Functional analyses of the interaction of chicken interleukin 23 subunit p19 with IL-12 subunit p40 to form the IL-23 complex.

This study represents the first description of the cloning of chicken IL-23p19 (ChIL-23α) and the function of the IL-23 complex in birds. Multiple alignment of ChIL-23α with other known IL-23α amino acid sequences revealed regions of amino acid conservation. The homologies of ChIL-23α, IL-12p35, and similar mammalian subunits ranged between 26% and 42%. ChIL-23α consisted of four exons and three introns; similar to those in humans and mice, and limited conservation of synteny between the human and chicken genomes was observed. Using bioinformatics tools, we identified the NF-κB, C/EBPα-β, c-Ju…

Draft whole genome sequence of the cyanide-degrading bacterium Pseudomonas pseudoalcaligenes CECT5344.

Pseudomonas pseudoalcaligenes CECT5344 is a Gram-negative bacterium able to tolerate cyanide and to use it as the sole nitrogen source. We report here the first draft of the whole genome sequence of a P. pseudoalcaligenes strain that assimilates cyanide. Three aspects are specially emphasized in this manuscript. First, some generalities of the genome are shown and discussed in the context of other Pseudomonadaceae genomes, including genome size, G + C content, core genome and singletons among other features. Second, the genome is analysed in the context of cyanide metabolism, describing genes

Sexual selection and maintenance of sex: evidence from comparisons of rates of genomic accumulation of mutations and divergence of sex-related genes in sexual and hermaphroditic species of Caenorhabditis.

Several hypotheses have been proposed to explain the persistence of dioecy despite the reproductive advantages conferred to hermaphrodites, including greater efficiency at purging deleterious mutations in the former. Dioecy can benefit from both mutation purging and accelerated evolution by bringing together beneficial mutations in the same individual via recombination and shuffling of genotypes. In addition, mathematical treatment has shown that sexual selection is also capable of mitigating the cost of maintaining separate sexes by increasing the overall fitness of sexual populations, and ge

Genome-Wide Characterization and Heat Responsive Expression Profiling of the HSP10 Co-Chaperonin Gene Family in Brassica juncea.

Heat stress disrupts protein homeostasis, yet the thermoregulatory roles of HSP10/CPN10 co-chaperonins in polyploid crops remain unexplored. This study conducted the first genome-wide characterization of the HSP10 gene family in allotetraploid Brassica juncea to identify heat tolerance candidates. Genome-wide identification of BjuHSP10 genes was performed across A and B subgenomes, followed by phylogenetic, gene structure, duplication, synteny, promoter, and structural analyses. Transcriptional responses were evaluated under moderate (28/24 °C) and severe (36/32 °C) heat stress conditions. Ten…

Genomic organization of the TCRα locus and essential roles of αβ T cells in antibacterial immunity in Nile tilapia.

T cells utilize diverse T cell receptors (TCRs) to recognize antigenic peptides and mediate adaptive immunity. However, the organization and function of αβ T cells in early vertebrates remain poorly understood. Here, we systematically characterized the TCRα locus and αβ T cell responses in Nile tilapia (Oreochromis niloticus) during Edwardsiella piscicida infection. Genomic analysis revealed that the TCRα locus in Nile tilapia exhibits a distinctive Vα(61)-Jα(82)-Cα(1)-Vα(65) structure with conserved synteny in the flanking regions, while the constant region retains essential residues for TCR/…

Phylogenomics Clarifies Plastome Reduction and Phylogenetic Relationships in Monotropoideae.

Plastome evolution and biogeographic history remain poorly understood in mycoheterotrophic lineages. Here, we sequenced and assembled the co … the more recently diverged Monotropa lineage. This reduction is accompanied by progressively poorer synteny and lineage-specific loss of rps23 in Monotropa. Phylogenomic analyses of 30 Ericaceae plastomes strongly supported Monotropoideae (bootstrap = 100) and clarified relationships among its 10 sampled genera. Genome-Wide Analysis of Fusarium oxysporum f. sp. cubense Peroxidases Reveals Oxidative-Stress Responses and Infection-Associated Expression.

Banana Fusarium wilt, caused by the soil-borne fungus Fusarium oxysporum f. sp. cubense (Foc), is a devastating disease and a major threat to global banana production. Here, we identified 24 genes predicted to encode heme-dependent peroxidase- or catalase-peroxidase-related proteins in Foc race 4 (Foc4), whereas nine genes encoding thiol-dependent peroxide-reducing proteins were catalogued separately. Phylogenetic, synteny, and Ka/Ks analyses indicated a conserved peroxidase repertoire under strong purifying selection, without substantial lineage-specific expansion. Motif, domain, gene-structu…
