Bone marrow-derived mesenchymal stem cells and the tumor microenvironment.

Over the last decade, there has been a growing interest in the role of mesenchymal stem cells (MSC) in cancer progression. These cells have the potential to give rise to a variety of mesenchymal cells like osteoblasts, chondrocytes, adipocytes, fibroblasts, and muscle cells. In contrast to their hematopoetic counterparts, MSC are not as clearly defined, which makes the interpretation of their role in cancer progression more complex. However, the nature of the relationship between MSC and tumor cells appears dual. Primary and metastatic tumors attract MSC in their microenvironment where they be

Lymphocyte-rich classical Hodgkin's lymphoma: distinctive tumor and microenvironment markers.

The existence, diagnostic features, and the biological and clinical relevance of lymphocyte-rich classical Hodgkin's lymphoma remain controversial. A comparative marker analysis of lymphocyte-rich classical Hodgkin's lymphoma, nodular lymphocyte-predominance Hodgkin's lymphoma, and of other subtypes of classical Hodgkin's lymphoma was carried out. Markers were selected focusing on B-cell lineage and transcription program (OCT.1, OCT.2, BOB.1, BCL6, PAX-5, GCET1, KLHL6, and BLIMP1), the NF-kappaB signaling pathway (REL-B, C-REL, TRAF-1, p-50, and MUM-1) and the T-cell microenvironment (CD3, CD5

The follicular lymphoma microenvironment: From tumor cell to host immunity.

Follicular lymphoma (FL) is a neoplasm derived from follicular germinal center cells. Like the normal components of this lymphoid structure, FL cells interact with various immune cells, such as the follicular helper T cells, suppressor regulatory T cells, dendritic cells, and histiocytes, that define the tumor microenvironment. Gene expression studies have shown that the nature of the tumor microenvironment predicts survival in patients with FL and may influence the response to immunotherapy and risk of transformation. The immune system may either promote or constrain tumor cell development, d…

Skeletal metastases: decreasing tumor burden by targeting the bone microenvironment.

Several common cancers often metastasize to the skeleton in advanced disease. Bone metastases are incurable and cause protracted, severe symptoms. Growth of tumor in bone is driven by a vicious cycle: tumor-secreted factors stimulate bone cells, which in turn release growth factors and cytokines. The bone-derived factors fuel the vicious cycle by acting back on the tumor cells. The vicious cycle offers novel targets for the treatment of advanced cancers. Treatments can inhibit bone cells (osteoclasts and osteoblasts) that are stimulated by tumor-secreted factors. Drugs can also inhibit tumor r

Spatial single-cell landscape of tumor-associated macrophages and their crosstalk with the tumor microenvironment.

Tumor-associated macrophages (TAMs) constitute a critical immune cell population within the tumor microenvironment (TME) and exhibit high functional heterogeneity. In this study, a pan-cancer atlas comprising 28 TAM subtypes was systematically constructed by integrating single-cell transcriptomic and spatial transcriptomic data from 291 human samples across 16 cancer types. The biological characteristics and spatial distribution patterns of these subtypes within the TME as well as the interaction mechanisms between TAMs and TME components closely associated with tumor progression were elucidat…

Tumor-Associated Neutrophils and Desmoplastic Reaction in the Breast Cancer Tumor Microenvironment: A Comprehensive Review.

The evolving tumor microenvironment (TME) plays a critical role in breast cancer tumorigenesis, growth, and metastatic potential. This study focuses on two key components of the TME: tumor-associated neutrophils (TANs) and the desmoplastic reaction (DR). We will analyze their multifaceted functions, emphasizing the significant mutual relationships among them, which dramatically affect disease outcomes and the effectiveness of treatments. TANs can either suppress or promote tumors, demonstrating notable functional flexibility in response to signals from their immediate environment. Concurrently…

Ectonucleotidases: Possible roles in the tumor microenvironment and influence on tumor progression in breast cancer.

Compared with nontumor tissue, the tumor microenvironment has a higher concentration of extracellular ATP. Extracellular ATP is degraded by the cooperative action of CD39 and ecto-5'-nucleotidase (CD73), thus leading to increases in the concentrations of adenosine and phosphate. This cooperative action can convert a proinflammatory environment characterized by a high concentration of ATP into an anti-inflammatory environment characterized by a high concentration of adenosine. In addition to its role in immune suppression, adenosine induces migration, metastasis and angiogenesis in breast cance…

Focusing on the interplay between tumor-associated macrophages and tumor microenvironment: from mechanism to intervention.

Immunotherapy has generated promising outcomes in cancer treatment; however, therapeutic responses are hampered by immunosuppression in the tumor microenvironment (TME). This has resulted in increased study of key immune cells in the TME as therapeutic interventions. Tumor-associated macrophages (TAMs), a major component of infiltrating immune cells in the TME, display high plasticity, largely dependent on cues received from their surroundings. Although significant progress in metabolomics and single-cell omics has unraveled the metabolic and functional heterogeneity of TAMs across several typ…

Targeted Nanophotoimmunotherapy Potentiates Cancer Treatment by Enhancing Tumor Immunogenicity and Improving the Immunosuppressive Tumor Microenvironment.

Cancer immunotherapy, such as immune checkpoint blockade, chimeric antigen receptor, and cytokine therapy, has emerged as a robust therapeutic strategy activating the host immune system to inhibit primary and metastatic lesions. However, low tumor immunogenicity (LTI) and immunosuppressive tumor microenvironment (ITM) severely compromise the killing effect of immune cells on tumor cells, which fail to evoke a strong and effective immune response. As an exogenous stimulation therapy, phototherapy can induce immunogenic cell death (ICD), enhancing the therapeutic effect of tumor immunotherapy. H…

Tumor microenvironment-responsive S-NSs-TPZ-ICG intelligent nanoplatforms for synergistically enhanced tumor multimodal therapy.

Nanosheet carriers loaded with drugs and phototherapeutics are used for effective cancer therapy, but the process remains challenging. Here, we prepared sulfur nanosheets (S-NSs) and then loaded tirapazamine (TPZ) and indocyanine green (ICG) with a loading efficiency of 6.3% and 94%, respectively. The obtained S-NSs-TPZ-ICG exhibits near-infrared (NIR) fluorescence, high 1O2 generation and photothermal conversion capabilities, good biocompatibility, and tumor microenvironment responsiveness. In vivo and in vitro experiments reveal that S-NSs-TPZ-ICG can be selectively decomposed under acidic a…

PRSS2 remodels the tumor microenvironment via repression of Tsp1 to stimulate tumor growth and progression.

The progression of cancer from localized to metastatic disease is the primary cause of morbidity and mortality. The interplay between the tumor and its microenvironment is the key driver in this process of tumor progression. In order for tumors to progress and metastasize they must reprogram the cells that make up the microenvironment to promote tumor growth and suppress endogenous defense systems, such as the immune and inflammatory response. We have previously demonstrated that stimulation of Tsp-1 in the tumor microenvironment (TME) potently inhibits tumor growth and progression. Here, we i…

Polyamine Depletion Strategies in Cancer: Remodeling the Tumor Immune Microenvironment to Enhance Anti-Tumor Responses.

Polyamine biosynthesis is frequently dysregulated in cancers, and enhanced flux increases intracellular polyamines necessary for promoting cell growth, proliferation, and function. Polyamine depletion strategies demonstrate efficacy in reducing tumor growth and increasing survival in animal models of cancer; however, mechanistically, the cell-intrinsic and cell-extrinsic alterations within the tumor microenvironment underlying positive treatment outcomes are not well understood. Recently, investigators have demonstrated that co-targeting polyamine biosynthesis and transport alters the immune l…

High-Dose Dexamethasone Manipulates the Tumor Microenvironment and Internal Metabolic Pathways in Anti-Tumor Progression.

High-dose dexamethasone (DEX) is used to treat chemotherapy-induced nausea and vomiting or to control immunotherapy-related autoimmune diseases in clinical practice. However, the underlying mechanisms of high-dose DEX in tumor progression remain unaddressed. Therefore, we explored the effects of high-dose DEX on tumor progression and the potential mechanisms of its anti-tumor function using immunohistochemistry, histological examination, real-time quantitative PCR (qPCR), and Western blotting. Tumor volume, blood vessel invasion, and levels of the cell proliferation markers Ki67 and c-Myc and

Macrophage- or neutrophil-expressed UPP1 drives tumor growth and immune suppression in the tumor microenvironment.

Uridine phosphorylase 1 (UPP1) has recently emerged as a metabolic regulator with critical roles in tumor cell biology. However, its expression pattern and functional significance in nonmalignant components of the tumor microenvironment (TME) remain largely unexplored. Here, we performed a comprehensive pan-cancer single-cell transcriptomic analysis revealing that UPP1 is highly expressed not only in tumor cells but also in tumor-associated macrophages (TAMs) and neutrophils (TANs). Clinically, UPP1 expression in TAMs and TANs correlates with poor prognosis and diminished response to immunothe…

Nanoplatforms synchronously activating tumor cell ferroptosis and remodeling the tumor microenvironment for reinforced cancer therapeutics.

Ferroptosis is a form of iron-dependent cell death pattern distinct from apoptosis, providing new targets for cancer therapeutics. By regulating cellular iron metabolism, destroying redox imbalance defenses and/or promoting lipid peroxidation, ferroptotic therapy induces ferroptosis of tumor cells, thereby circumventing resistance issues associated with the overwhelmingly used apoptosis-based treatments. Importantly, there are myriad interactions between ferroptotic tumor cells and the tumor microenvironment (TME), especially for solid tumors, which would greatly impact therapeutic outcomes. T…

A tumor-targeting gambogic acid-loaded nanovaccine reprograms the tumor microenvironment for enhanced cancer immunotherapy.

Nanovaccines for co-delivering antigens and adjuvants have demonstrated certain efficacy in cancer immunotherapy. However, these vaccines are challenged by poor biocompatibility, insufficient targeting ability, and attenuated performance in the immunosuppressive tumor microenvironment (TME). In this study, we successfully constructed a cancer cell membrane (CCM)-coated nanovaccine loaded with an active component from traditional Chinese medicine, gambogic acid (GA), using a magnesium/aluminum layered double hydroxide (LDH) as the functional carrier (CCM-LDH@GA). CCM serves as a source of tumor…
