BACKGROUND & AIMS: The Dubin-Johnson syndrome is characterized by conjugated hyperbilirubinemia and by impaired secretion of anionic conjugates from hepatocytes into bile. Absence of the multidrug-resistance protein 2 (MRP2; symbol ABCC2), an adenosine triphosphate-dependent conjugate export pump, from the hepatocyte canalicular membrane is the molecular basis of this syndrome. The aim of this study was the elucidation of all exon-intron boundaries of the MRP2 gene as a prerequisite for the analysis of mutations in patients with Dubin-Johnson syndrome.
METHODS: Exon-intron boundaries of MRP2

The increasing prevalence of multidrug-resistant (MDR) Aeromonas salmonicida (AS) threatens both animal and human health by limiting the effectiveness of conventional antibiotics. To identify effective therapeutic strategies, we evaluated the activity of gentamicin (GENTA), levofloxacin (LEV), and cefotaxime sodium (CFT) as individual treatments and in combination using zebrafish and rainbow trout infection models. In vitro checkerboard assays showed consistent synergistic effects in the GENTA-CFT combination, as confirmed by Combenefit analysis. In vivo experiments revealed that AS infection

Tuberculosis (TB) therapy demands novel agents with distinct structures, targets, and mechanisms. Herein, through phenotypic screening, we identified a phenoxazinone scaffold questiomycin A (QA) with potent activity against Mycobacterium tuberculosis (MTB) H37Rv strain (MIC: 0.41 μg/mL) and broad-spectrum activity against drug-resistant clinical isolates (MIC: 0.16-0.31 μg/mL). Structural optimization of QA yielded lead B10 with enhanced activity against drug-resistance MTB (MIC: 0.063-0.25 μg/mL) and good intracellular antimycobacterial activity. In addition, B10 showed improved in vitro and

Since the introduction into clinical practice of vinca alkaloids, tubulin has become a key and well-established target of modern antineoplastic chemotherapy. When taxanes became available their broad spectrum of activity was striking and opened up new horizons for cancer patients' treatment. However, taxanes' susceptibility to drug resistance caused by the drug efflux pump protein, P-glycoprotein, is not infrequent and their use may be limited by poor solubility, synthetic problems and toxicity. The epothilones are a new class of chemotherapeutic agents with a mechanism of action similar to th

Staphylococcus aureus has been reported as one of the most difficult to treat. In the search for new treatment alternatives, isolated plant substances such as phenolic compounds, have demonstrated the ability to reverse bacterial resistance. The present study aims to evaluate the inhibitory action of caffeic acid and gallic acid on efflux pumps from S. aureus resistant strains. The broth microdilution assay was carried out to obtain the MICs of caffeic acid and gallic acid while the efflux pump inhibition test was assessed through the reduction of the minimum inhibitory concentration of the an

Diagnostic cytogenetic abnormalities are considered important prognostic factors in patients with acute myeloid leukaemia (AML). However, the prognostic assessments have mainly been derived from patients with AML aged <60 years. Two recent studies of AML patients of 60 years and older proposed prognostic classifications with distinct discrepancies. To further study the prognostic value of cytogenetic abnormalities in this patient population, we have evaluated cytogenetic abnormalities in a series of 293 untreated patients with AML aged 60 years and older, included in a randomised phase 3 trial

Biofilm formation by Pseudomonas aeruginosa is a considerable challenge in treating infections. We aimed to investigate the inhibitory effect of crocin, naringin, and naringenin on biofilm formation capacity and the expression of the lecA gene by this organism. One hundred unrepeated P. aeruginosa isolates were collected from hospitalized patients and were identified. The antibiotic resistance pattern of the isolates was determined using the disk agar diffusion method. The minimum inhibitory concentration (MIC) of naringin, naringenin, and crocin was determined by a micro broth dilution test.

Antimicrobial Resistance (AMR) represents a defining crisis of modern healthcare, severely limiting therapeutic options and driving a global increase in clinical mortality. Central to this crisis is Klebsiella pneumoniae, a ubiquitous gut commensal that has evolved into a formidable opportunistic pathogen through its remarkable ability to transition from a harmless organism to a hypervirulent, Multidrug-Resistant (MDR) threat. This review examines that pathogenic transition, emphasizing the dangerous convergence of virulence and resistance traits particularly within carbapenem-resistant lineag

BACKGROUND: The Bacillus Calmette-Guérin (BCG) vaccine is widely administered to prevent severe tuberculosis but can cause serious adverse events, including disseminated BCGosis, in immunocompromised individuals. However, studies investigating the in vivo genetic adaptation and microevolution of this live-attenuated vaccine during prolonged infection remain limited.
METHODS: Two clinical Mycobacterium bovis BCG isolates (BCG01 and BCG02) and a lot-matched vaccine strain (VAC) underwent whole-genome sequencing. Phenotypic drug susceptibility testing was performed on the clinical isolates. Geno

The clinical management of severe infections caused by Enterobacterales co-producing KPC and metallo-beta-lactamases is challenging. In these situations, the use of aztreonam-avibactam (AZA), or the association of aztreonam (ATM) with ceftazidime-avibactam (CZA), in scenarios where AZA is not available, has demonstrated clinical efficacy. However, the time to obtain results of in vitro antimicrobial susceptibility test may be a concern. We aimed to evaluate DOT-MGA (Direct-on-target microdroplet growth assay) as a tool for the rapid determination of susceptibility to AZA, as well as CZA plus A

Klebsiella pneumoniae are a leading cause of healthcare-associated infections worldwide. In particular, strains expressing extended-spectrum β-lactamases (ESBLs) and carbapenemases pose serious treatment challenges, leading the World Health Organization (WHO) to designate ESBL and carbapenem-resistant Enterobacteriaceae as 'critical' threats to human health. Research efforts to combat these pathogens can be supported by accessibility to diverse and clinically relevant isolates for testing novel therapeutics. Here, we describe a panel of 100 diverse K. pneumoniae isolates that are publicly avai

Globally, cervical cancer is the fourth most common cancer among women and the second most common cancer in India after breast cancer, with the highest burden occurring in low- and middle-income countries. Although platinum-based chemotherapy and concurrent chemoradiotherapy improve outcomes in early-stage disease, multidrug resistance (MDR) remains a major cause of treatment failure in persistent, recurrent, and metastatic cervical cancer. Olaparib, a poly(ADP-ribose) polymerase inhibitor (PARPi), has emerged as a promising targeted therapeutic agent; however, it is not currently approved as
