The synthesis of myelin and brain subcellular membrane proteins in the offspring of rats fed ethanol during pregnancy.

Pregnant Long-Evans rats received either: (1) liquid diet containing 5.15% ethanol; (2) liquid diet pair fed to (1) for total calories; or (3) liquid diet ad libitum. These special diets were administered from the 5th through the 18th days of gestation. Dams received standard laboratory chow and water ad libitum before and after the test interval. Additional dams received standard chow and water throughout the study. Birth weights of offspring in the ethanol group were lower than for offspring of the pair-fed or control groups, and their subsequent growth lagged behind the other groups. Neonat

Impaired brain growth in neonatal rats exposed to ethanol.

Infant rat pups, fed through intragastric cannulas from postnatal day 4 through day 18, showed a 19 percent reduction in total brain weight when ethanol was included in their diet on days 4 through 7. This reduction in brain weight occurred even though body growth in the experimental rats was equal to that of their littermate controls. The ethanol-exposed animals were markedly hypoactive during the period of drug administration, then displayed gross body tremors for 3 to 5 days. Throughout the study, the animals treated with ethanol had poor motor coordination and were hyperresponsive. These b

Liver abnormalities in three patients with fetal alcohol syndrome.

Liver abnormalities were found in three patients with fetal alcohol syndrome. The histological appearance was different in each case. Thick, sclerotic central veins were seen in two of the three cases. One patient had features typical of congenital hepatic fibrosis and cystic disease of the kidneys. Findings in these patients indicate that some cases of congenital hepatic fibrosis might be caused by high maternal alcohol ingestion in pregnancy.

Stem cell therapy: social recognition recovery in a FASD model.

To better understand the cellular pathogenetic mechanisms of fetal alcohol spectrum disorder (FASD) and the therapeutic benefit of stem cell treatment, we exposed pregnant rats to ethanol followed by intravenous administration of neural stem cells (NSCs) complexed with atelocollagen to the new born rats and studied recovery of GABAergic interneuron numbers and synaptic protein density in the anterior cingulate cortex, hippocampus and amygdala. Prenatal ethanol exposure reduced both parvalbumin-positive phenotype of GABAergic interneurons and postsynaptic density protein 95 levels in these area

Prenatal Alcohol Exposure Results in Sex-Specific Alterations in Circular RNA Expression in the Developing Mouse Brain.

Fetal alcohol spectrum disorders (FASD) are heterogeneous disorders associated with alcohol exposure to the developing fetus that are characterized by a range of adverse neurodevelopmental deficits. Despite the numerous genomics and genetic studies on FASD models, the comprehensive molecular understanding of the mechanisms that underlie FASD-related neurodevelopmental deficits remains elusive. Circular RNAs (circRNAs) are a subtype of long non-coding RNAs that are derived from back-splicing and covalent joining of exons and/or introns of protein-coding genes. Recent studies have shown that cir…

Do Canadian prenatal record forms integrate evidence-based guidelines for the diagnosis of a FASD?

OBJECTIVES: Prenatal alcohol exposure is a significant public health issue with lifelong psychological, emotional and financial costs associated with caring for an affected individual. In 2005, the Public Health Agency of Canada and Health Canada's First Nations and Inuit Health Branch developed evidence-based guidelines for the diagnosis of a Fetal Alcohol Spectrum Disorder (FASD). We examined the extent to which prenatal records across Canadian provinces and territories currently integrate key recommendations from these guidelines. METHODS: A content analysis of prenatal record forms retrie

Feasibility and potential benefits of an attention and executive function intervention on metacognition in a mixed pediatric sample.

The term "metacognition" describes thinking about a cognitive phenomenon or, more simply put, thinking about thinking . Metacognition involves using knowledge about one's cognitive processes to change behavior, including monitoring and controlling cognition. Metacognition is vital for learning and is often more difficult for children with neurodevelopmental concerns (e.g. Attention Deficit Hyperactivity Disorder [ADHD], Fetal Alcohol Spectrum Disorder [FASD], Autism Spectrum Disorders [ASD]), possibly due to underlying deficits in attention and executive functioning (EF). The present study eva

Student experiences in a novel interprofessional neurodevelopmental clinic: a qualitative study.

BACKGROUND: Student-led clinics can provide low-cost speciality care and practical interprofessional education (IPE) opportunities. In Australia, there are currently limited speciality services available that provide neurodevelopmental assessments that consider fetal alcohol spectrum disorder (FASD) as one possible outcome. The aim of the current study was to understand student experiences in a novel interprofessional student-led clinic for children and adolescents with suspected or confirmed prenatal alcohol exposure. METHOD: Seventeen allied health university students (11 occupational thera

Prenatal Alcohol Exposure Inhibits Transient Expression of Autophagy and Synaptic Proteins in Developing Brain.

INTRODUCTION: Neuronal apoptosis and consequent inhibition of autophagy, with loss of synaptic connections are central events in the genesis of fetal alcohol spectrum disorders (FASD). However, studies of molecular mechanisms of autophagy in human fetal brain are limited. Recently, prenatal exposure to EtOH was associated with reduced miRNA-9 levels in fetal brain-derived exosomes (FB- Es) isolated from maternal plasma, which correlated with small eyes, an anatomical hallmark of fetal alcohol syndrome (FAS). Since miR-9 targets several genes that regulate synaptic plasticity, EtOH-induced inhi…

Optic nerve hypoplasia: comparative effects in children and rats exposed to alcohol during pregnancy.

Children with the fetal alcohol syndrome often have ocular anomalies. These include abnormalities of the eyes and adnexa (strabismus, blepharoptosis, epicanthus), as well as intraocular defects (cataract, glaucoma, persistent hyperplastic primary vitreous, retinal and optic nerve anomalies). Based on the clinical results in an ophthalmological study of a group of Swedish children with the fetal alcohol syndrome, in which optic nerve hypoplasia was found in up to one-half of the group, an experimental study was designed in rats pre- and perinatally exposed to alcohol by means of a liquid diet.

Ethanol-induced teratogenic alterations in developing cardiomyocytes in culture.

The development of an in vitro cardiogenesis model was designed to enhance our understanding of the mechanism(s) of ethanol teratogenicity. Growth and development events in the model are similar to in vivo events. Time-specific and event-specific windows of biokinetic activities during both hyperplastic and hypertrophic growth are easily controlled and independently investigated. Systematic study of the effects of ethanol on the embryogenesis model, from committed blast cells to mature, functioning cells was accomplished to determine at what stage or stages of the growth and development paradi

Risk factors for adverse life outcomes in fetal alcohol syndrome and fetal alcohol effects.

Clinical descriptions of patients with Fetal Alcohol Syndrome (FAS) and Fetal Alcohol Effects (FAE) suggest major problems with adaptive behavior. Five operationally defined adverse outcomes and 18 associated risk/protective factors were examined using a Life History Interview with knowledgeable informants of 415 patients with FAS or FAE (median age 14 years, range 6-51; median IQ 86, range 29-126). Eighty percent of these patients were not raised by their biological mothers. For adolescents and adults, the life span prevalence was 61% for Disrupted School Experiences, 60% for Trouble with the

Effects of fetal alcohol exposure on infant reaction time.

Fetal alcohol exposure is associated with slower reaction times (RTs) in children, suggesting an alcohol-related deficit in "speed of central processing." This study examined effects of prenatal alcohol exposure on a new paradigm which, for the first time, directly assesses RT in infancy. RT was assessed in 103 Black, inner-city, 6.5-month-olds born to women recruited prenatally based on alcohol consumption during pregnancy. Maternal drinking was related to longer RTs and to fewer fast responses, after controlling for potential confounders. The incidence of fast performance was reduced in infa

Analysis of facial shape in children gestationally exposed to marijuana, alcohol, and/or cocaine.

The association between fetal marijuana and/or alcohol exposure and facial features resembling fetal alcohol syndrome was investigated in a sample of 80 children. Standardized lateral and frontal facial photographs were taken of 40 children, 5 to 7 years of age, whose mothers reported frequent use of marijuana during the first trimester of pregnancy and 40 children whose mothers reported no use of marijuana during pregnancy. The marijuana-exposed and unexposed children were group-matched on alcohol exposure prior to and during pregnancy, sex, race, and age at the time of assessment. The photog

Comparative metabolic effects of chronic ethanol intake and undernutrition in pregnant rats and their fetuses.

Female rats receiving ethanol in the drinking water before and during gestation (ET) were compared to pair-fed animals (PF) and normal controls (C) fed ad libitum. On the 21st day of gestation the maternal body and liver weight, blood glucose, and plasma protein concentrations were lower in ET and PF animals as compared to C. In contrast to C or PF mothers, ET-fed mothers had higher circulating beta-hydroxybutyrate and triacyglyceride levels and beta-hydroxy-butyrate/acetoacetate ratio. Liver triacylglycerides were increased whereas liver glycogen concentration was reduced in ET-fed animals. O

Perinatal maternal ethanol effects on pregnant mice and on offspring viability and growth: influences of exposure time and weaning diet.

C57BL/6 mice were maintained on liquid diets containing 27.5% ethanol-derived calories beginning on gestational day 12. For one group, the liquid diet was replaced with laboratory chow on the morning of gestational day 18, approximately 24 hr before delivery. For a second group, the diet was continued for 7 days postnatally. Two additional groups were pair-fed isocaloric sucrose diets, and a final group was maintained on normal laboratory chow. The mice consumed sufficient amounts of diet to provide near 30 g ethanol/kg/day during pregnancy and slightly greater amounts during the first postpar
