AIMS: Obesity is characterized by a low-grade systemic inflammatory state and adipose tissue (AT) dysfunction, which predispose individuals to the development of insulin resistance (IR) and metabolic disease. However, a subset of obese individuals, referred to as metabolically healthy obese (MHO) individuals, are protected from obesity-associated metabolic abnormalities. Here, we aim at identifying molecular factors and pathways in adipocytes that are responsible for the progression from the insulin-sensitive to the insulin-resistant, metabolically unhealthy obese (MUHO) phenotype.

This study tested whether the glycogen-accumulating effect of chronic in vivo pharmacological 5'AMP-activated protein kinase (AMPK) activation could improve glycemic control under conditions of insulin deficiency. Male Wistar rats were rendered diabetic through the administration of streptozotocin (STZ) and then treated for 7 consecutive days with the AMPK activator 5-aminoimidazole-4-carboxamide-1-β-D-ribofuranoside (AICAR). Subsequently, glycogen content and synthesis, glucose oxidation, and fatty acid oxidation (FAO) were determined in oxidative and glycolytic skeletal muscles. Glycemia, in

A palmitate linked to the inositol in glycosylphosphatidylinositol (GPI) is removed in the endoplasmic reticulum immediately after the conjugation of GPI with proteins in most cells. Previously, we identified PGAP1 (post GPI attachment to proteins 1) as a GPI inositoldeacylase that removes the palmitate from inositol. A defect in PGAP1 caused a delay in the transport of GPI-anchored proteins (GPI-APs) from the endoplasmic reticulum to the cell surface in Chinese hamster ovary cells, although the cell-surface expression of GPI-APs in the steady state was normal. Nevertheless, in most cells, GPI

Metabolic dysfunction-associated steatotic liver disease (MASLD) is characterized by dysregulated hepatic lipid metabolism and persistent inflammatory remodeling. Apigenin has lipid-regulatory and anti-inflammatory activities, but the molecular basis linking these effects remains incompletely understood. This study investigated whether SMAD4-associated signaling contributes to the coordinated regulation of hepatocyte lipogenesis and macrophage inflammatory activation by apigenin. C57BL/6J mice were fed a high-fat diet (HFD) to establish MASLD and treated with apigenin. Palmitic acid (PA)-treat

BACKGROUND: Diabetic kidney disease (DKD) is the leading cause of chronic kidney disease (CKD) and end-stage renal disease in type 2 diabetes mellitus (T2DM) patients. Recent studies have demonstrated that pregnancy zone protein (PZP), a hepatic cytokine, plays a role in regulating glucose and lipid metabolism. This retrospective study aimed to investigate the connection between plasma PZP concentrations and DKD.
MATERIALS AND METHODS: All participants were stratified by estimated glomerular filtration rate (eGFR) and urine albumin-to-creatinine ratio (UACR). The discovery cohort included 130

AIMS/HYPOTHESIS: Metabolic dysfunction-associated steatotic liver disease (MASLD) is a common feature of obesity and type 2 diabetes. Under lipotoxic stress, hepatocytes release small extracellular vesicles (sEVs) which act locally and contribute to MASLD progression, but their role in beta cell function and development of type 2 diabetes remains largely unexplored. We aimed to examine whether hepatocyte-derived sEVs (Hep-sEVs) under lipotoxic conditions impact on liver and pancreas inflammation and subsequent effects on beta cell function.
METHODS: Primary mouse hepatocytes and Huh7 human he

Metabolic associated fatty liver disease (MAFLD) is the leading cause of chronic liver disease worldwide. Various studies have demonstrated that traditional Chinese medicine alleviates MAFLD by suppressing PANoptosis. Although Oridonin (ORI) is a traditional Chinese medicine that has been used in hepatosteatosis, its underlying mechanism for treating MAFLD, especially regarding PANoptosis, remains unclear. Therefore, this study aims to investigate the ameliorative effects of ORI on MAFLD by inhibiting PANoptosis and to elucidate the underlying mechanisms. A vivo model of MAFLD was constructed

BACKGROUND/AIMS: The molecular mechanisms driving nonalcoholic steatohepatitis (NASH) progression are poorly understood. This research examines the involvement of chaperone-mediated autophagy (CMA) in NASH progression.
METHODS: Hepatic CMA activity was analysed in NASH mice and patients. Lysosome-associated membrane protein 2A (LAMP2A) was knocked down or overexpressed to assess the effects of hepatocyte-specific CMA on NASH progression. Mice received a high-fat diet or a methionine and choline-deficient diet to induce NASH. Palmitic acid was employed to mimic lipotoxicity-induced hepatocyte

BACKGROUND: Panax japonicus (T. Nees) C.A. Mey., a member of the Araliaceae family, has been utilized for medicinal purposes by the Tujia and Hmong ethnic groups in China for over a thousand years. In traditional Chinese medicine, metabolic dysfunction-associated fatty liver disease (MAFLD) is classified as a damp-heat syndrome, and P. japonicus is believed to strengthen the spleen and the stomach, promoting fat transformation. Previous studies have shown that the total saponins from P. japonicus (TSPJ) are the primary active constituents that exert hepatoprotective, lipid-lowering, and anti-i

A hyperstable lipase from Thermotoga naphthophila (TnLip) was cloned and overexpressed as a soluble and active monomeric protein in an effectual mesophilic host system. Sequence study revealed that TnLip is a peptidase S9 prolyl oligopeptidase domain (acetyl esterase/lipase-like protein), belongs to alpha/beta (α/β)-hydrolase superfamily containing a well-conserved α/β-hydrolase fold and penta-peptide (GLSAG) motif. Various cultivation and induction strategies were applied to improve the heterologous expression and bacterial biomass, but TnLip intracellular activity was enhanced by 14.25- fold

Liraglutide (LRG), one agonist of glucagon-like peptide-1 receptor (GLP1R), has multiple lipid-lowering effects in type 2 diabetes mellitus, however, studies on the role of LRG in saturated fatty acid-induced bone loss are limited. Therefore, our aim was to investigate whether LRG reduces palmitate (PA)-induced apoptosis and whether the mechanism involves PKA/β-catenin/Bcl-2/Bax in osteoblastic MC3T3-E1 cells. MC3T3-E1 cells were treated with different concentrations of PA, LRG, or pretreated with Exendin 9-39 and H89, cell viability, intracellular reactive oxygen species (ROS), cAMP levels, a

Diabetic patients are susceptible to infectious diseases. Bacterial invasion activates immune cells such as macrophages through interaction between LPS and TLR4, and induces the expression of inflammatory mediators, including IL-1β and TNF-α, which play key roles in the elimination of infections. Unregulated overproduction or underproduction of these cytokines has been reported as a major factor in the development of septic shock, immune deficiency, and autoimmunity. Recent studies found that metabolic abnormalities of diabetes, such as hyperglycemia and dyslipidemia, played a major role in mo

Pad Thai, a Thai dish of stir-fried rice noodles and other ingredients, is one of the culinary heritages of Thailand. In the southern region of Thailand, Pad Thai has different characteristics from other areas because coconut milk and curry paste are used to produce a thick sauce prior to cooking. To commercially distribute this uniquely local culinary treat in a convenient form all over Thailand and other export markets, a shelf-stable sauce using heat sterilization should be developed. Retort processing technology with retort pouches can be used for this purpose. However, phase separation an
