Cryoglobulins and pyroglobulins: an overview.

Cryoglobulins are serum proteins with heterogeneous etiopathogenetic and immunochemical properties. What they have in common is temperature-dependent insolubility, in that at temperatures below 37 degrees C (often around 4 degrees C) they precipitate, and then redissolve at 37 degrees C. When the etiopathogenesis of the cryoglobulinemia is unknown, which is true for many patients, the condition is called idiopathic or essential cryoglobulinemia, whereas it is termed secondary whenever it appears to be associated with one of several diseases. Cryoglobulinemia has indeed been found in patients w

Discovery of Antiamebic Compounds That Inhibit Cysteine Synthase From the Enteric Parasitic Protist Entamoeba histolytica by Screening of Microbial Secondary Metabolites.

Amebiasis is caused by infection with the protozoan parasite Entamoeba histolytica. Although metronidazole has been a drug of choice against amebiasis for decades, it shows side effects and low efficacy against asymptomatic cyst carriers. In addition, metronidazole resistance has been documented for bacteria and protozoa that share its targets, anaerobic energy metabolism. Therefore, drugs with new mode of action or targets are urgently needed. L-cysteine is the major thiol and an essential amino acid for proliferation and anti-oxidative defense of E. histolytica trophozoites. E. histolytica p

Severe corneal melting with necrotic sloughing in Acanthamoeba keratitis mimicking corneal endotheliitis.

BACKGROUND: Acanthamoeba keratitis (AK) is a sight-threatening corneal infection that may initially lack typical epithelial or perineural findings and mimic other infectious or inflammatory anterior segment disorders. Endotheliitis-like presentations of AK with stromal edema, keratic precipitates, Descemet membrane folds, and anterior chamber inflammation have been reported, but progression to severe stromal necrosis with spontaneous sloughing of necrotic corneal tissue is rarely documented. CASE PRESENTATION: A 48-year-old contact lens wearer presented with right eye pain, redness, and decre

Hyphosphere interactions: P-solubilizing fungi modulate AMF phosphatase activity and mycorrhizal symbiosis via exudate-mediated communication.

Arbuscular mycorrhizal fungi (AMF) form symbiotic associations with plant roots, enhancing water and nutrient absorption. Phosphate-solubilizing fungi (PSF) can solubilize and mineralize phosphorus, an essential nutrient with low bioavailability, and eventually interact with AMF. However, the understanding of how they interact in the hyphosphere, where root influence is absent, remains limited. Furthermore, the effect of PSF on the phosphatase activity of AMF, related to the P efficiency in acquisition and utilization, within the hyphosphere and mycorrhizosphere zones, remains unclear. RNAi of Mannosidase-Ia in the Colorado potato beetle and changes in the midgut and peritrophic membrane.

BACKGROUND: In addition to its role in the digestive system, the peritrophic membrane (PM) provides a physical barrier protecting the intestine from abrasion and against pathogens. Because of its sensitivity to RNA interference (RNAi), the notorious pest insect, the Colorado potato beetle (CPB, Leptinotarsa decemlineata), has become a model insect for functional studies. Previously, RNAi-mediated silencing of Mannosidase-Ia (ManIa), a key enzyme in the transition from high-mannose glycan moieties to paucimannose N-glycans, was shown to disrupt the transition from larva to pupa and the metamorp

Untargeted Metabolomics Approach for the Discovery of Environment-Related Pyran-2-ones Chemodiversity in a Marine-Sourced Penicillium restrictum.

Very little is known about chemical interactions between fungi and their mollusc host within marine environments. Here, we investigated the metabolome of a Penicillium restrictum MMS417 strain isolated from the blue mussel Mytilus edulis collected on the Loire estuary, France. Following the OSMAC approach with the use of 14 culture media, the effect of salinity and of a mussel-derived medium on the metabolic expression were analysed using HPLC-UV/DAD-HRMS/MS. An untargeted metabolomics study was performed using principal component analysis (PCA), orthogonal projection to latent structure discr

Microbial production of eumelanin from a desert-derived Aspergillus terreus: statistical optimization, comprehensive characterization, and in silico insights into breast cancer-associated targets.

BACKGROUND: Naturally derived microbial products are increasingly explored as sustainable sources of bioactive compounds for targeted cancer applications. Among these, eumelanin is a nitrogen-containing polymeric pigment produced by various microorganisms, including fungi, and is distinguished by its unique physicochemical properties, biocompatibility, and free radical scavenging capacity. Nevertheless, eumelanin-producing fungi from extreme desert habitats remain poorly explored, particularly those isolated from the Wadi Allaqi Biosphere Reserve, and comprehensive studies integrating producti

Candida albicans Goliath cells pioneer biofilm formation.

Candida albicans is a fungal pathogen that can cause life-threatening bloodstream infections. Indwelling medical devices, such as catheters, play a critical role in their progression, as C. albicans can colonize these devices and form biofilms, which can then seed the bloodstream, resulting in recurrent candidemia. Under conditions of zinc restriction, C. albicans forms enlarged Goliath cells. Here, we show that Goliath cells exhibit enhanced adhesion to host tissue under static conditions, particularly to vaginal and oral epithelial cell monolayers. Moreover, Goliath cells are fully capable o

Synthesis, Characterization, Antimicrobial Activity and Molecular Modeling Studies of Novel Indazole-Benzimidazole Hybrids.

Background/Objectives: In this work, a series of six new indazole-benzimidazole hybrids (M1-M6) were designed, synthesized, and fully characterized. The design of these compounds was based on the combination of two pharmacophoric units, indazole and benzimidazole, both known for their broad spectrum of biological activities. Methods: The molecular hybridization strategy was planned to combine these scaffolds through an effective synthetic pathway, using 6-nitroindazole, two 2-mercaptobenzimidazoles, and 1,3- or 1,5-dihaloalkanes as key precursors, affording the desired hybrids in good yields a

Human α-Defensin 6: A Small Peptide That Self-Assembles and Protects the Host by Entangling Microbes.

Human α-defensin 6 (HD6) is a 32-residue cysteine-rich peptide that contributes to innate immunity by protecting the host at mucosal sites. This peptide is produced in small intestinal Paneth cells, stored as an 81-residue precursor peptide named proHD6 in granules, and released into the lumen. One unusual feature of HD6 is that it lacks the broad-spectrum antimicrobial activity observed for other human α-defensins, including the Paneth cell peptide human α-defensin 5 (HD5). HD6 exhibits unprecedented self-assembly properties, which confer an unusual host-defense function. HD6 monomers self-as

A broadly conserved fungal chorismate mutase targets the plant shikimate pathway to regulate salicylic acid production and other secondary metabolites.

The molecular dynamics of plant-pathogen interactions are complex, involving a constant defense race between host plants and pathogens. Plants deploy antimicrobial metabolites and physiological responses to inhibit pathogens, while pathogens counteract these defenses using secreted proteinaceous effectors. Chorismate mutases (CMs) are effectors largely studied in biotrophic fungi and nematodes for their ability to hijack the host shikimate pathway by diverting chorismate from salicylic acid (SA) biosynthesis, subsequently undermining SA-mediated defenses against biotrophic infection. While CMs
