in various environmental scenarios. This review aims to bridge this gap through discussion of O2•- generation pathways under both natural and controlled conditions. The merits and limitations of the generation and detection methods under various conditions are compared, w

The landscape of cognitive impairment in superoxide dismutase 1-amyotrophic lateral sclerosis.

Although mutations in the superoxide dismutase 1 gene account for only a minority of total amyotrophic lateral sclerosis cases, the discovery of this gene has been crucial for amyotrophic lateral sclerosis research. Since the identification of superoxide dismutase 1 in 1993, the field of amyotrophic lateral sclerosis genetics has considerably widened, improving our understanding of the diverse pathogenic basis of amyotrophic lateral sclerosis. In this review, we focus on cognitive impairment in superoxide dismutase 1-amyotrophic lateral sclerosis patients. Literature has mostly reported that c

Reactivity of trans-Resveratrol toward Electrogenerated Superoxide in N,N-Dimethylformamide.

The reactivity of 5-[(E)-2-(4-hydroxyphenyl)ethen-1-yl]benzene-1,3-diol (trans-resveratrol) and related compounds toward electrogenerated superoxide radical anion (O2•-) were investigated using electrochemistry, in situ electrolytic electron spin resonance, and in situ electrolytic ultraviolet-visible spectral measurements, in N,N-dimethylformamide (DMF) with the aid of density functional theory (DFT) calculations. The quasi-reversible cyclic voltammogram of dioxygen/O2•- was modified by the presence of trans-resveratrol, suggesting that the electrogenerated O2•- was scavenged by trans-resvera

Machine Learning Guided Discovery of Superoxide Dismutase Nanozymes for Androgenetic Alopecia.

Androgenetic alopecia (AGA) is a common form of hair loss, which is mainly caused by oxidative stress induced dysregulation of hair follicles (HF). Herein, a highly efficient manganese thiophosphite (MnPS3) based superoxide dismutase (SOD) mimic was discovered using machine learning (ML) tools. Remarkably, the IC50 of MnPS3 is 3.61 μg·mL-1, up to 12-fold lower than most reported SOD-like nanozymes. Moreover, a MnPS3 microneedle patch (MnMNP) was constructed to treat AGA that could diffuse into the deep skin where HFs exist and remove excess reactive oxygen species. Compared with the widely use

Inhibitory effects of superoxide dismutase 3 on IgE production in B cells.

Immunoglobulin E (IgE) functions as a first-line defense against parasitic infections. However, aberrant production of IgE is known to be associated with various life-threatening allergic diseases. Superoxide dismutase 3 (SOD3) has been found to suppress IgE in various allergic diseases such as allergic conjunctivitis, ovalbumin-induced allergic asthma, and dust mite-induced atopic dermatitis-like skin inflammation. However, the role of SOD3 in the regulation of IgE production in B cells remains elusive. In this study, we investigated the effect of SOD3 on LPS/IL-4 and anti-CD40/IL-4-mediated

Transcription factor-driven alternative localization of Cryptococcus neoformans superoxide dismutase.

Cryptococcus neoformans is an opportunistic fungal pathogen whose pathogenic lifestyle is linked to its ability to cope with fluctuating levels of copper (Cu), an essential metal involved in multiple virulence mechanisms, within distinct host niches. During lethal cryptococcal meningitis in the brain, C. neoformans senses a Cu-deficient environment and is highly dependent on its ability to scavenge trace levels of Cu from its host and adapt to Cu scarcity to successfully colonize this niche. In this study, we demonstrate for this critical adaptation, the Cu-sensing transcription factor Cuf1 di

Additive effects of metal excess and superoxide, a highly toxic mixture in bacteria.

Heavy metal contamination is a serious environmental problem. Understanding the toxicity mechanisms may allow to lower concentration of metals in the metal-based antimicrobial treatments of crops, and reduce metal content in soil and groundwater. Here, we investigate the interplay between metal efflux systems and the superoxide dismutase (SOD) in the purple bacterium Rubrivivax gelatinosus and other bacteria through analysis of the impact of metal accumulation. Exposure of the Cd2+ -efflux mutant ΔcadA to Cd2+ caused an increase in the amount and activity of the cytosolic Fe-Sod SodB, thereby

Superoxide dismutating molecules rescue the toxic effects of PINK1 and parkin loss.

Reactive oxygen species exert important functions in regulating several cellular signalling pathways. However, an excessive accumulation of reactive oxygen species can perturb the redox homeostasis leading to oxidative stress, a condition which has been associated to many neurodegenerative disorders. Accordingly, alterations in the redox state of cells and mitochondrial homeostasis are established hallmarks in both familial and sporadic Parkinson's disease cases. PINK1 and Parkin are two genes which account for a large fraction of autosomal recessive early-onset forms of Parkinson's disease an

Portacaval shunting causes differential mitochondrial superoxide production in brain regions.

The portacaval shunting (PCS) prevents portal hypertension and recurrent bleeding of esophageal varices. On the other hand, it can induce chronic hyperammonemia and is considered to be the best model of mild hepatic encephalopathy (HE). Pathogenic mechanisms of HE and dysfunction of the brain in hyperammonemia are not fully elucidated, but it was originally suggested that the pathogenetic defect causes destruction of antioxidant defense which leads to an increase in the production of reactive oxygen species (ROS) and the occurrence of oxidative stress. In order to gain insight into the pathoge

Inhibition of copper-zinc superoxide dismutase activity by selected environmental xenobiotics.

The function of Cu,Zn-SOD is to dismutate superoxide into hydrogen peroxide and oxygen. This task is fulfilled due to structural nuances of the enzyme. Many environmental xenobiotics have been proved to inhibit Cu,Zn-SOD. Those compounds could be found not only in industrial sewage, cigarettes and various chemical agents - some of them are used as drugs, drug production substrates or are the product of drug biotransformation. Cu,Zn-SOD exposition to these compounds leads to inhibition due to: copper ion chelation, unfolding the structure of the enzyme, affecting residues vital for activity mai
