Loss of myelinated axons is specific to the central nervous system in a mouse model of the fetal alcohol syndrome.

We have previously developed a mouse model of the fetal alcohol syndrome, the outcome of which is a late loss of myelinated axons in the optic nerve of offspring of alcohol-treated mice between 9 and 15 wk of age. We have extended this study to investigate whether this axon loss is stable and specific to the central nervous system. Pregnant female (C57BL/6/Wlds x CBA) F1 mice were injected intraperitoneally with a single dose of a 25% solution of ethanol (v:w), on d 12 of gestation. Control animals were given a similar volume of saline at the same time. Litters were taken at 12 (n = 18) and 23

Effects of prenatal exposure to ethanol on the number of axons in the pyramidal tract of the rat.

We examined the effect of gestational ethanol exposure on the number of axons in the caudal pyramidal tract. Between gestational day (G) 6 and G21, inclusive, pregnant rats were fed a liquid ethanol-containing diet (Et), an isocaloric liquid control diet (Ct), or a diet of chow and water (Ch). On postnatal day 30, the offspring of these rats were killed and their caudal medullas were processed for electron microscopy. The overall size of the pyramidal tract and the space occupied by the axons was smaller in the Et-treated rats than in the Ct-treated rats. The myelinated axons were smaller and

The effect of maternal ethanol intoxication on erythrocyte ghost fluidity in new-born rat pups.

We administered ethanol to pregnant rats and determined cholesterol, lipid phosphorus and fluorescence anisotropy of diphenylhexatriene (DPH) and of trimethylaminophenyl hexatriene (TMA-DPH) in erythrocyte ghosts of newborn pups and of their mothers. Cholesterol content was different in dams and pups and changed after treatment. Age, but not ethanol, affected lipid phosphorus. Either age (dams versus pups) or treatment affected the curves of DPH fluorescence anisotropy (r) versus temperature (T), but those of TMA-DPH did not change, indicating that only the inner core of the membrane was influ

Fetal alcohol exposure attenuates lipopolysaccharide-induced fever in rats.

Exposure to alcohol in utero can lead to long-lasting impairments of immune functions and to decreased resistance to infectious agents. We studied the effects of fetal alcohol exposure (FAE) in rats on the core body temperature response to an exogenous challenge of the immune system with lipopolysaccharide (LPS). We report that FAE rats show markedly decreased LPS-induced fever [i.e., they require a higher dose than control rats to show any LPS-induced hyperthermia (50 micrograms/kg vs. 10 micrograms/kg)], and even with the higher LPS dose they manifest a weaker hyperthermia, which declines fa

Protective effect of Liv.52 on alcohol-induced fetotoxicity.

The adverse effects of maternal alcohol consumption on the development of the fetus are well known. The adverse effects of ethanol on the liver are now believed to be due to acetaldehyde formed as an intermediate metabolite of ethanol. Liv.52 has been shown to bring about faster elimination of acetaldehyde from the body and thus prevent alcoholic liver damage. Other toxic effects of alcohol may also be due to acetaldehyde and may be prevented by Liv.52. In this study, rats were given 20% (v/v) ethanol in drinking water, during the gestation period, and the effect on maternal body weight and fe

Animal models of prenatal alcohol exposure.

Since the first reports of fetal alcohol syndrome (FAS), thousands of studies have examined the effects of antenatal alcohol exposure in humans and in animal models. Research with animal models has led to discoveries which would be difficult if not impossible in human subjects. Most importantly, these models have resulted in valuable insights into the actions of alcohol on various parts of the developing embryo and have helped researchers come closer to identifying the mechanisms of its teratogenic action. Both the direct and indirect biological effects of alcohol exposure in utero appear to w

In utero ethanol exposure decreases the biosynthesis of phosphatidylserine in rat pup cerebrum.

Phosphatidylserine is enriched in the brain and has been implicated to play a role in regulating neuronal membrane functions. In this study, three experimental protocols were used to examine the effects of in utero ethanol exposure on phosphatidylserine biosynthesis in rat pup brain, namely, (1) assay of the serine base-exchange enzyme activity in brain microsomes, (2) incubation of brain slices with [3H] serine, and (3) incorporation in vivo of [3H]serine into phosphatidylserine as well as serine-related phospholipids in brain. Results from all three protocols point to a decrease in phosphati

Effect of ethanol on the release of prostaglandins from ovine fetal brain stem during gestation.

Prostaglandins (PGs) have been implicated as mediators of the ethanol-induced suppression of ovine fetal breathing movements (FBM). The objectives of the present study were to determine the ontogeny of the in vitro efflux of PGE2 and 6-keto PGF1 alpha in ovine fetal brain stem during the second half of gestation and to determine the effect of in vitro ethanol exposure on the efflux of these PGs. Ovine fetal brain stem tissue was obtained at mean gestational ages of 80 days (n = 6), 105 days (n = 10), and 135 days (n = 16) by rapid excision following maternal euthanization. Tissue slices (400 m

Developmental thermoregulatory deficits in prenatal ethanol exposed long- and short-sleep mice.

Sensitivity to alcohol may influence the severity of prenatal alcohol effects. To examine this hypothesis, the ontogeny of thermoregulation was measured in prenatal ethanol exposed offspring of mice selected for differences in alcohol sensitivity. Pregnant long-sleep (LS) and short-sleep (SS) mice were exposed to 3 or 4 g/kg ethanol or an isocaloric amount of maltose-dextrin twice per day from day 7 through 18 of pregnancy. Doses were given six hours apart via gavage. Nonintubated lab chow controls were included for both genotypes. Offspring were fostered at birth to lactating mice of an outbr

Basic animal research.

Recent research with animal models of the fetal alcohol syndrome has resulted in advances in the areas of prenatal and postnatal growth retardation, facial dysmorphology, immune system deficiencies, endocrine dysfunction, and central nervous system alterations, including behavior. The rat is the most common species used in these studies, although similar experimental changes have been produced in a limited number of studies in other species. The specific deleterious effects of ethanol on the fetus are dose-related and dependent on the stage of gestation, as with other fetotoxic agents. However

Learning and memory in rats exposed pre- and postnatally to alcohol. An attempt at pharmacological control.

Using conditioned-reflex methods for active and passive avoidance with punishment reinforcement, we found pronounced memory deficits in 12-week old rats exposed perinatally to alcohol (FAS rats). Impairment of memory was observed not only with the high dose of 9 g ethanol/kg body weight (ingested with tap water in a 6% solution) to which dams were exposed during pregnancy and lactation, but also with the ten-fold lower dose of 1 g ethanol/kg body weight (0.6% ethanol). The nootropic drugs citicholine, piracetam and meclofenoxate administered orally for five days before the training session wer

Changes with age in the proliferative response of splenic T cells from rats exposed to ethanol in utero.

The fetal alcohol syndrome is associated with altered immunity. Several laboratories have confirmed that rodents exposed to ethanol in utero demonstrate both diminished proliferative responses of T cells to mitogens and diminished proliferative responses of T-blast cells to human recombinant interleukin 2 (rIL2). We examined the developmental time course of these altered immune responses by testing the immune function of in utero ethanol-exposed rats at various ages. We found that while diminished splenic T cell proliferative responses could not be detected at 2 weeks, they were present at 6 w

Suppression of immune responsiveness: sex differences in prenatal ethanol effects.

Exposure to ethanol in utero results in changes in the offspring's developing immune system, including thymus lymphocyte subpopulation shifts and functional lymphocyte changes that persist in adult animals. The present study was designed to define further the extent of changes in the immune system that result from fetal ethanol exposure and to compare effects in male and female offspring. In adulthood, male and female offspring from Sprague-Dawley dams fed an ethanol-containing liquid diet (alcohol, A), an isocaloric liquid control diet (pair-fed, PF), or laboratory chow and water (control, C)

Ethanol effect on the ionic transfer through isolated human amnion. I. Preventive and antagonistic actions of some nutrients and of their synthetic congeners.

The inhibition of the placental transfer of nutrients was studied in the pathogenesis of the fetal alcohol syndrome. Ethanol reduces the ionic transfer through the human amnion. In this study, the preventive and antagonistic actions of natural nutrients (Mg2+, Ca2+, taurine, homotaurine, gamma-aminobutyric acid) and of their synthetic congeners (Ca2+-taurinate, Ca2+-acetyltaurinate, Ca2+-acetylhomotaurinate) were observed on the amniotic conductance which was modified by ethanol. All the molecules, except Mg, demonstrated protective actions on the fetal side. The effect varied with the molecul

Growth response after administration of pharmacological doses of growth hormone in rats.

Increased levels of endogenous growth hormone (GH) during childhood and adolescence result in a significant elevation of final height both in humans and in laboratory animals. We investigated the growth promoting effects of the administration of pharmacological doses of human GH (25 to 100 IU/kg/day) in 7 rats with severe intrauterine growth retardation (IUGR) due to artificially induced embryofetal alcohol syndrome (EFAS). Such doses of GH led to a marked growth spurt during the time of administration (14 days) exceeding the expected daily length and weight gain significantly, compared to pla

Effects of physostigmine on shuttle avoidance in rats exposed prenatally to ethanol.

Shuttle avoidance performance following pretreatment with physostigmine was assessed in 85- to 100-day-old rats whose mothers consumed a liquid diet consisting of 35% ethanol-derived calories (EDC) during pregnancy. Offspring of pair-fed (0% EDC) and ad lib lab chow (LC) dams served as controls. Animals received either 0, 0.1 or 0.2 mg/kg physostigmine sulfate prior to acquisition training in a shuttle avoidance apparatus. Training consisted of 50 trials/day for 4 days. Thirty-five percent EDC rats made fewer avoidances than controls during acquisition training. Treatment with physostigmine re

Prenatal ethanol exposure affects temperature responses of adult rats to pentobarbital and diazepam alone and in combination with ethanol.

Long-term effects of prenatal alcohol exposure on body temperature responses to pentobarbital and diazepam and to either drug in combination with ethanol were studied in adult rats who were the offspring of dams fed a 5.0% w/v ethanol-containing liquid diet during the last 2 weeks of gestation. Adult offspring of pair-fed and chow-fed dams served as nutritional and normal controls, respectively. Pentobarbital (6.25-25.0 mg/kg) and diazepam (2.5-10.0 mg/kg) produced significantly greater dose-related hypothermic responses in females than males. Following either pentobarbital or diazepam adminis
