In vitro evaluation of immunomodulatory, anti-diabetic, and anti-cancer molecular mechanisms of Tribulus terrestris extracts.

Dampened immunity and impaired wound healing in diabetic patients may lead to diabetic foot ulcer disease, which is the leading cause of limb amputations and hospitalization. On the other hand, cancer is the most significant cause of mortality globally, accounting for over 10 million fatalities in 2020, or nearly one in every six deaths. Plants and herbs have been used to treat chronic diseases due to their essential pharmaceutical attributes, such as mitigating drug resistance, ameliorating systemic toxicities, reducing the need for synthetic chemotherapeutic agents, and strengthening the immu

Emodin enhances host antiviral immunity against Micropterus salmoides rhabdovirus by activating RLR signaling in largemouth bass.

Micropterus salmoides rhabdovirus (MSRV) is a highly pathogenic virus that causes substantial mortality in largemouth bass, yet effective antiviral agents remain unavailable. In this study, we evaluated the antiviral potential of 20 plant-derived small molecules and identified emodin as the most effective inhibitor. At 25 mg/L, emodin reduced MSRV nucleoprotein gene expression in epithelioma papulosum cyprinid (EPC) cells by 93 %. Time-course analysis revealed a strong prophylactic effect, with viral inhibition increasing with longer pretreatment durations and peaking at 24 h (87 % inhibition)

Chemical Stimulants and Stressors Impact the Outcome of Virus Infection and Immune Gene Expression in Honey Bees (Apis mellifera).

Western honey bees (Apis mellifera) are ecologically, agriculturally, and economically important plant pollinators. High average annual losses of honey bee colonies in the US have been partially attributed to agrochemical exposure and virus infections. To examine the potential negative synergistic impacts of agrochemical exposure and virus infection, as well as the potential promise of phytochemicals to ameliorate the impact of pathogenic infections on honey bees, we infected bees with a panel of viruses (i.e., Flock House virus, deformed wing virus, or Sindbis virus) and exposed to one of thr

Nutritional Wheat Amylase-Trypsin Inhibitors Promote Intestinal Inflammation via Activation of Myeloid Cells.

BACKGROUND & AIMS: Wheat amylase-trypsin inhibitors (ATIs) are nutritional activators of innate immunity, via activation of the toll-like receptor 4 (TLR4) on myeloid cells. We aimed to characterize the biologic activity of ATIs in various foods and their effect on intestinal inflammation. METHODS: We selected 38 different gluten-containing and gluten-free products, either unprocessed (such as wheat, rye, barley, quinoa, amaranth, soya, lentils, and rice) or processed (such as pizza, pasta, bread, and biscuits). ATIs were extracted and their biological activities determined in TLR4-responsive

Acquired resistance to Listeria monocytogenes during a secondary infection in a murine model fed dietary lipids.

OBJECTIVE: The ω-3 polyunsaturated fatty acids can suppress immune system functions. This property may cause adverse effects by impairing host resistance to infection. The present study focused on estimating the impact of different dietary lipids on the immune system of mice after a secondary infection with Listeria monocytogenes. METHODS: BALB/c mice were divided into five dietary groups of olive oil, fish oil, sunflower oil, high-oleic sunflower oil, or low fat that was administered for 8 wk. The mice were immunized with 10(3) colony-forming units. Thirty-eight days later, each mouse was ch

Comparative genomic identification and validation of β-defensin genes in the Ovis aries genome.

BACKGROUND: β-defensins are small, cationic, antimicrobial peptides found in species across the plant and animal kingdoms. In addition to microbiocidal activity, roles in immunity as well as reproduction have more recently been documented. β-defensin genes in Ovis aries (domestic sheep) have been poorly annotated, having been identified only by automatic gene prediction algorithms. The objective of this study was to use a comparative genomics approach to identify and characterise the β-defensin gene repertoire in sheep using the bovine genome as the primary reference. RESULTS: All 57 currentl

Lack of enhanced T-helper cell function in uremic patients with circulating alloreactive antibodies.

Some uremic patients with a history of blood transfusion, pregnancy, and previous transplantation maintain high levels of alloreactive cytotoxic antibodies in the absence of continuous exogenous allogenic stimuli and are thus considered sensitized to the major histocompatibility proteins. To differentiate into antibody-producing cells, B lymphocytes must interact with T-helper (CD4+) cells. Whether ongoing help from these cells is necessary for the B cells to continue producing cytotoxic alloreactive antibodies in these sensitized uremic patients is unknown. To gain insight into the cellular m

Plants and Plant-Derived Compounds Mediate Protection Across Diverse Pathological Conditions by Targeting the PANoptosis Pathways.

PANoptosis is a complex inflammatory programmed cell death pathway that integrates the key mechanistic features of pyroptosis, apoptosis, and necroptosis. It represents a crucial innate immune mechanism for eliminating infected or damaged cells; however, its dysregulation is implicated in the pathogenesis of a wide spectrum of diseases, including sepsis, myocardial infarction, acute organ injuries, and cancer. Consequently, targeting PANoptosis has garnered significant interest as a novel therapeutic strategy. This review explored the potent modulatory effects of herbal medicines and their bio

Tomato Brown Rugose Fruit Virus Evades Tm-22-Mediated Resistance by Avoiding the Induction of Tm-22 Self-Association.

Tomato brown rugose fruit virus (ToBRFV) overcomes Tm-22-mediated resistance, posing a serious threat to global tomato production. We previously identified that key residues in the movement protein (MP)-H67, N125, K129, A134, I147 and I168 are essential for ToBRFV evasion of Tm-22 resistance. However, the underlying mechanisms remain unclear. In Tm-22-transgenic Nicotiana benthamiana plants at later infection stages, MP mutations H67C, N125A, K129Q and A134N abolished ToBRFV-GFP cell-to-cell and systemic movement, whereas I168N abolished ToBRFV-GFP systemic but not cell-to-cell movement, and I

Genome-Wide Analysis of CsCAX Genes and Functional Characterization of CsCAX3 Revealing Its Negative Role in Citrus Bacterial Disease Resistance.

Cation/proton antiporters (CAXs) are key membrane transporters involved in plant development and stress adaptation. In this study, five CsCAX genes (CsCAX1-CsCAX5) were identified from the Citrus sinensis genome through comprehensive bioinformatic analysis. All CsCAX proteins are hydrophobic with 10-11 transmembrane domains, predominantly composed of α-helices and random coils, and are localized to either the plasma membrane or vacuole. Phylogenetic analysis classified them into two subfamilies (IA and IB). Promoter prediction identified stress- and hormone-responsive cis-elements (e.g., LTR,
