Congenital bovine spinal dysmyelination is caused by a missense mutation in the SPAST gene.

Bovine spinal dysmyelination (BSD) is a recessive congenital neurodegenerative disease in cattle (Bos taurus) characterized by pathological changes of the myelin sheaths in the spinal cord. The occurrence of BSD is a longstanding problem in the American Brown Swiss (ABS) breed and in several European cattle breeds upgraded with ABS. Here, we show that the disease locus on bovine chromosome 11 harbors the SPAST gene that, when mutated, is responsible for the human disorder hereditary spastic paraplegia (HSP). Initially, SPAST encoding Spastin was considered a less likely candidate gene for BSD

Permethrin may induce adult male mouse reproductive toxicity due to cis isomer not trans isomer.

Permethrin, the most popular insecticide among the synthetic pyrethroids, has been used worldwide to control a wide range of insects in agriculture, forestry, public health, and homes. Humans may have suffered potential exposure to this compound. The commercial formulation of permethrin contains trans and cis isomers. Here, at the same dosage, we made a comparison of the reproductive effects between these two isomers. Male adult ICR mice were orally administered trans- or cis-permethrin daily for 6 weeks at a dose of 0 or 70 mg/(kg day). In the cis-permethrin exposure group, the caudal epididy

Vicianin hydrolase is a novel cyanogenic beta-glycosidase specific to beta-vicianoside (6-O-alpha-L-arabinopyranosyl-beta-D-glucopyranoside) in seeds of Vicia angustifolia.

The cyanogenic disaccharide glycoside, vicianin [mandelonitrile beta-vicianoside (6-O-alpha-L-arabinopyranosyl-beta-D-glucopyranoside)], is accumulated in seeds of Vicia angustifolia var. segetalis. Vicianin hydrolase (VH) catalyzes the hydrolysis of vicianin into mandelonitrile and a disaccharide vicianose. VH was purified from the seeds using DEAE-, CM- and Con A-Sepharose chromatography, and the molecular mass of the purified VH was estimated to be 56 kDa on SDS-PAGE. The N-terminal amino acid sequence of the purified VH was determined, and a cDNA encoding VH was obtained. The deduced VH pr

La Crosse bunyavirus nonstructural protein NSs serves to suppress the type I interferon system of mammalian hosts.

La Crosse virus (LACV) is a mosquito-transmitted member of the Bunyaviridae family that causes severe encephalitis in children. For the LACV nonstructural protein NSs, previous overexpression studies with mammalian cells had suggested two different functions, namely induction of apoptosis and inhibition of RNA interference (RNAi). Here, we demonstrate that mosquito cells persistently infected with LACV do not undergo apoptosis and mount a specific RNAi response. Recombinant viruses that either express (rLACV) or lack (rLACVdelNSs) the NSs gene similarly persisted and were prone to the RNAi-med

Immunohistochemical/immunogold detection and distribution of the endosymbiont Wolbachia of Dirofilaria immitis and Brugia pahangi using a polyclonal antiserum raised against WSP (Wolbachia surface protein).

Intracellular bacteria in filarial nematodes were described as early as the 1970s, yet it was only with the work on Dirofilaria immitis, the agent of canine and feline heartworm disease, that these microorganisms were identified as belonging to Wolbachia, a genus known for encompassing bacteria infecting insects and other arthropods. The implications for the presence of intracellular bacteria in filarial nematodes is now the subject of intense research, particularly regarding their role in the immunology and pathogenesis of disease in infected humans and animals and as a possible target for th

Roles for scalloped and vestigial in regulating cell affinity and interactions between the wing blade and the wing hinge.

The scalloped and vestigial genes are both required for the formation of the Drosophila wing, and recent studies have indicated that they can function as a heterodimeric complex to regulate the expression of downstream target genes. We have analyzed the consequences of complete loss of scalloped function, ectopic expression of scalloped, and ectopic expression of vestigial on the development of the Drosophila wing imaginal disc. Clones of cells mutant for a strong allele of scalloped fail to proliferate within the wing pouch, but grow normally in the wing hinge and notum. Cells overexpressing

A novel GWT1 A397T substitution combined with TAC1B F214I reduces manogepix susceptibility and fosmanogepix efficacy in Candida auris.

Fosmanogepix (FMGX) is the prodrug of manogepix (MGX), which targets Gwt1, an inositol acyltransferase encoded by GWT1 and required for glycosylphosphatidylinositol anchor biosynthesis. To identify genetic changes associated with reduced MGX susceptibility in Candida auris, we exposed strain NCPF8971 to MGX. A stable isolate with an MGX minimum inhibitory concentration (MIC) of 1.0 µg/mL compared with 0.03 µg/mL for the parental strain carried GWT1 A397T and TAC1B F214I substitutions. Reciprocal CRISPR-Cas9 editing demonstrated that GWT1 A397T, TAC1B F214I, and the combined substitutions incre

Biochemical and histopathological investigation to study the impact of pyriproxyfen exposure on ovarian morphology and reproductive function in female rats.

BACKGROUND: Pyriproxyfen (PYR) is a pyridine-based broad-spectrum insect growth regulator and pesticide which works as an analogue of juvenile hormone. Its exposure to aquatic animals and crops is linked with various hazardous effects on biological functions. We aimed to find the possible reprotoxic effects of pyriproxyfen in adult female Sprague-Dawley rats through histological and biochemical approaches. METHODS: Adult female rats were assigned to four groups and were administered 0 mg/kg (Control), 62 mg/kg b.w, 124 mg/kg b.w, and 186 mg/kg b.w., of PYR dissolved in distilled water for 28

[Characterization of sequences, expression profiling, and natural allelic variation analysis of the MYC gene family in sorghum (Sorghum bicolor)].

Sorghum aphid (Melanaphis sacchari) and head smut fungi (Sporisorium reilianum) infesting sorghum cause delayed growth and development, and reduce yield and quality. This study use bioinformatics and molecular biological approaches to profile the gene expression pattern during sorghum development and under pest infestation, and analyzed the natural allelic DNA variation of sorghum MYC gene family. The findings provide insights for potential application in breeding the stress resistant and high productivity sorghum varieties. The results indicated that there are 28 MYC genes identified in sorgh

Co-infection of dengue and Zika viruses mutually enhances viral replication in the mosquito Aedes aegypti.

BACKGROUND: The mosquito Aedes aegypti transmits two of the most serious mosquito-borne viruses, dengue virus (DENV) and Zika virus (ZIKV), which results in significant human morbidity and mortality worldwide. The quickly shifting landscapes of DENV and ZIKV endemicity worldwide raise concerns that their co-circulation through the Ae. aegypti mosquito vector could greatly exacerbate the disease burden in humans. Recent reports have indicated an increase in the number of co-infection cases in expanding co-endemic regions; however, the impact of co-infection on viral infection and the detailed m

Bioaccumulation and Metabolic Effects of the Endocrine Disruptor Methoprene in the Lobster, Homarus americanus.

Methoprene is a pesticide that acts as a juvenile hormone agonist. Although developed initially against insects, it has since been shown to have toxic effects on larval and adult crustaceans. Methoprene was one of several pesticides applied to the Western Long Island Sound (WLIS) watershed area during the summer of 1999; the other pesticides were malathion, resmethrin, and sumethrin. These pesticides were applied as part of a county-by-county effort to control the mosquito vector of West Nile Virus. Subsequently, the seasonal lobster catches from the WLIS have decreased dramatically. The letha

t-[35S]butylbicyclophosphorothionate binding sites in invertebrate tissues.

Specific high affinity binding of the cage convulsant t-[35S]butylbicyclophosphorothionate (TBPS) was observed in membrane homogenates of housefly heads and crayfish abdominal muscles. [35S]TBPS binding in these two invertebrate tissues was inhibited by biologically active cage convulsants, picrotoxin analogs, and barbiturates. The housefly binding sites were inhibited most potently by several insecticides. Approximately 50% of total binding was displaceable by excess (0.1 mM) nonradioactive TBPS, picrotoxinin, ethyl bicyclophosphate, or dieldrin. Optimal binding assay conditions for housefly

Yeasts with potential to be used as microbial inoculants for sustainable agriculture in future.

Sustainability in agricultural production has emerged as one of the most significant concerns in the 21st Century, which recommends use of less chemical inputs having adverse effects on soil health and environment. The sustainability in the crop production system can be achieved by using beneficial soil microorganisms as microbial inoculants. A large number of studies have been carried out on bacteria and multicellular fungi which are used currently as microbial inoculants in sustainable agriculture. Yeasts, which are unicellular fungi are traditionally used for producing beverages and foods l

Acrosomal protein NEP4 is required for sperm retention in, but not entry into, the female seminal receptacle in Drosophila melanogaster.

Sperm storage in the female reproductive tract is essential for efficient progeny production in animals with internal fertilization. This is particularly so for insects, especially females that have specialized organs for long-term storage. Nep4 mutant males of Drosophila melanogaster produce an average number of morphologically normal sperm, but they cannot fertilize eggs because Nep4 sperm nuclei fail to decondense after entry into the egg. Furthermore, although Nep4 mutant sperm enter the sperm storage organs in a typical manner, they are largely lost from the seminal receptacle within 24 h

New Structures of Human Formylglycine-generating Enzyme Reveal Features Important for Catalysis, Disease and Structure-based Drug Design.

The formylglycine-generating enzyme (FGE) post-translationally modifies the active site of all human sulfatases. Mutations in the SUMF1 gene encoding FGE may lead to catalytically inactive FGE or destabilize the protein. The resulting lack of sulfatase modification causes the rare disease multiple sulfatase deficiency (MSD). Previously, FGE required elastase treatment for crystallization and the structures lacked copper, although FGE is a copper-dependent enzyme. Here, we show that highly active human FGE purified from insect cells natively contains one copper ion and we report six new crystal

Advances and challenges in building microbial cell factories for phytohormone biomanufacturing toward sustainable agriculture.

The long-term and extensive use of conventional chemical pesticides has led to a series of problems, including pesticide resistance in plant pathogens and insect pests, environmental pollution, and risks to the safety of agricultural products. These challenges have created an urgent need for green, efficient, and sustainable alternative pest management strategies. As endogenous signaling molecules, phytohormones play crucial roles in regulating plant growth and development, enhancing stress tolerance, and inducing disease resistance. Owing to their environmental compatibility, target specifici

Botulinum toxin dysregulates calcium activity and prevents Dpp/BMP secretion from epithelial cells in the developing fly wing.

It has long been thought that neurons are the only cells to utilize ion flux to temporally regulate secretion for cell-to-cell signaling, but we discovered that ectoderm and mesenchymal cells are excitable, undergo voltage-dependent calcium waves, and harness calcium influx to control secretion of essential morphogens, bone morphogenetic proteins (BMPs). Furthermore, altering ion flux during morphogenesis results in craniofacial and limb abnormalities in organisms as diverse as insects and mammals, suggesting that the role for electrical signaling is conserved. In neurons, calcium activates th

Autochthonous canine cutaneous leishmaniasis caused by Leishmania (Mundinia) martiniquensis in Thailand: First evidence with successful intralesional amphotericin B treatment and a review of animal infections worldwide.

Autochthonous human leishmaniasis caused by Leishmania (Mundinia) martiniquensis has been increasingly reported in Northern Thailand. However, the potential animal reservoirs for this parasite remain unidentified. Herein, we report the first documented case of canine cutaneous leishmaniasis caused by L. martiniquensis in a dog from Chiang Mai Province. The patient presented with a firm, non-ulcerative nodule (0.3 × 0.4 cm) on the concave surface of the right ear pinna. Fine-needle aspiration cytology revealed protozoan amastigotes, suggestive of Leishmania infection. The parasite was successfu
