Features in essential tremor and the development of Parkinson's disease vs. parkinsonism.

BACKGROUND: Patients with essential tremor (ET) are at higher risk to develop Parkinson's disease (PD). Recent studies suggest that propranolol (common treatment for ET) can augment pathologic expression of alpha-synuclein. We studied features associated with the development of Parkinson's disease with antecedant essential tremor (ET-PD) compared with ET-plus with parkinsonism (PK). DESIGN: Retrospective case series from a tertiary movement disorders center including patients with ET and PD, found to have ET-PD or ET-plus (PK). RESULTS: We analyzed two groups: (1) ET-plus (PK) (n = 33) and (…

Cell type transcriptomic modules reveal shared molecular mechanisms in Alzheimer's and Parkinson's disease.

BACKGROUND: Historically, Alzheimer's disease (AD) and Parkinson's disease (PD) have been investigated as 2 distinct disorders of the brain. However, a few similarities in neuropathology and clinical symptoms have been documented over the years. Traditional single-gene centric studies, such as differential gene expression analyses, have struggled to unravel the molecular basis for the observed pathological links between AD and PD. RESULTS: We tailor a latent factor framework to analyze synchronous gene co-expression changes in AD or PD at sub-cell-type resolution. Utilizing large, single-nucl

The Interplay between Intracellular Iron Homeostasis and Neuroinflammation in Neurodegenerative Diseases.

Iron is essential for life. Many enzymes require iron for appropriate function. However, dysregulation of intracellular iron homeostasis pro … onse by producing amyloid-beta (Aβ) fibrils and neurofibrillary tangles in Alzheimer's disease, and alpha-synuclein aggregation in Parkinson's disease. This interplay shows that intracellular iron homeostasis is vital to maintain inflammatory homeostasis. Here, we review the role of iron homeostasis in inflammation based on recent findings.

Sex-dependent alterations in motor and anxiety-like behavior of aged bacterial peptidoglycan sensing molecule 2 knockout mice.

Peptidoglycan recognition proteins (PGRPs) are key sensing-molecules of the innate immune system that specifically detect bacterial peptidog … ot in the amygdala. Pglyrp2 KO female mice exhibited increased expression levels of spinophilin and alpha-synuclein in the frontal cortex, while exhibiting decreased expression levels of synaptophysin, gephyrin and spinophilin in the amygdala. Our findings suggest a novel role for Pglyrp2asa key regulator of motor and anxiety-like behavior in late life.

Imaging nanoscopic co-assemblies of α-synuclein and Tau in human samples.

α-synuclein (αS), amyloid-β (Aβ), and Tau aggregates are hallmarks of many neurodegenerative diseases, yet the prevalence and nature of their nanoscopic co-assemblies remain poorly understood. Here, we adapt a single-molecule pull-down (SiMPull) assay to profile αS, Aβ, and Tau co-assemblies across post-mortem brain tissue homogenates, soluble brain fractions, human serum, cerebrospinal fluid, and saliva. We show that αS-Tau co-assemblies are abundant across all sample matrices, accounting for ~10-70% of total αS aggregates in both Parkinson's disease (PD) and control samples. Notably, αS-Tau

Imbalance in excitation and inhibition in sgACC is associated with depression in dementia with Lewy bodies.

Depression in dementia with Lewy bodies (DLB) is a common neuropsychiatric symptom associated with reduced quality in life. Structural, functional and neurochemical abnormalities in glutamatergic and GABAergic neurotransmission are implicated in the pathophysiology of depression, showing changes in regions involved in emotional processing, including the subgenual cingulate cortex (sgACC), which shows pathological changes in DLB. Using post-mortem tissue from DLB patients and controls, we assessed synaptic and neurochemical changes within the sgACC in relation to depression in DLB. We identifie

Angiotensin II-induced hypertension does not exacerbate α-SYNUCLEIN pathology in a mouse model of Parkinson's disease.

Parkinson's disease (PD) is characterized by progressive degeneration of nigrostriatal dopaminergic neurons accompanied by chronic neuroinflammation and α-SYNUCLEIN (α-SYN) aggregation. Increasing evidence suggests that cardiovascular comorbidities, including arterial hypertension, may contribute to neurodegenerative processes through vascular dysfunction and sustained inflammatory signaling. However, whether hypertension directly exacerbates α-SYN-induced neurodegeneration and inflammasome activation remains unclear. Here, we investigated the impact of chronic Angiotensin II (AngII)-induced h

Schedule-dependent neuroprotection by pioglitazone in a novel model of α-synucleinopathy in rats: Integrated behavioural and histological outcomes.

BACKGROUND AND PURPOSE: α-Synucleinopathies are neurodegenerative disorders characterized by the aggregation and propagation of misfolded α-synuclein. In Parkinson's disease (PD), the most common α-synucleinopathy, the progression of motor and nonmotor deficits, and dopaminergic neuron loss, are closely linked to the spreading of misfolded α-synuclein. Pioglitazone, a PPARγ agonist, has shown neuroprotective effects in preclinical models. However, its dosing and its effects on behaviour and neuropathology have not been explored in a model of α-synucleinopathy. Here, we evaluate the safety and

Calcium overload induced mitochondrial and lysosomal dysfunction is regulated by Tousled-like kinase in a-synucleinopathy.

As a pathological hallmark of Parkinson's disease (PD), a-synucleinopathy induces various cellular damages, including calcium overload, mitochondrial and autophagic dysfunction, ultimately resulting in dopaminergic neuron death. However, the hierarchy of these detrimental events remains unclear. It is well established that a-synuclein can induce calcium overload through diverse mechanisms. To assess whether calcium overload plays a crucial detrimental role, we established a calcium overload model in Drosophila and conducted genetic screening. Our findings indicate that calcium overload caused

Glycerol 3-phosphate acyltransferase exacerbates α-synuclein-induced toxicity by increasing lipid peroxidation.

Although multiple cellular pathways have been implicated in α-Synuclein (α-syn)-associated Parkinson's disease (PD), the role of lipid metabolism remains elusive. In this study, we identify Drosophila mino, which encodes the mitochondrial isoform of the lipid synthesis enzyme glycerol 3-phosphate acyltransferase (GPAT), as a potent modifier of α-syn. Silencing the expression of mino significantly suppresses α-syn-induced PD phenotypes in Drosophila, including dopaminergic neuronal loss and locomotion defects as well as circadian rhythm-related activities, whereas mino overexpression yields opp

Ulcerative colitis model triggers gut α-Synuclein aggregation without brain involvement or neuronal loss in female rats.

INTRODUCTION: Despite being the second most common neurodegenerative disorder, the mechanisms underlying the onset and progression of Parkinson's disease (PD) remain poorly understood, and no curative treatment is currently available. The Braak hypothesis offers an intriguing framework for explaining both the origin and development of the disease, proposing that PD begins in the gut and subsequently spreads to the brain. METHODS: In previous studies, our group developed a novel PD model in which peripheral inflammation, triggered by administering dextran sodium sulphate (DSS) in the drinking

TMEM106B deficiency exacerbates α-synuclein aggregation in Parkinson's disease.

Parkinson's disease is an age-related neurodegenerative disease that is characterized by the deposition of α-synuclein aggregates in the brain. Nevertheless, the molecular mechanisms that regulate α-synuclein aggregation have not yet been identified fully. TMEM106B is a lysosomal transmembrane protein that has been reported to be associated with brain ageing and neurodegenerative diseases, including Parkinson's disease. Here we show that TMEM106B is reduced in the brains of patients with Parkinson's disease. Knockout of Tmem106b increases the formation of α-synuclein aggregates in primary neur

Plasma cfDNA hypermethylation at SNCA intron 1 as a potential blood-based epigenetic signal in Parkinson's disease and multiple system atrophy.

BACKGROUND: The accumulation of α-synuclein (SNCA) in the central nervous system is a hallmark of Parkinson's disease (PD) and multiple system atrophy (MSA). SNCA intron 1 methylation is implicated in SNCA transcriptional regulation and may serve as a peripheral epigenetic signal in synucleinopathies. However, studies of SNCA methylation in leukocyte-derived DNA have yielded inconsistent results. We aimed to evaluate whether cell-free DNA (cfDNA)-based SNCA intron 1 methylation differs in PD or MSA compared with normal controls (NC). METHODS: Plasma cfDNA was collected from 105 patients with

Highly sensitive chemiluminescence imaging of misfolded proteins in neurodegenerative models.

Protein misfolding in the brain is a key pathological hallmark of neurodegenerative diseases. Optical imaging of misfolded proteins in disease models is essential for elucidating etiology and early diagnosis. However, developing specific optical imaging probes for each misfolded protein is time-consuming and challenging, leaving many pathological targets without effective detection tools, especially for in vivo imaging. Here, we present a dual-mode chemiluminescence strategy that enables both generic and specific detection of misfolded proteins using a single probe platform. In the generic mod

Ten-Year Retrospective Study on the Diagnostic Value of Three-Site Skin Biopsy for Detecting pS129-α-Synuclein Biomarker: Cervical and Leg Dual-Site Biopsy as an Effective and Less Invasive Alternative.

BACKGROUND: The in vivo detection of phosphorylated-α-synuclein (pS129-α-syn) via immunofluorescence in cutaneous nerve fibers has emerged as a promising biomarker for diagnosing synucleinopathies, including Parkinson's disease, dementia with Lewy bodies and multiple system atrophy (MSA). However, the variability in biopsy protocols, particularly regarding the choice and number of anatomical sites, limits standardisation and clinical applicability. OBJECTIVE: To evaluate the diagnostic accuracy of different biopsy site combinations for pS129-α-syn detection in a large cohort of patients with

Investigation of correlation between cholesterol intake, apolipoprotein B and Parkinson's disease related genes in guinea pigs feeding a high-fat diet containing cholesterol.

Apolipoprotein B (Apo B), which is involved in the transport of cholesterol, is thought to be associated with neurodegenerative diseases such as Parkinson's disease in addition to atherosclerosis and cardiovascular diseases. We aimed to investigate the possible correlation between cholesterol intake, Apo B and parkin RING domain-containing E3 ubiquitin protein ligase (PARKIN), phosphatase and tensin homologue (PTEN)-induced kinase 1 (PINK1) and α-synuclein (SNCA), which have an important role in Parkinson's disease.
