Influenza virus A(H1N1)pdm09 hemagglutinin polymorphism and associated disease in southern Germany during the 2010/11 influenza season.

A novel influenza A virus emerged in early 2009 to cause the first influenza pandemic of the 21(st) century. Understanding the evolution of influenza virus is crucial to determine pathogenesis, vaccine efficacy, and resistance to antiviral drugs. In this study, we investigated the molecular evolution of influenza virus A(H1N1)pdm09 in the 2010/11 influenza season in southern Germany by sequence analysis of the influenza virus hemagglutinin gene from 25 patients with mild, moderate, and severe disease. Phylogenetic analysis revealed co-circulation of different genetic groups. The D222G mutation

Dual color microsphere encoded by fluorescent protein and Cy5-labeled anti-Tag ssDNA for multiplex detection of respiratory pathogens.

Respiratory diseases are challenging to diagnose rapidly and accurately due to diverse pathogens, overlapping symptoms, and high infection rates. Here, we developed a suspension array based on magnetic microspheres encoded with blue fluorescent protein (mTagBFP) and Cy5-labeled anti-Tag ssDNA (MB@mTagBFP&anti-Tag-Cy5) for simultaneous detection of five core respiratory pathogens. Multiplex PCR was performed using 5'-Tag-labeled upstream primers and 5'-biotinylated downstream primers, yielding dual-labeled products (Tag and biotin). The resulting complexes (MB@mTagBFP&anti-Tag-Cy5-Tag-dsDNA-phy

Multiplex PCR-Mass Spectrometry Mini-Sequencing Technology Detected Antibiotic Resistance of Helicobacter pylori to Six Antibiotics.

The abuse of antibiotics has led to widespread resistance to Helicobacter pylori (H. pylori) in the population. There is an urgent need to establish a method to detect multiple antibiotic resistance rapidly. This study aimed to construct a novel strategy for the high-throughput detection of H. pylori's resistance to varying antibiotics using multiplex PCR-mass spectrometry mini-sequencing (mPCR-MS mini-sequencing) technology. This study detected the resistance of H. pylori to six antibiotics using eight mutated sites (23S rRNA-2143; pbp1A-1667, 1684, 1240; gyrA-261, 271, 573; and 16S rRNA-928)

Molecular diagnosis of respiratory viral infections in cystic fibrosis, sputum versus nasopharyngeal samples.

BACKGROUND: Respiratory viral infections in people with Cystic Fibrosis (pwCF) significantly impact disease progression, and their molecular diagnosis should be part of the recommendations. Sputum, the routinely used specimen for bacterial culture, could also be used for viral molecular diagnosis, avoiding an additional invasive nasopharyngeal swab or aspirate. Our primary objective was to evaluate whether invasive nasopharyngeal samples (NP) and non-invasive sputum specimens are equally accurate for the diagnosis of viral infections in pwCF. METHODS: We conducted a prospective and multicentr

Rapid detection of carbapenem resistance in Klebsiella pneumoniae clinical isolates: Flow cytometry as an alternative to multiplex PCR.

BACKGROUND: Klebsiella pneumoniae is a leading cause of diverse infections, making rapid diagnosis and effective treatment critical to reducing adverse outcomes. Carbapenems, the most potent beta-lactam antibiotics, are widely used due to their broad-spectrum activity. However, the rise and spread of carbapenem-resistant strains pose a significant global health threat. We aimed to compare the sensitivity and specificity of flow cytometry versus multiplex PCR for detecting carbapenemase classes A, B, and D in clinical isolates of K. pneumoniae. METHODS: A total of 671 clinical samples were col

Ct Values Do Not Predict Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) Transmissibility in College Students.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is highly contagious and has caused significant medical/socioeconomic impacts. Other than vaccination, effective public health measures, including contact tracing, isolation, and quarantine, is critical for deterring viral transmission, preventing infection progression and resuming normal activities. Viral transmission is affected by many factors, but the viral load and vitality could be among the most important ones. Although in vitro studies have indicated that the amount of virus isolated from infected individuals affects the succ

Sensitivity assessment of workflows detecting rare circulating cell-free DNA targets: A study design proposal.

The field of liquid biopsy has seen extensive growth in recent decades, making it one of the most promising areas in molecular diagnostics. Circulating cell-free DNA (ccfDNA) especially is used as an analyte in a growing number of diagnostic assays. These assays require specified preanalytical workflows delivering ccfDNA in qualities and quantities that facilitate correct and reliable results. As each step and component used in the preanalytical process has the potential to influence the assay sensitivity and other performance characteristics, it is key to find an unbiased experimental setup t

A combination of naso- and oropharyngeal swabs improves the diagnostic yield of respiratory viruses in adult emergency department patients.

BACKGROUND: Along with the current development of molecular diagnostic methods of respiratory viruses, the bedside patient sampling techniques need to be evaluated. We here asked the question whether the addition of an oropharynx swab to the traditional nasopharynx swab might improve the diagnostic yield of multiplex PCR analysis. Ct values from the two sampling sites were compared as well as patient tolerability. METHODS: In an emergency department in Malmö, Sweden, 98 adult patients with respiratory disease were sampled both from the nasopharynx and oropharynx for virus diagnostics by PCR.

Detection of HPV RNA molecules in stratified mucin-producing intraepithelial lesion (SMILE) with concurrent cervical intraepithelial lesion: a case report.

BACKGROUND: Stratified mucin-producing intraepithelial lesion (SMILE) is a rare precursor lesion in the uterine cervix that is considered a variant of adenocarcinoma in situ (AIS). Although human papillomavirus (HPV) is thought to be related to the development of SMILE, there is little information available on the detection of HPV integrated into the lesion. CASE PRESENTATION: A 30-year-old female underwent a routine uterine cervical cancer screening, and her Pap smear indicated the possible existence of atypical glandular cells. A cervical biopsy with endocervical curettage was performed. Th

Monitoring arbovirus in Thailand: Surveillance of dengue, chikungunya and zika virus, with a focus on coinfections.

Infections caused by arboviruses such as dengue virus (DENV), chikungunya virus (CHIKV), and Zika virus (ZIKV) frequently occur in tropical and subtropical regions. These three viruses are transmitted by Aedes (Ae.) aegypti and Ae. albopictus. In Thailand, the highest incidence of arbovirus infection and the high circulation of Aedes mosquito mainly occurs in the Southern provinces of the country. Few studies have focused on the incidence of co-infection of arboviruses in this region. In the present study, a cross-sectional study was conducted on a cohort of 182 febrile patients from three hos

Early-Onset Obesity: Unrecognized First Evidence for GNAS Mutations and Methylation Changes.

CONTEXT: Early-onset obesity, characteristic for disorders affecting the leptin-melanocortin pathway, is also observed in pseudohypoparathyroidism type 1A (PHP1A), a disorder caused by maternal GNAS mutations that disrupt expression or function of the stimulatory G protein α-subunit (Gsα). Mutations and/or epigenetic abnormalities at the same genetic locus are also the cause of pseudohypoparathyroidism type 1B (PHP1B). However, although equivalent biochemical and radiographic findings can be encountered in these related disorders caused by GNAS abnormalities, they are considered distinct clini
