Sex-Specific Patterns of Taste Dysfunction, Their Relationships with α-Synuclein Profiling, and Supervised Learning-Based Diagnosis in Parkinson's Disease (PD).

Taste impairment is a little-known non-motor Parkinson's disease (PD) feature with potential diagnostic value. However, its biological basis and sex-specific patterns remain unclear. We combined psychophysical taste testing, salivary α synuclein (αsyn) profiling, genotyping of four SNCA polymorphisms, and Supervised Learning (SL) within a unified, sex-aware analytical framework to analyze sensory, molecular, and genetic correlates of gustatory dysfunction in 99 PD patients and 60 healthy controls. Overall taste identification was markedly reduced in PD, independently of sex. However, males and

Aberrant Cerebellar-Recipient Thalamic Activity in Two Mouse Models with Prominent Tremor or Bradykinesia.

The motor thalamus functions as a gateway between subcortical and cortical motor circuits, relaying information critical to movement initiation and execution. Inputs to motor thalamus from the basal ganglia and cerebellum form functionally distinct circuits responsible for different aspects of healthy and dysfunctional motor control. Here, we use mouse models that capture different features of Parkinson's disease pathophysiology-alpha-synuclein (α-syn) aggregation and dopamine loss-to study how thalamic subcircuits are altered under different pathophysiological conditions. Using a trans-synapt…

N-terminal acetylation reduces synuclein pathology in models of Parkinson's disease.

α-synuclein protein is a major constituent of pathological intracellular inclusions such as Lewy bodies found in brains of patients with Parkinson's disease and other synucleinopathies. Whereas α-synuclein phosphorylation has been much studied, comparatively less work has been devoted to other post-translational modifications such as acetylation, especially given that N-terminally acetylated α-synuclein is the most abundant endogenous form of the protein in the brain. In this study, using multiple in vitro and in vivo models of Parkinson's disease, we sought to understand the pathological cons

TogoPhosTAC as a delivery-ready platform for targeted protein dephosphorylation.

Phosphorylation-targeting chimeras (PhosTACs) enable targeted protein dephosphorylation by recruiting phosphatases through induced proximity. However, the direct recruitment of phosphatase subunits or holoenzymes with small molecules remains challenging, as suitable ligands are scarce and often compromise enzymatic activity or cellular function. Here, we present togoPhosTAC, a hybrid modality that integrates a small-molecule PhosTAC, an engineered FKBP12F36V-phosphatase, and a lipid nanoparticle delivery system. This strategy allows delivery of preassembled PhosTAC-FKBP12F36V-phosphatase compl

Deletion or immunotherapeutic blockade of FcγRIIb (CD32b) impairs α-Syn propagation in vivo.

Parkinson's disease (PD), the most prevalent neurodegenerative movement disorder, is characterised by widespread alpha-synuclein (α-Syn) pathology in the brain and the progressive degeneration of nigrostriatal dopaminergic neurons. Experimental and clinicopathological data support a templated, prion-like component to α-Syn spread; however, the cellular mechanisms governing fibril uptake and intercellular transmission remain incompletely defined. Fc gamma receptor IIb (FcγRIIb or CD32b), the sole cross-species inhibitory Fcγ receptor, has been implicated in α-Syn internalisation in vitro, thoug…

MPP+ induces α-synuclein oligomerization and neurite impairment by disrupting mitochondrial function and Akt signaling, with apigenin emerging as a potential neuroprotective agent.

Parkinson's disease (PD) is a devastating neurodegenerative disorder characterized by α-synuclein accumulation and mitochondrial impairment. Mitigating α-synuclein aggregation and mitochondrial dysfunction are prominent strategies in PD treatment. Apigenin, a dietary flavonoid, exhibits anti-inflammatory neuroprotective potential in PD models. However, it is still unclear whether apigenin possesses any impact on α-synuclein aggregation and mitochondrial dysfunction. Here, the effects and underlying mechanism of apigenin were investigated using MPP+-induced PD-like pathology in SH-SY5Y cells. O

Altered ATP13A2/PARK9 Levels Influence α-Synuclein Accumulation in Neurons via Phagocytosis and Secretion in Glial Cells.

(1) Background: Parkinson's disease (PD) is characterized by the pathological accumulation of α-synuclein (α-syn) containing Lewy bodies (LBs) and Lewy neurites (LNs) within neurons. Growing evidence indicates that α-syn may propagate throughout the nervous system in a manner similar to prion-like transmission. Extracellular vesicles (EVs) may contribute to this pathway. We and others have reported that ATP13A2/PARK9 deficiency results in decreased EVs while its overexpression leads to increased EV generation. For analyzing EV-mediated α-syn secretion in neighboring neurons, we planned to alte

A non-redundant role of EAAT3 for ATP synthesis mediated by GDH in dopaminergic neuronal cells: a new avenue for glutamate metabolism and protection in Parkinson's disease.

Parkinson's disease (PD) is a devastating neurodegenerative disorder with a distinct loss of the nigrostriatal dopaminergic pathway. Despite the multiplicity in etiology, alterations that disrupt neuronal integrity can be traced back to defects in fundamental processes that typically run under mitochondrial inputs. Evidence indicates that mitochondrial activities are hierarchically integrated with the energetic performance of these organelles, so that an interesting perspective holds that interventions aimed at improving mitochondrial bioenergetics can potentially mitigate the severity of PD p

Plasma extracellular vesicle pathognomonic proteins as the biomarkers of the progression of Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder for which reliable blood biomarkers to predict disease progression remain elusive. Plasma extracellular vesicles (EVs) have gained attention as a promising biomarker platform due to their stability and ability to cross the blood-brain barrier. This study explored the potential of EV-cargo proteins, specifically α-synuclein, tau, and β-amyloid, as biomarkers of PD progression. A cohort of 55 people with PD (PwP) and 58 healthy controls (HCs) underwent annual assessments of plasma EV proteins, cognition, and motor symptoms. EVs

α-synuclein fibrils per se but not α-synuclein seeded aggregation causes mitochondrial dysfunction and cell death in human neurons.

One of the major histopathological features of Parkinsons's disease - intracellular Lewy bodies - consists of misfolded α-synuclein. This protein can self-assemble, spread through the brain and seed its own aggregation. Aggregated α-synuclein is shown to induce mitochondrial dysfunction that leads to neuronal loss. Using human iPSC-derived SNCA triplication (3xSNCA) and isogenic control (ISO) neurons we studied whether acute exposure to fibrillar α-synuclein, or its seeding properties, induce effects on mitochondrial function and toxicity. Chronic exposure of neurons to fibrillar α-synuclein (

Early α-Synuclein Pathology Induces Neuroinflammation and Decreases Topoisomerase IIβ Expression in A53T Mice.

Neuroinflammation in the pathogenesis of neurodegenerative diseases is still a subject of discussion, as it might be either the cause or the result of neurodegeneration. Misfolded or improperly aggregated α-synuclein in Parkinson's Disease, can cause inflammatory responses, indicating a mechanistic relationship between α-synuclein pathology and neuroinflammation. The aim of this study was to investigate the role of DNA topoisomerase IIβ, the key player of DNA topology, in the neuroinflammation process in an age-dependent transgenic Parkinson's Disease mouse model overexpressing the A53T mutant

Early α-synuclein aggregation decreases corticostriatal glutamate drive and synapse density.

Neuronal inclusions of α-synuclein (α-syn) are pathological hallmarks of Parkinson's disease (PD) and Dementia with Lewy Bodies (DLB). α-Syn pathology accumulates in cortical neurons which project to the striatum. To understand how α-syn pathology affects cortico-striatal synapses at early time points before significant dopamine neuron loss, pre-formed α-syn fibrils (PFF) were injected into the striatum to induce endogenous α-syn aggregation in corticostriatal-projecting neurons. Electrophysiological recordings of striatal spiny projection neurons (SPNs) from acute slices found a significant d

Pathological α-synuclein perturbs nuclear integrity.

Pathological aggregates of α-synuclein are a hallmark of a group of neurodegenerative disorders collectively termed synucleinopathies. The physiological function of α-synuclein, and the detrimental effects of the pathological variants of α-synuclein have been widely debated, but recent evidence has suggested an emerging consensus on a critical role for α-synuclein in regulating synaptic function. However, a controversial role for α-synuclein in nuclear function in both normal and pathogenic states has been proposed, and the degree to which α-synuclein localizes within the nucleus and subsequen

Effects on neuroprotection and α-synuclein expression of a Canna coccinea rhizome extract.

Parkinson's disease (PD) is the second most common neurodegenerative disorder in the elderly, currently with no cure. Its mechanisms are not well understood, however α-synuclein protein aggregation plays a central role in the pathogenesis of PD, leading to neurodegeneration. We demonstrated that in a PD model dietary in Caenorhabditis elegans treatment with an extract from the rhizome of Canna coccinea decreased the accumulation of α-synuclein. The extract showed low toxicity to neuro 2a cells and protected them from oxidative damage with H2O2 as a model for neurodegeneration. We studied the c

Molecular dynamics simulation of the aggregation and folding mechanism of α-synuclein 47-56 in core peptide fragments induced by α-synuclein pentamer template.

The misfolding of intrinsically disordered α-synuclein protein, which can form β-sheet-rich fibrillar amyloid structures, is closely associated with Parkinson's disease (PD). The peptide α-synuclein 47-56 has been identified as the toxic core of α-synuclein and plays a pivotal role in the aggregation and misfolding processes of this protein. Investigating the template induction behavior of this peptide is crucial for elucidating the molecular mechanisms underlying α-synuclein misfolding and aggregation. To explore the molecular mechanism of the peptide α-synuclein 47-56, guided by the α-synucl

Altered reactivity to threatening stimuli in Drosophila models of Parkinson's disease, revealed by a trial-based assay.

The fruit fly Drosophila melanogaster emerges as an affordable, genetically tractable model of behavior and brain diseases. However, despite the surprising level of evolutionary conservation from flies to humans, significant genetic, circuit-level, and behavioral differences hinder the interpretability of fruit fly models for human disease. Therefore, to allow a more direct fly-versus-human comparison, we surveyed the rarely exploited, rich behavioral repertoire of fruit flies with genetic alterations relevant to Parkinson's disease (PD), including overexpression of human mutant Parkin or α-sy

Intact microdissection of stellate ganglia in a Parkinson's disease model reveals aggregation of mutant human α-synuclein in their cell bodies.

Cardiac dysautonomia plays an important role in understanding Parkinson's disease (PD), with recent studies highlighting the presence of α-synuclein in cardiac tissue. We hypothesise that sympathetic dysregulation observed in PD may involve pathological changes caused by α-synuclein in stellate ganglia (SG). This study aimed to investigate α-synucleinopathy in SG of the genetic PD murine animal model. Mice overexpressing Ala30Pro (A30P) mutant α-synuclein were used. We here demonstrate a technique for meticulously dissecting SG. The collected SG from the transgenic mice were immunolabelled wit

Alzheimer's disease and its co-pathologies: Implications for hippocampal degeneration, cognitive decline, and the role of APOE ε4.

INTRODUCTION: In neurodegenerative dementias, the co-occurrence and interaction of amyloid β peptide (Aβ), tau pathology, and other pathological lesions confound their individual contributions to neurodegeneration and their modulation by risk factors. METHODS: We analyzed 480 post mortem human brains (ages 50-99) using regression and structural equation models to assess the relationships among Aβ, tau, limbic-predominant age-related TDP-43 encephalopathy neuropathological changes (LATE-NC), α-synuclein, other age-related lesions, and apolipoprotein E (APOE) ε4, as well as their effects on CA1

α-Synuclein interacts directly with AP2 and regulates its binding to synaptic membranes.

α-Synuclein mutation and aggregation are associated with several neurodegenerative disorders, including Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy. It is expressed in the presynaptic compartment where it regulates clathrin mediated synaptic vesicle endocytosis. We have shown that α-synuclein regulates clathrin lattice size and curvature in vitro. However, the molecular mechanism by which this occurs remains unknown. Here, we show a strong colocalization between the heterotetrametric clathrin adaptor protein-2 (AP2) and α-synuclein at presynapses. Moreover, we r

Neuronally Derived Extracellular Vesicles' Oligomeric and p129-α-Synuclein Levels for Differentiation of Parkinson's Disease from Essential Tremor.

Clinical differentiation between Parkinson's disease (PD) and essential tremor (ET) may be challenging, highlighting the need for easily assessable diagnostic biomarkers. Neuronally derived extracellular vesicles (NDEVs) have been proposed as a peripheral matrix that can well recapitulate the cellular composition of neurons. We investigated the clinical usefulness of NDEV oligomeric and p129-α-synuclein levels in discriminating between patients with PD and those with ET. NDEV oligomeric and p129-α-synuclein species were assessed using an ELISA in 43 patients with PD, 21 patients with ET, and 4
