Maladaptive decision making and substance use outcomes in high-risk individuals: preliminary evidence for the role of 5-HTTLPR variation.

OBJECTIVE: Individuals with multiple alcohol-dependent (AD) relatives are at increased risk for substance use disorders (SUDs). Prospective, longitudinal studies of high-risk (HR) individuals afford the opportunity to determine potential risk markers of SUDs. The current study assessed the effect of familial risk and genetic variation on Iowa Gambling Task (IGT) performance and tested for an association between IGT performance and SUD outcomes. METHOD: Individuals from multiplex AD families (n = 63) and low-risk (LR; n = 45) control families, ages 16-34 years, were tested using a computerized

Time-based and event-based prospective memory in autism spectrum disorder: the roles of executive function and theory of mind, and time-estimation.

Prospective memory (remembering to carry out an action in the future) has been studied relatively little in ASD. We explored time-based (carry out an action at a pre-specified time) and event-based (carry out an action upon the occurrence of a pre-specified event) prospective memory, as well as possible cognitive correlates, among 21 intellectually high-functioning children with ASD, and 21 age- and IQ-matched neurotypical comparison children. We found impaired time-based, but undiminished event-based, prospective memory among children with ASD. In the ASD group, time-based prospective memory

Life With Cognitive-Communication Deficits After the NFL: A First-Person Narrative.

PURPOSE: This article presents a co-authored, first-person narrative that offers an intimate look at the long-term cognitive-communication consequences of repetitive head impacts sustained during a professional football career. Through the lived experiences of former National Football League cornerback D.J. Johnson, and with light annotation from peer-reviewed literature, the narrative illustrates how concussion and traumatic brain injury can affect memory, speech, and everyday participation years after retirement. KEY INSIGHTS: Johnson describes progressive word-finding difficulty, slowed an

Chronic cannabidiol administration modulates depressive and cognitive alterations induced by social isolation in male mice.

Cannabidiol (CBD), a non-psychotropic compound derived from Cannabis sativa, is known for its potential therapeutic effects on central nervous system (CNS) disorders. This study investigates the effects of chronic CBD administration on depressive and cognitive alterations induced by social isolation in male C57BL/6 mice. The experimental design involved adult mice subjected to either group housing or 12 weeks of social isolation. Behavioral assessments, including the sucrose preference test, open field test, light/dark box, novel object recognition, and tail suspension test, were performed to

Dynamics and immunomodulation of cognitive deficits and behavioral changes in non-severe experimental malaria.

Data recently reported by our group indicate that stimulation with a pool of immunogens capable of eliciting type 2 immune responses can restore the cognitive and behavioral dysfunctions recorded after a single episode of non-severe rodent malaria caused by Plasmodium berghei ANKA. Here we explored the hypothesis that isolated immunization with one of the type 2 immune response-inducing immunogens, the human diphtheria-tetanus (dT) vaccine, may revert damages associated with malaria. To investigate this possibility, we studied the dynamics of cognitive deficits and anxiety-like phenotype follo

The role of cerebral microbleeds in the incidence of post-stroke dementia.

Microbleeds are a marker of cerebrovascular disease however its role in incident post-stroke dementia (PSD) remains unclear. We investigated whether microbleeds are associated with incident PSD, domain-specific cognitive impairment and cognitive decline over a 2-year follow-up; and whether microbleeds interact with acute stroke-related infarcts to synergistically affect cognitive outcomes. In a cohort of patients with first-episode mild ischemic stroke and no pre-stroke dementia, we found patients with 3 or more mixed microbleeds (presence of both lobar and deep) were 4 times more at risk of d

Neurocognitive profile of outpatients with unipolar depressive disorders.

Introduction: It is widely accepted that major depressive disorder (MDD) is associated with impairments in several cognitive domains in inpatients, but cognitive functioning has barely been systematically investigated in depressed outpatients. The aims of the present study were to examine cognitive functioning in depressed outpatients in comparison with healthy controls and to determine whether cognitive deficits vary as a function of different disorder characteristics and demographic variables. Method: We compared 103 outpatients with recurrent MDD in an acute phase of the illness with 103 he

Forebrain-specific ablation of phospholipase Cγ1 causes manic-like behavior.

Manic episodes are one of the major diagnostic symptoms in a spectrum of neuropsychiatric disorders that include schizophrenia, obsessive-compulsive disorder and bipolar disorder (BD). Despite a possible association between BD and the gene encoding phospholipase Cγ1 (PLCG1), its etiological basis remains unclear. Here, we report that mice lacking phospholipase Cγ1 (PLCγ1) in the forebrain (Plcg1f/f; CaMKII) exhibit hyperactivity, decreased anxiety-like behavior, reduced depressive-related behavior, hyperhedonia, hyperphagia, impaired learning and memory and exaggerated startle responses. Child sexual abuse and the development of psychiatric disorders: a neurobiological trajectory of pathogenesis.

Child sexual abuse (CSA) is an important public health problem with long-standing neurobiological, developmental, and psychiatric abnormalities. The present review analyzes the long-term effects of CSA from a developmental, psychiatric morbidity, neurochemical and neurobiological perspective and then tries to posit a developmental neurobiological trajectory from CSA to the genesis of psychopathology in later life. The role of various neurotransmitters such as serotonin and dopamine affected by CSA are discussed. Serotonin abnormalities have been reported in various studies among participants e

An attentional scope model of rumination.

Rumination, defined as repetitive thinking about negative information, has been found to lead to serious maladaptive consequences, including longer and more severe episodes of major depression. In this review, we present and discuss research findings motivated by the formulation that individual differences in cognitive processes that control how information is processed influence the likelihood that thoughts will become repetitive and negative. Several studies have demonstrated that a tendency to ruminate (i.e., trait rumination) is related to difficulties updating working memory (WM) and dise

Neural markers of negative symptom outcomes in distributed working memory brain activity of antipsychotic-naive schizophrenia patients.

Since working memory deficits in schizophrenia have been linked to negative symptoms, we tested whether features of the one could predict the treatment outcome in the other. Specifically, we hypothesized that working memory-related functional connectivity at pre-treatment can predict improvement of negative symptoms in antipsychotic-treated patients. Fourteen antipsychotic-naive patients with first-episode schizophrenia were clinically assessed before and after 7 months of quetiapine monotherapy. At baseline, patients underwent functional magnetic resonance imaging while performing a verbal n-

Forensic significance of the limbic psychotic trigger reaction.

During the "decade of the brain," competent expert testimony should encompass widely neglected, even novel, neurophysiologically plausible explanations for otherwise unexplainable acts. In the case presented here, a sudden, out-of-character, motiveless, unplanned homicidal attack was committed by a patient who demonstrated flat affect, preserved consciousness, and memory of the episode. Transient autonomic hyperactivation and psychosis were suddenly experienced when the victim happened to move his mouth while eating. Following a sudden memory revival of repetitive but moderate bodily stresses,

Hepatico-jejuno-duodenostomy reconstruction following excision of choledochal cysts in children.

Ten children with choledochal cysts were treated at Children's Memorial Hospital in Chicago between 1979 and 1980 and at Henrietta Egleston Hospital for Children or Grady Memorial Hospital in Atlanta between 1980 and 1987. Age at the time of operation ranged from one to 46 months. There were eight girls and two boys with Type I choledochal cysts. One child with a dilated but histologically normal common bile duct underwent a choledochoduodenostomy. The remaining nine patients had excision of the choledochal cyst. Continuity with the GI tract was then re-established with a valved isolated jejun

Association of early-life caregiver mental health and education with incidence of mild cognitive impairment among the Chinese population: A longitudinal observational study.

BACKGROUND: Mild cognitive impairment (MCI) is a critical prodromal stage of dementia, yet the role of early-life psychosocial determinants, particularly caregiver characteristics, remains underexplored. OBJECTIVE: To investigate the associations of caregiver mental health and education with offspring MCI incidence, and to examine their joint effects. METHODS: We utilized data from the China Health and Retirement Longitudinal Study (CHARLS), involving 8140 participants aged ≥45 years. The mental health (categorized as normal or abnormal) and educational level (categorized as illiterate, basi

Trace elements and cognition in the general population: The HUNT study.

BACKGROUND: Trace elements play essential roles in neurobiology, but evidence linking whole blood concentrations to cognitive outcomes in the general population remains limited. This study examined whether concentrations of eight trace elements: selenium, mercury, cadmium, arsenic, lead, manganese, copper, and zinc were associated with cognitive performance assessed approximately six years later in a Norwegian general population cohort from the HUNT3 Survey. METHODS: Whole blood trace element concentrations were measured using high resolution inductively coupled plasma mass spectrometry, with

Intellectual disability and structural defects of the CaV2.1 channel in episodic ataxia type 2: correlation using an AI prediction model.

BACKGROUND: Episodic ataxia type 2 (EA2) results from pathogenic variants in CACNA1A that encodes the CaV2.1 P/Q-type calcium channel. The molecular basis of cognitive impairments requires further elucidation in EA2. OBJECTIVE: To correlate AI-predicted structural alterations of the CaV2.1 channel with intellectual function observed in patients with EA2. METHODS: Using AlphaFold3, we modeled the wild-type and variant CACNA1A proteins. Structural similarity between the wild-type and variant proteins was quantified using the Template Modeling (TM) score. To assess degree of truncation, the rel

The impact of a 12-month randomized exercise intervention on cognitive function and brain volume in adults with down syndrome.

BACKGROUND: Adults with Down syndrome (DS) have a high risk for Alzheimer's disease (AD). While exercise improves cognition and brain health in the general population, few long-term studies have evaluated its effects in DS. OBJECTIVE: We examined the impact of a 12-month remotely delivered exercise program on cognitive function and brain volume in adults with DS. METHODS: 81 adults with DS (mean age = 27 years) were randomized to high-frequency remote exercise (3 sessions/week, RH), low-frequency remote exercise (1 session/week, RL), or a support and education control (SE).
