Colonic immune stimulation by targeted oral vaccine.

BACKGROUND: Currently, sufficient data exist to support the use of lactobacilli as candidates for the development of new oral targeted vaccines. To this end, we have previously shown that Lactobacillus gasseri expressing the protective antigen (PA) component of anthrax toxin genetically fused to a dendritic cell (DC)-binding peptide (DCpep) induced efficacious humoral and T cell-mediated immune responses against Bacillus anthracis Sterne challenge. METHODOLOGY/PRINCIPAL FINDING: In the present study, we investigated the effects of a dose dependent treatment of mice with L. gasseri expressing …

Targeting the inflammasome and adenosine type-3 receptors improves outcome of antibiotic therapy in murine anthrax.

AIM: To establish whether activation of adenosine type-3 receptors (A3Rs) and inhibition of interleukin-1β-induced inflammation is beneficial in combination with antibiotic therapy to increase survival of mice challenged with anthrax spores. METHODS: DBA/2 mice were challenged with Bacillus anthracis spores of the toxigenic Sterne strain 43F2. Survival of animals was monitored for 15 d. Ciprofloxacin treatment (50 mg/kg, once daily, intraperitoneally) was initiated at day +1 simultaneously with the administration of inhibitors, and continued for 10 d. Two doses (2.5 mg/kg and 12.5 mg/kg) of a…

Multigenic control and sex bias in host susceptibility to spore-induced pulmonary anthrax in mice.

Mechanisms underlying susceptibility to anthrax infection are unknown. Using a phylogenetically diverse panel of inbred mice and spores of Bacillus anthracis Ames, we investigated host susceptibility to pulmonary anthrax. Susceptibility profiles for survival time and organ pathogen load differed across strains, indicating distinct genetic controls. Tissue infection kinetics analysis showed greater systemic dissemination in susceptible DBA/2J (D) mice but a higher terminal bacterial load in resistant BALB/cJ (C) mice. Interestingly, the most resistant strains, C and C57BL/6J (B), demonstrated a…

Anthrax lethal toxin suppresses murine cardiomyocyte contractile function and intracellular Ca2+ handling via a NADPH oxidase-dependent mechanism.

OBJECTIVES: Anthrax infection is associated with devastating cardiovascular sequelae, suggesting unfavorable cardiovascular effects of toxins originated from Bacillus anthracis namely lethal and edema toxins. This study was designed to examine the direct effect of lethal toxins on cardiomyocyte contractile and intracellular Ca(2+) properties. METHODS: Murine cardiomyocyte contractile function and intracellular Ca(2+) handling were evaluated including peak shortening (PS), maximal velocity of shortening/ relengthening (± dL/dt), time-to-PS (TPS), time-to-90% relengthening (TR(90)), intracellul…

Effect of aluminum hydroxide adjuvant and formaldehyde in the formulation of rPA anthrax vaccine.

The serological response and efficacy of Bacillus anthracis recombinant protective antigen (rPA) vaccines formulated with aluminum hydroxide adjuvant, either with or without formaldehyde, were evaluated in rabbits. Rabbits that had been injected with a single dose of 25 microg of rPA adsorbed to 500 microg of aluminum in aluminum hydroxide gel (Alhydrogel) had a significantly higher quantitative anti-rPA IgG ELISA titers (p<0.0001) and toxin neutralizing antibody (TNA) assay titers (p<0.0001) than rabbits tested at the next lowest concentration of aluminum (158 microg). Rabbits injected with t…

Dual-ligand lanthanide metal-organic framework for ratiometric fluorescence detection of the anthrax biomarker dipicolinic acid.

Dipicolinic acid (DPA) is a unique biomarker of Bacillus anthracis. Development of a simple, fast, sensitive and timely DPA detection method is of great importance and interest for preventing mass disease outbreaks and treatment of anthrax. In this work, a novel lanthanide-doped fluorescence probe was constructed by coordination of Eu3+ with bifunctional UiO-66-(COOH)2-NH2 MOFs materials for efficient monitoring DPA. UiO-66-(COOH)2-NH2 MOFs were prepared using Zr4+ as a metal node, 1,2,4,5-benzenetetracarboxylic acid (H4BTC) and 2-aminoterephthalic acid (NH2-BDC) as bridging ligand through a s…

The role of l-histidine as molecular tongs: a strategy of grasping Tb3+ using ZIF-8 to design sensors for monitoring an anthrax biomarker on-the-spot.

In this study, a novel lanthanide-doped nanoprobe for monitoring dipicolinic acid (DPA), a unique biomarker of Bacillus anthracis, was constructed by coordination of Tb3+ with l-histidine (His) functionalized ZIF-8 (His@ZIF-8). After being functionalized with His, the resultant His@ZIF-8 had abundant carboxyl and amino groups, which like tongs help His@ZIF-8 "grasp" Tb3+ firmly to form a stable lanthanide-doped nanoparticle (His@ZIF-8/Tb3+). Owing to the unsaturated coordination of Tb3+ with the amino acid group, the resultant His@ZIF-8/Tb3+ showed reserved response sites of Tb3+ to DPA becaus…

A Nasal Vaccine Displaying Anthrax Antigen on the Surface of Lactiplantibacillus plantarum Induces Protective Mucosal Immunity against Anthrax Toxin.

Anthrax, particularly inhalational anthrax, poses a severe threat due to its high fatality rate and potential misuse as a biological weapon. Current anthrax vaccines require multiple injections, which limits their practicality and underscores the need for improved, user-friendly vaccination strategies. In this study, we developed a novel nasal booster vaccine platform aimed at preventing inhalational anthrax by inducing mucosal immunity at the pathogen's entry site, the respiratory tract. We successfully engineered a biologically safe Lactiplantibacillus plantarum (L. plantarum) strain to disp

Secondary Structure Preferences of the Anthrax Toxin Protective Antigen Translocase.

In order for many proteins to move across hydrophobic membrane bilayers, they must be unfolded and translocated by a membrane-embedded channel. These translocase channels interact with the substrate proteins they translocate via hydrophobic pore loops and cleft structures called clamps. The molecular basis for how clamps facilitate unfolding and translocation is poorly understood. Anthrax toxin is composed of three proteins, a translocase channel-forming subunit, called protective antigen (PA), and two substrate proteins, called lethal factor (LF) and edema factor. Oligomeric PA forms a large

Immunization of Mice with Anthrax Protective Antigen Limits Cardiotoxicity but Not Hepatotoxicity Following Lethal Toxin Challenge.

Protective immunity against anthrax is inferred from measurement of vaccine antigen-specific neutralizing antibody titers in serum samples. In animal models, in vivo challenges with toxin and/or spores can also be performed. However, neither of these approaches considers toxin-induced damage to specific organ systems. It is therefore important to determine to what extent anthrax vaccines and existing or candidate adjuvants can provide organ-specific protection against intoxication. We therefore compared the ability of Alum, CpG DNA and the CD1d ligand α-galactosylceramide (αGC) to enhance prot

Inhalational anthrax (Ames aerosol) in naïve and vaccinated New Zealand rabbits: characterizing the spread of bacteria from lung deposition to bacteremia.

There is a need to better understand inhalational anthrax in relevant animal models. This understanding could aid risk assessment, help define therapeutic windows, and provide a better understanding of disease. The aim here was to characterize and quantify bacterial deposition and dissemination in rabbits following exposure to single high aerosol dose (> 100 LD(50)) of Bacillus anthracis (Ames) spores immediately following exposure through 36 h. The primary goal of collecting the data was to support investigators in developing computational models of inhalational anthrax disease. Rabbits were …
