Lqh-8/6 is a minor fraction isolated from the venom of the scorpion Leiurus quinquestriatus hebraeus. Here we describe the purification, amino acid sequencing and solution structure determination by NMR and molecular modeling of this peptide. Lqh-8/6 is a small polypeptide (38 residues) which contains 8 half-cystines and is highly similar to another venom component, chlorotoxin. Standard homonuclear methods were used to sequentially assign the proton NMR spectra and to collect spatial restraints for structure determination. Two populations, identified early in the assignment step, are in slow

The timing and mechanism by which a host insect initiates an immune response are critical to successful defense against infection. Pathogen recognition, a prerequisite for host defense, has long been recognized to take place during the insect epidermis invasion by fungus. Here we report that insect can sense the fungal pathogen before host cuticle is penetrated by fungus. We discovered the upstream pattern recognition receptor (PRR) genes of the Toll pathway were upregulated in both the integument and fat body early during fungal germination on the epicuticle of Locusta migratoria manilensis.

Cystathionine β-synthase (CBS), the key enzyme in the transsulfuration pathway, links methionine metabolism to the biosynthesis of cellular redox controlling molecules. CBS catalyzes the pyridoxal-5'-phosphate-dependent condensation of serine and homocysteine to form cystathionine, which is subsequently converted into cysteine. Besides maintaining cellular sulfur amino acid homeostasis, CBS also catalyzes multiple hydrogen sulfide-generating reactions using cysteine and homocysteine as substrates. In mammals, CBS is activated by S-adenosylmethionine (AdoMet), where it can adopt two different c

BACKGROUND: Chagas disease is most often transmitted to humans by Trypanosoma cruzi infected triatomine bugs, and remains a significant cause of morbidity and mortality in Central and South America. Control of Chagas disease has relied mainly on vector eradication. However, development of insect resistance has prompted us to develop a paratransgenic strategy to control vectorial transmission of T. cruzi. Here, the potential role of recombinant endoglucanases as anti-trypanosomal agents for paratransgenic application is examined. The surface of T. cruzi is covered by a thick coat of mucin-like

Rift Valley fever virus (RVFV) is a Phlebovirus (Bunyaviridae family) transmitted by mosquitoes. It infects humans and ruminants, causing dramatic epidemics and epizootics in Africa, Yemen, and Saudi Arabia. While recent studies demonstrated the importance of the nonstructural protein NSs as a major component of virulence in vertebrates, little is known about infection of mosquito vectors. Here we studied RVFV infection in three different mosquito cell lines, Aag2 cells from Aedes aegypti and U4.4 and C6/36 cells from Aedes albopictus. In contrast with mammalian cells, where NSs forms nuclear

The determination of the redox properties of the cofactor in heme proteins provides fundamental insight into the chemical characteristics of this wide-spread class of metalloproteins. For the preparation of the ferroheme state, probably the most widely applied reductant is sodium dithionite, which at neutral pH has a reduction potential well below the reduction potential of most heme centers. In addition to the heme iron, some heme proteins, including the nitrophorins (NPs), contain cysteinecysteine disulfide bonds. In the present study, the effect of dithionite on the disulfides of NP4 and NP

Neuron-glia interactions are necessary for the formation of the longitudinal axon trajectories in the Drosophila central nervous system. Longitudinal glial cells are required for axon guidance and fasciculation, and pioneer neurons for trophic support of the glia. Neuregulin is a neuronal molecule that controls glial survival in the vertebrate nervous system. The Drosophila protein Vein has structural similarities with Neuregulin. We show here that Vein functions like a Neuregulin to maintain glial cell survival. We present direct in vivo evidence at single-cell resolution that Vein is produce

Here we review recent insights in the genetics of skin cancer susceptibility as gleaned from studies of three hereditary syndromes: basal cell nevus syndrome, familial melanoma/dysplastic nevus syndrome, and xeroderma pigmentosum. We provide a brief synopsis of the recent findings related to these syndromes in an attempt to illustrate several emerging themes in the genetics of skin cancer. These themes include 1) the recent identification of multiple cancer susceptibility genes that occur in a myriad of cellular regulatory pathways; 2) the relative specificity of certain regulatory pathways to

BACKGROUND: Sumoylation is a type of post-translational modification that is implicated in the regulation of numerous cellular events. However, its role in the function of human sperm has not yet been characterized.
METHODS AND RESULTS: In this study, both immunofluorescence and electron microscopy revealed that small ubiquitin-like modifiers (SUMO) SUMO1 and SUMO2/3 were highly enriched in the neck area of human sperm that is associated with the redundant nuclear envelope and were also detectable in the flagella and some head regions. Similar localization patterns of SUMO were also observed

Dominant Drop (Dr) mutations are nearly eyeless and have additional recessive phenotypes including lethality and patterning defects in eye and sensory bristles due to cis-regulatory lesions in the cell cycle regulator string (stg). Genetic analysis demonstrates that the dominant small eye phenotype is the result of separate gain-of-function mutations in the closely linked muscle segment homeobox (msh) gene, encoding a homeodomain transcription factor required for patterning of muscle and nervous system. Reversion of the Dr(Mio) allele was coincident with the generation of lethal loss-of-functi

Freshwater fish farming is one of the fastest-growing food sectors, contributing significantly to global food security. Despite the impressive global growth of aquaculture, high operational costs remain a major challenge, with feed production costs constituting the highest proportion of expenses. To reduce reliance on expensive materials, alternative ingredients have been explored, but issues such as reduced digestibility may arise. To address these challenges, various processing methods have been developed, ranging from traditional to advanced technologies. This review provides comprehensive

Diabrotica undecimpunctata ssp. howardi, the southern corn rootworm or eastern 12-spotted cucumber beetle, is a generalist insect herbivore that causes damage and yield loss to several crops in North America, including maize. Unresolved phylogenetic relationships within and among D. undecimpunctata subspecies are impacting current quarantine policies. We report the chromosome-level haploid genome assembly, icDiaUnde3, constructed using HiFi and Hi-C read data from a single male D. undecimpunctata collected and identified as subspecies howardi based on geographic location and morphology. The genus Laggera (Asteraceae) comprises medicinal plants traditionally used for treating inflammatory, infectious, and respiratory disorders. However, a comprehensive review of its phytochemistry and pharmacological potential is still limited. A systematic literature search was conducted across major scientific databases, including PubMed, Scopus, Web of Science, and Google Scholar, covering publications through April, 2026. Keywords such as "Laggera", "phytochemistry", "essential oil", "pharmacology", and "synthesis" were used. Relevant studies were selected based on their focus on chemical

We have previously shown that the slender form of Trypanosoma (T.) brucei is able to infect teneral tsetse flies, develop to the first fly form, which is the procyclic form, and complete the life cycle in the insect vector (Schuster et al., 2021). Further, analysis of the transmission index (TI; defined as the number of salivary gland infections relative to the number of midgut infections) revealed a higher TI for slender as compared to stumpy forms under laboratory conditions, which included the addition of N-acetylglucosamine (NAG) to the infective bloodmeal. Here, we show that slender trypa

In decapod crustaceans, red pigment-concentrating hormone (RPCH) is considered an ortholog of insect adipokinetic hormone (AKH); both belong to the conserved arthropod AKH/RPCH neuropeptide family. Research on crustacean RPCH has focused primarily on pigmentary functions, but the tissue-level roles of RPCH beyond its role in pigmentation are poorly understood. In this study, we used RNA interference to knock down the rpch gene in Litopenaeus vannamei (Lv-rpch) and integrated transcriptomic analysis, reverse transcription quantitative polymerase chain reaction (RT-qPCR) validation, biochemical

The A subunit of thyrotropin receptor (TSHR) is thought to be the crucial gene mediating stimulatory autoantibodies in Graves' diease (GD), but it remains unclear what the molecular basis of this pathological antibody response is. Stimulatory TSHR autoantibodies may induce activation of multiple signalling pathways in GD, modulate chemokine exposure and further stimulate immune imbalance. In this study, we prepared TSHR 289 protein by using insect baculovirus expression, adenovirus-expressed TSHR289 immunised mice, and obtained three mouse anti-TSHR monoclonal antibodies (mAbs), 1A4, 7C3 and 2

Chimeric peptides comprised of amino acid sequence motifs found within hormones, neuropeptides, and insect or lizard toxins are now under investigation for their potential use in therapeutics. Here, we report the discovery of one such peptide designated as Black Widow Spider-Exendin-4 (BW-Ex-4). It consists of a putative G protein-coupled receptor (GPCR) binding domain present within αLatrotoxin (αLTX) isolated from Latrodectus, and fused to N- and C- terminal motifs found within the glucagon-like peptide-1 receptor (GLP-1R) agonist Exendin-4 isolated from Heloderma. Vaccination is the most effective way to prevent infectious diseases and control outbreaks, and nanoparticles are highly suitable platforms for protein-based vaccines. Human adenovirus type 7 (Ad7) is one of the main pathogens causing severe pneumonia. A safe and effective vaccine against Ad7 infection is urgently needed. Penton-dodecahedron (Pt-Dd) nanoparticles, can be generated by the spontaneous assembly of the penton during the replication cycle of human adenovirus type 3. Adenovirus type 3 Pt-Dd has been studied as drug delivery vehicles. However, little attention has been paid to whethe

The poultry industry is a critical source of affordable protein worldwide; however, it faces continuous threats from various poultry diseases that significantly impact public health, economic stability, and food security. Knowledge of and examination of the transmission routes, risk factors, and environmental survival characteristics of the most important pathogens affecting poultry populations, as well as the importance of strict biosecurity, are pivotal. Transmission routes are split into direct and vector-borne pathways, and indirect ways, which include infections via contaminated surfaces

The tomato leafminer (Tuta absoluta) is a highly destructive global quarantine pest. The methoprene-tolerant (Met) protein, a member of the bHLH/PAS family of transcription factors, forms complexes with other family members to transduce the juvenile hormone signal, which regulates insect growth and development. However, the functions of the TaMet gene have rarely been studied in T. absoluta. Herein, we investigated the significance of TaMet in T. absoluta. Spatiotemporal expression analysis revealed that TaMet exhibited comparable expression patterns in males and females, with high expression
