[Effect of low-dose primaquine treatment on Plasmodium vivax recurrence and transmission-blocking activity in southwest Ethiopia: a longitudinal cohort study.

BACKGROUND: Existing malaria control strategies for Plasmodium vivax are challenging due to its dormant and relapsing liver stages, as well … r one year. To assess the effect of different treatment regimens on transmission-blocking activity, Anopheles arabiensis mosquitoes were used in direct membrane-feeding assays (DMFA) at baseline (pre-treatment) and on day 42 (post-treatment). Based on polymerase chain reaction (PCR) positivity, the time to the first recurrence was estimated using Kaplan-Mei …

Human and entomological determinants of malaria transmission in the Lihir Islands of Papua New Guinea: A cross-sectional study.

BACKGROUND: The Lihir Islands of Papua New Guinea, located in an area with high burden of malaria and hosting a large mining operation, offer a unique opportunity to study transmission. There, we investigated human and vector factors influencing malaria transmission. METHODS: In 2019, a cross-sectional study was conducted on 2,914 individuals assessing malaria prevalence through rapid diagnostic tests (RDT), microscopy, and quantitative PCR (qPCR). A logistic regression analysis identified infection-associated factors. Anopheles species distribution, biting behaviours, and sporozoite carriage…

Repeat controlled human Plasmodium falciparum infections delay bloodstream patency and reduce symptoms.

Resistance to clinical malaria takes years to develop even in hyperendemic regions and sterilizing immunity has rarely been observed. To evaluate the maturation of the host response against controlled repeat exposures to P. falciparum (Pf) NF54 strain-infected mosquitoes, we systematically monitored malaria-naïve participants through an initial exposure to uninfected mosquitoes and 4 subsequent homologous exposures to Pf-infected mosquitoes over 21 months (n = 8 males) (ClinicalTrials.gov# NCT03014258). The primary outcome was to determine whether protective immunity against parasite infection

A single-cell atlas of Plasmodium falciparum transmission through the mosquito.

Malaria parasites have a complex life cycle featuring diverse developmental strategies, each uniquely adapted to navigate specific host environments. Here we use single-cell transcriptomics to illuminate gene usage across the transmission cycle of the most virulent agent of human malaria - Plasmodium falciparum. We reveal developmental trajectories associated with the colonization of the mosquito midgut and salivary glands and elucidate the transcriptional signatures of each transmissible stage. Additionally, we identify both conserved and non-conserved gene usage between human and rodent para

Understanding heterogeneities in mosquito-bite exposure and infection distributions for the elimination of lymphatic filariasis.

It is well known that individuals in the same community can be exposed to a highly variable number of mosquito bites. This heterogeneity in bite exposure has consequences for the control of vector-borne diseases because a few people may be contributing significantly to transmission. However, very few studies measure sources of heterogeneity in a way which is relevant to decision-making. We investigate the relationship between two classic measures of heterogeneity, spatial and individual, within the context of lymphatic filariasis, a parasitic mosquito-borne disease. Using infection and mosquit

The impact of repeated rounds of mass drug administration with diethylcarbamazine plus albendazole on bancroftian filariasis in Papua New Guinea.

BACKGROUND: This study employed various monitoring methods to assess the impact of repeated rounds of mass drug administration (MDA) on banc … igenemia, and antifilarial antibodies also decreased significantly after MDA. Filarial DNA rates in Anopheles punctulatus mosquitoes that had recently taken a blood meal decreased from 15.1% to 1.0% (a 92.3% decrease). CONCLUSIONS/SIGNIFICANCE: MDA had dramatic effects on all filariasis parameters in the study area and also reduced incidence rates. Follow- …

Onchocerciasis, malaria and trypanosomiasis in three resettlement schemes in western Ethiopia.

Epidemiological studies were carried out among 180 randomly chosen settler and 180 non-settler households in the three resettlement schemes of Kishe, Gera and Didessa located in river valleys and highland areas of Illubabor Administrative Region in western Ethiopia. Up to 49% of the indigenous populations (in Kishe) and 0.9% of the settlers had onchocerciasis, with a mean density of 13.6 filariae per slide/skin snip for indigenous people and 9.4 for settlers. Onchocerciasis prevalence rates were higher in males than females and were highest in the 20-59 age group. In the Kishe scheme, rates we

The treatment and control of parasitic diseases.

In many parts of world, transmission of malaria due to Plasmodium falciparum can no longer be controlled by insecticides. Furthermore, this species has developed an amazing capacity for resistance to polyvalent drugs. The advent of new drugs and the possibility of developing a vaccine offer some hope for control of falciparum malaria. The fact that the spread of many parasitic diseases is promoted by contaminated water supplies resulting from inadequate waste disposal raises important questions about the relevance of current research. The control of some endemic infections, like Chagas' diseas

"Referral patterns and associated factors among adult patients admitted with Severe Malaria at Shedeho Meket Primary Hospital, Northeast Ethiopia".

INTRODUCTION: Malaria is a protozoan disease transmitted by the bite of infected female Anopheles mosquitoes, with severe and fatal cases predominantly caused by Plasmodium falciparum. Despite a lower prevalence compared to other endemic African nations, malaria remains a significant public health challenge in Ethiopia. This study aimed to assess the referral patterns and associated factors among adult patients admitted with severe malaria at Shedeho Meket Primary Hospital in Northeast Ethiopia. METHODS: An institution-based retrospective cross-sectional study was conducted at Shedeho Meket P…

Determinants of Malaria Vaccine Acceptance: A Systematic Review and Meta-Analysis of Awareness, Acceptance, Hesitancy, and Willingness to Pay.

BACKGROUND: Malaria is a life-threatening disease caused by Plasmodium parasites, transmitted through the bites of infected female Anopheles mosquitoes. It remains a major global health issue, with 263 million cases and 597,000 deaths in 2023, primarily affecting young children and pregnant women. This review evaluates awareness, acceptance, hesitancy, and willingness to pay (WTP) for the RTS,S/AS01 malaria vaccine, along with the key factors influencing these outcomes. METHODS: A comprehensive literature search was conducted in Web of Science, PubMed, and Embase, covering publications up to …

Bioinformatics characterization of Plasmodium knowlesi apical membrane antigen 1 (PkAMA1) for multi-epitope vaccine design.

Malaria caused by Plasmodium knowlesi species has become a public health concern, especially in Malaysia. Plasmodium knowlesi parasite which originates from the macaque species, infects human through the bite of the Anopheles mosquitoes. Research on malaria vaccine has been a continuous effort to eradicate the malaria infection, yet there is no vaccine against P. knowlesi malaria to date. Apical membrane antigen 1 (AMA1) is a unique surface protein of all apicomplexan parasites that plays a crucial role in parasite-host cell invasion and thus has been a long-standing malaria vaccine candidate.…

Molecular and epidemiological characterization of imported malaria cases in Chile.

BACKGROUND: Chile is one of the South American countries certified as malaria-free since 1945. However, the recent increase of imported malaria cases and the presence of the vector Anopheles pseudopunctipennis in previously endemic areas in Chile require an active malaria surveillance programme. METHODS: Specimens from 268 suspected malaria cases-all imported-collected between 2015 and 2018 at the Public Health Institute of Chile (ISP), were diagnosed by microscopy and positive cases were included for epidemiological analysis. A photo-induced electron transfer fluorogenic primer real-time PCR…
