Phytochemical Analysis and Trypanocidal Activity of Marrubium incanum Desr.

The rationale inspiring the discovery of lead compounds for the treatment of human parasitic protozoan diseases from natural sources is the well-established use of medicinal plants in various systems of traditional medicine. On this basis, we decided to select an overlooked medicinal plant growing in central Italy, Marrubium incanum Desr. (Lamiaceae), which has been used as a traditional remedy against protozoan diseases, and to investigate its potential against Human African trypanosomiasis (HAT). For this purpose, we assayed three extracts of different polarities obtained from the aerial par

Hepatic extracellular signal-regulated kinase 2 suppresses endoplasmic reticulum stress and protects from oxidative stress and endothelial dysfunction.

BACKGROUND: Insulin signaling comprises 2 major cascades: the insulin receptor substrate/phosphatidylinositol 3'-kinase/protein kinase B and Ras/Raf/mitogen-activated protein kinase/kinase/ERK pathways. While many studies on the tissue-specific effects of the insulin receptor substrate/phosphatidylinositol 3' -kinase/protein kinase B pathway have been conducted, the role of the other cascade in tissue-specific insulin resistance has not been investigated. High glucose/fatty acid toxicity, inflammation, and oxidative stress, all of which are associated with insulin resistance, can activate ERK.

Effects of rosiglitazone on abnormal lipid kinetics in HIV-associated dyslipidemic lipodystrophy: a stable isotope study.

HIV-associated dyslipemic lipodystrophy (HADL) is a heterogeneous syndrome of fat redistribution, hypertriglyceridemia, and insulin resistance, associated with markedly accelerated rates of lipolysis, intraadipocyte and intrahepatic reesterification, and very low-density lipoprotein-triglyceride synthesis and release. The objective of the study was to determine if rosiglitazone can ameliorate these lipid kinetic defects in patients with HADL. Infusions of [(13)C(1)]palmitate and [(2)H(5)]glycerol were used to measure total and net lipolysis, adipocyte and hepatic reesterification, and plasma f

Short-term milk production responses differ when balancing dietary 18-carbon fatty acids using high-oleic soybeans or a commercially available stearic acid-enriched supplement.

Recent evidence suggests that dietary 16- and 18-carbon fatty acids (FA) play an important role in influencing milk fat synthesis and nutrient partitioning. However, limited research has directly compared the effects of oleic- versus stearic-based supplemental sources under controlled conditions. We evaluated the effects of supplying 18-carbon FA primarily as oleic acid (cis-9 C18:1) from high-oleic soybeans versus stearic acid (C18:0) from a commercially available C18:0-enriched supplement, while balancing for similar 16-carbon FA intake, on short-term production responses in mid-lactation Ho

Palmitoylation of oncogenic NRAS is essential for leukemogenesis.

Activating mutations of NRAS are common in acute myeloid leukemia, chronic myelomonocytic leukemia, and myelodysplastic syndrome. Like all RAS proteins, NRAS must undergo a series of post-translational modifications for differential targeting to distinct membrane subdomains. Although farnesylation is the obligatory first step in post-translational modifications of RAS, to date, successes of therapies targeting farnesyl protein transferase are modest. Other RAS modifications, such as palmitoylation, are required for optimal plasma membrane association of RAS proteins. However, the relative impo

The SGLT2 Inhibitor Empagliflozin Directly Binds Cardiac JNK3, Uncouples c-Jun/c-Fos Signaling, and Confers Cardioprotection in Diabetic Cardiomyopathy.

BACKGROUND: Sodium-glucose cotransporter 2 (SGLT2) inhibitors, including empagliflozin (EMPA), confer significant cardioprotection in type 2 diabetes in addition to lowering glucose levels. However, their direct targets in cardiac tissue and molecular mechanisms underlying their effects in diabetic cardiomyopathy (DCM) remain unclear. The aim of the present study was to identify a direct cardiac target of EMPA and define the downstream signaling mechanisms mediating its cardioprotective effects in DCM. METHODS: A kinase activity screening of 107 kinases was used to identify candidate targets

Liquiritigenin ameliorates adipocyte insulin resistance by inhibiting pro-inflammatory polarization of macrophages via regulation of the mtROS/OXPHOS axis.

Obesity-induced insulin resistance (IR) is closely associated with chronic inflammation and metabolic dysfunction in adipose tissue, in which macrophage polarization plays a pivotal role. Liquiritigenin (LQ), a natural flavonoid derived from licorice, exhibits potential metabolic regulatory effects; however, its role in high-fat diet (HFD)-induced obesity and insulin resistance, as well as the underlying mechanisms, remains unclear. In this study, an HFD-induced obese mouse model combined with multi-omics analyses and an in vitro co-culture system was employed to investigate the effects of LQ.

Tirzepatide attenuates cardiac microvascular injury and ferroptosis-related lipid peroxidation in diabetic mice: endothelial responses in the context of partial PRKN silencing.

BACKGROUND: Diabetic metabolic stress damages the cardiac microvasculature. We evaluated tirzepatide-associated cardiac and endothelial effects, including endpoint patterns after partial silencing of parkin RBR E3 ubiquitin protein ligase (PRKN) and lipidated microtubule-associated protein 1 light chain 3 beta (LC3B-II) accumulation during lysosomal blockade. METHODS: Male C57BL/6J mice received a normal diet, a high-fat diet (HFD), or HFD/streptozotocin (STZ); HFD and HFD/STZ cohorts received tirzepatide or vehicle for 8 weeks. Metabolic, cardiac, microvascular, lipid-peroxidation, iron, and

Phase-dependent disruption of microbiome-metabolome coordination is associated with diet-induced MASLD.

Metabolic-dysfunction-associated steatotic liver disease (MASLD) is typically attributed to caloric overload, lipotoxicity, and static gut dysbiosis, but how chronic diet alters the temporal organization of gut-liver communication remains unclear. We combined a phase-stratified multiomics framework, cecal 16S rRNA profiling, dual-compartment (cecum and serum) metabolomics, and hepatic clock and lipogenic gene expression in C57BL/6J male mice fed a high-fat, palmitate, and cholesterol-enriched (FPC) diet containing high sucrose for 22 wk. FPC feeding was associated with severe MASLD and markedl

Hepatocyte NAT10-mediated ac4C regulation contributes to TGFβ1-associated Kupffer cell -HSC communication in steatotic liver disease.

BACKGROUND AND OBJECTIVE: Obesity and metabolic syndrome impose chronic lipotoxic stress on hepatocytes, contributing to macrophage reprogramming and fibrosis progression in steatotic liver disease. However, how soluble hepatocyte-derived factors regulate Kupffer cell-hepatic stellate cell (HSC) crosstalk remains unclear. This study investigated the role of hepatocyte NAT10-mediated N4-acetylcytidine (ac4C) RNA modification and explored potential ac4C-dependent targets involved in this process. METHODS: Hepa1-6 cells were exposed to palmitic acid with or without NAT10 knockdown, and nd key fi

Dietary addition of magnesium hydride nanoparticles: a breakthrough in combating high-fat diet-induced chronic kidney disease.

A substantial body of evidence indicates a positive correlation between dyslipidemia and an elevated risk of chronic kidney disease, with renal interstitial fibrosis frequently serving as a common pathway in the advanced stages of chronic kidney disease progression. Hydrogen has anti-inflammatory and antioxidant properties, and magnesium hydride nanoparticle is a material with high hydrogen storage capacity. Magnesium hydride -fortified feed is capable of releasing hydrogen gas steadily and continuously within the digestive tract. A 12-week high-fat diet significantly elevated the serum urea a

Exogenous hydrogen sulfide improves non-alcoholic fatty liver disease by inhibiting endoplasmic reticulum stress/NLRP3 inflammasome pathway.

Non-alcoholic fatty liver disease (NAFLD) is a common chronic liver disease worldwide, and its exact pathogenesis has not been fully studied. Hydrogen sulfide (H2S) is the third gas signaling molecule discovered in mammals, following nitric oxide and carbon monoxide. It has the effects of anti-inflammation, anti-apoptosis, and so on, thereby playing an important role in many diseases. However, the role and mechanism of exogenous H2S in NAFLD are not fully understood. In this study, we constructed in vitro and in vivo NAFLD models by feeding mice a high-fat diet and stimulating hepatocytes with

Transcriptomic and Proteomics Analysis of a Lipid-Loaded HepaRG Model for Steatosis Reveals Altered Regulation in Lipid and Xenobiotic Metabolism.

INTRODUCTION: Hepatic lipid accumulation (steatosis) is an early indicator of non-alcoholic fatty liver disease (NAFLD), preceding fibrosis and cirrhosis. Understanding its effects on drug-metabolizing enzymes (DMEs) and transporters is crucial for assessing potential alterations in drug disposition among NAFLD patients. This study aimed to replicate steatosis in an in vitro HepaRG cell model and analyze its impact on DMEs and transporters. METHODS: Differentiated HepaRG cells were treated with a mixture of saturated (palmitate) and unsaturated (oleate) fatty acids (in a 1:2 ratio at 0.5 mM),

Targeting Ferroptosis Restores the Antiviral Activity of CD8+ T Cells During Chronic Hepatitis B Virus Infection.

BACKGROUND & AIMS: CD8+ T cells play a crucial role in antiviral immunity; however, hepatitis B virus (HBV)-specific CD8+ T cells become dysfunctional during chronic HBV (CHB) infection. Blocking inhibitory pathways only partially restores efficient antiviral responses, suggesting that the mechanism underlying CD8+ T-cell dysfunction is complicated. This study aimed to investigate whether HBV-specific CD8+ T cells undergo ferroptosis, examine its correlation with T-cell dysfunction, and elucidate the underlying mechanism and potential intervention strategies. METHODS: Analysis of CD8+ T cells

Peripheral amyloid-β clearance mediates cognitive impairment in non-alcoholic fatty liver disease.

BACKGROUND: Non-alcoholic fatty liver disease (NAFLD) is a prevalent risk factor for cognitive impairment. Cerebral amyloid-β (Aβ) accumulation, as an important pathology of cognitive impairment, can be caused by impaired Aβ clearance in the periphery. The liver is the primary organ for peripheral Aβ clearance, but the role of peripheral Aβ clearance in NAFLD-induced cognitive impairment remains unclear. METHODS: We examined correlations between NAFLD severity, Aβ accumulation, and cognitive performance in female Sprague-Dawley rats. The impact of NAFLD on hepatic Aβ clearance and the involve

Resveratrol protects against a high-fat diet-induced neuroinflammation by suppressing mitochondrial fission via targeting SIRT1/PGC-1α.

Various health issues have emerged due to consuming high-fat diets (HFD), particularly the detrimental impact they have on mitochondrial dynamics and subsequet cognition functions. Specially, mitochondrial fission can serve as an upstream signal in the regulation of cortical inflammation and neural pyroptosis. Our study was designed to verify the existence of neuroinflammation in the pathogenesis of HFD-induced cognitive dysfunction and demonstrated that resveratrol (RSV) attenuated neural deficits via regulation of cortical mitochondrial fission. A total of 50 male Sprague Dawley rats were ra
