Surveillance methods of circulating antibiotic resistance genes (ARGs) are of utmost importance in order to tackle what has been described as one of the greatest threats to humanity in the 21st century. In order to be effective, these methods have to be accurate, quickly deployable, and scalable. In this study, we compare metagenomic shotgun sequencing (TruSeq DNA sequencing) of wastewater samples with a state-of-the-art PCR-based method (Resistomap HT-qPCR) on four wastewater samples that were taken from hospital, industrial, urban and rural areas. ARGs that confer resistance to 11 antibiotic

BACKGROUND: Mycobacterium bovis BCG is a live, attenuated tuberculosis vaccine. While the vaccine protects infants from tuberculosis, complications including disseminated infections have been reported following vaccination. Genetically diverse BCG sub-strains now exist following continuous passaging of the original Pasteur strain for vaccine manufacture. This genetic diversity reportedly influences the severity of disseminated BCG infections and the efficacy of BCG immunization.
METHODS: M. bovis BCG was isolated from infants suspected of being infected with tuberculosis. The whole genome of

Because a shortage of new antimicrobial agents is a critical issue at present, and with the spread of multidrug-resistant (MDR) pathogens, the use of fosfomycin to treat infections is being revisited as a "last-resort option." This drug offers a particular benefit in that it is more effective against bacteria growing under oxygen-limited conditions, unlike other commonly used antimicrobials, such as fluoroquinolones and aminoglycosides. In this study, we showed that Escherichia coli strains, including enterohemorrhagic E. coli (EHEC), were more susceptible to fosfomycin when grown anaerobicall

BACKGROUND: Extended-spectrum cephalosporins (ESCs) are third and fourth generation cephalosporin antimicrobials used in humans and animals to treat infections due to multidrug-resistant (MDR) bacteria. Resistance to ESCs (ESC-R) in Enterobacterales is predominantly due to the production of extended-spectrum β-lactamases (ESBLs) and plasmid-mediated AmpC β-lactamases (AmpCs). The dynamics of ESBLs and AmpCs are changing across countries and host species, the result of global transmission of ESC-R genes. Plasmids are known to play a key role in this dissemination, but the relative importance of

Staphylococcus epidermis is one of the most frequent causes of device-associated infections due to biofilm formation. Current reports noted that subinhibitory concentrations of antibiotics induce biofilm production in some bacteria. Accordingly, we evaluated the effect of exposure of different subinhibitory concentrations of cloxacillin, cefazolin, clindamycin, and vancomycin on the biofilm formation of methicillin-resistant S. epidermidis (MRSE). Antimicrobial susceptibility testing and minimum inhibitory/bactericidal concentration of antimicrobial agents were determined. MRSE isolates were s

BACKGROUND: Klebsiella pneumoniae is a well-known opportunistic pathogen in humans and can cause chronic obstructive pulmonary disease (COPD) in Cystic Fibrosis (CF) patients. This pathogen has developed resistance to β-lactam antibiotics due to the expression of extended-spectrum beta-lactamase (ESBL) genes, leading to current treatment challenges in these patients. Bacteriophages are alternative and effective treatment options against multidrug-resistant (MDR) pathogens. In this study, a lytic bacteriophage was isolated from untreated sewage, tested against multidrug-resistant K. pneumoniae

Pseudomonas aeruginosa (P. aeruginosa) is one of the most common ones that harm fish. P. aeruginosa has been regarded as one of the most significant threats to the fishing industry, which also affects public health. Thus, the present investigation was done in two steps; the first step was to examine the prevalence and the antibiogram of P. aeruginosa among Nile tilapia (Oreochromas niloticus (O. niloticus)) from aquaculture farms in Kafr El-shiekh Governorate with an emphasis on their antibiotic resistance genes (BlaTEM, tetA, and sul1). The second step was to investigate the effect of levamis

Background/Objectives: The spread of antimicrobial resistance (AMR) is a growing global health concern. Wild animals can play an important role in the amplification and dissemination of AMR and in conservation efforts aiming at controlling diseases in vulnerable wild animal populations. These animals can serve as reservoirs for antibiotic resistance genes and are key in the spread of AMR across ecosystems and hosts. Therefore, monitoring AMR in wild animals is crucial in tackling the spread of resistance in the environment and human population. This study investigated the phenotypic and genoty

BACKGROUND: The rise of carbapenem-resistant hypervirulent Klebsiella pneumoniae (CR-hvKp) strains represents a major clinical threat, particularly in Asia. These strains possess both extensive drug resistance and enhanced virulence traits, leading to high morbidity and mortality. This study aimed to characterize the genomic profile of a CR-hvKp isolate (BB-7) obtained from a clinical case in eastern India and to investigate its resistance and virulence mechanisms.
METHODS: K. pneumoniae BB-7 was obtained from the urine sample of a 75-year-old male and identified by using the VITEK2 system an

UNLABELLED: Many multidrug-resistant (MDR) bacteria have evolved through the accumulation of antibiotic resistance genes (ARGs). Although the potential risk of probiotics as reservoirs of ARGs has been recognized, strategies for blocking the transfer of ARGs while using probiotics have rarely been explored. The probiotic Escherichia coli Nissle 1917 (EcN) has long been used for treating intestinal diseases. Here, we demonstrate frequent transfer of ARGs into EcN both in vitro and in vivo, raising concerns about its potential risk of accumulating antibiotic resistance. Given that no CRISPR-Cas

BACKGROUND: Infant gut microbiota is highly malleable, but the long-term longitudinal impact of antibiotic exposure in early life, together with the mode of delivery on infant gut microbiota and resistome, is not extensively studied.
METHODS: Two hundred and eight samples from 45 infants collected from birth until 2 years of age over five time points (week 1, 4, 8, 24, year 2) were analysed. Based on shotgun metagenomics, the gut microbial composition and resistome profile were compared in the early life of infants divided into three groups: vaginal delivery/no-antibiotic in the first 4 days

Studies on antimicrobial resistance (AMR) profiles and epidemiological affirmation for AMR transmission are limited in fisheries and aquaculture settings. Since 2015, based on Global Action Plan on AMR by World Health Organization (WHO) and World Organization for Animal Health (OIE), several initiatives have been under taken to enhance the knowledge, skills and capacity to establish AMR trends through surveillance and strengthening of epidemiological evidence. The focus of this study was to determine the prevalence of antimicrobial resistance (AMR), its resistance profiles and molecular charac
