Mass spectrometric analysis of L-cysteine metabolism: physiological role and fate of L-cysteine in the enteric protozoan parasite Entamoeba histolytica.

UNLABELLED: L-cysteine is essential for virtually all living organisms, from bacteria to higher eukaryotes. Besides having a role in the syn … -thiazolidine-4-carboxylic acids are involved in the defense against oxidative stress. IMPORTANCE: Amebiasis is a human parasitic disease caused by the protozoan parasite Entamoeba histolytica. In this parasite, L-cysteine is the principal low-molecular-weight thiol and is assumed to play a significant role in supplying the amino acid during trophozoite in …

Ultrastructural characterization and pathogenicity of Allovohlkamfia spelaea in a murine model: Neuropulmonary infections and therapeutic potential of ellagic acid.

BACKGROUND: Allovahlkampfia spelaea (A. spelaea) is a free-living amoeba that has recently been recognized to cause Acanthamoeba-like keratitis, the treatment of which is complex. The pathogenic potential of Allovahlkampfia spp. remains unexplored. This study characterized A. spelaea through ultrastructural morphological analysis and investigated the pathogenic potential of the A. spelaea strain KS1, which was isolated from a patient with keratitis, in a murine model, with a focus on neuro-pulmonary infections. Additionally, this study assessed the therapeutic effectiveness of ellagic acid (EA

Type 2 diabetes mellitus BALB/c mice are more susceptible to granulomatous amoebic encephalitis: Immunohistochemical study.

Granulomatous amoebic encephalitis (GAE) is a chronic, difficult to resolve infection caused by amphizoic amoebae of the genus Acanthamoeba, which in most cases occurs in immunosuppressed persons or with chronic diseases such as diabetes. In this study, we describe the early events of A. culbertsoni infection of GAE in diabetic mice model. Diabetes was induced in male BALB/c mice, with a dose of streptozotocin (130 mg/kg). Healthy and diabetic mice were inoculated via intranasal with 1 × 106 trophozoites of A. culbertsoni. Then were sacrificed and fixed by perfusion at 24, 48, 72 and 96 h post

Immune responses against protozoan parasites: a focus on the emerging role of Nod-like receptors.

Nod-like receptors (NLRs) have gained attention in recent years because of the ability of some family members to assemble into a multimeric protein complex known as the inflammasome. The role of NLRs and the inflammasome in regulating innate immunity against bacterial pathogens has been well studied. However, recent studies show that NLRs and inflammasomes also play a role during infections caused by protozoan parasites, which pose a significant global health burden. Herein, we review the diseases caused by the most common protozoan parasites in the world and discuss the roles of NLRs and infl

Thermophilic potentially pathogenic amoebae isolated from natural water bodies in Poland and their molecular characterization.

The free-living amoebae (FLA) may live in the environment and also within other organisms as parasites and then they are called amphizoic. They are potentially pathogenic for humans and animals and are found in water that is a source of infection. The aim of this study was molecular detection and identification of these FLA in natural water bodies in North-Western Poland to evaluate the risk of the pathogenic amoebae infections. We examined surface water samples collected from 50 sites and first, the tolerance thermic test was performed in order to select thermophilic, potentially pathogenic s

Primary amoebic meningoencephalitis due to Naegleria fowleri.

Primary amoebic meningoencephalitis (PAM) due to Naegleria fowleri was detected in a 36-year-old, Indian countryman who had a history of taking bath in the village pond. He was admitted in a semi comatosed condition with severe frontal headache, neck stiffness, intermittent fever, nausea, vomiting, left hemiparesis and seizures. Computerized tomography (CT) scan of brain showed a soft tissue non-enhancing mass with erosion of sphenoid sinus. However CSF findings showed no fungal or bacterial pathogen. Trophozoites of Naegleria fowleri were detected in the direct microscopic examination of CSF

Fatal infections with Balamuthia mandrillaris (a free-living amoeba) in gorillas and other Old World primates.

Balamuthia mandrillaris is a newly described free-living amoeba capable of causing fatal meningoencephalitis in humans and animals. Because the number of human cases is rapidly increasing, this infection is now considered an important emerging disease by the medical community. A retrospective review of the pathology database for the Zoological Society of San Diego (the San Diego Zoo and San Diego Wild Animal Park) for the period July 1965 through December 1994 revealed five cases of amoebic meningoencephalitis, all in Old World primates. The infected animals were a 3-year, 10-month-old female

Medical problems of foreign-born adopted children.

Over 8000 foreign-born children, almost all from third-world countries, are adopted by citizens of the United States each year. Most primary care practitioners do not have enough experience to evaluate and manage their problems efficiently. We reviewed the medical care of the foreign-born adoptees followed up in a single pediatric group practice. The 128 children arrived from one of eight countries in Asia or Latin America at ages ranging from 1 month to 10 years; 57% were female. The median duration of follow-up was 20 months. The most common problems identified included deficient immunizatio

Seroprevalence of Naegleria fowleri in the Houston-Galveston Texas Population.

Naegleria fowleri is a free-living amoeba and the causative agent of Primary Amebic Meningoencephalitis (PAM), a rare yet almost always fatal disease that primarily affects children. While only 431 PAM cases have been reported worldwide, the southern states of the United States, including Texas, report the highest number of cases. Despite the high mortality of the disease, studies have identified antibodies to N. fowleri in healthy individuals suggesting that exposure to this pathogen is common, but disease is rare. Here, we investigated the seroprevalence of N. fowleri in the general populati
