Postnatal ethanol exposure blunts upregulation of GABAA receptor currents in Purkinje neurons.

Recently, we found that early postnatal ethanol exposure inhibits the maturation of GABAA receptors (GABAARs) in developing medial septum/diagonal band (MS/DB) neurons, suggesting that these receptors may represent a target for ethanol related to fetal alcohol syndrome (FAS). To determine whether GABAARs on other neurons are also sensitive to a postnatal ethanol insult, postnatal day (PD) 4-9, rat pups were artificially reared and exposed to ethanol (4.5 g kg-1 day-1, 10.2% v/v). The pharmacological profile of acutely dissociated cerebellar Purkinje cell GABAARs from untreated, artificially re

Defective intracellular processing of lactase-phlorizin hydrolase protein in rats prenatally exposed to ethanol.

We have previously shown that fetal exposure to ethanol in rats produces both structural and biochemical abnormalities in absorptive enterocytes. Among the indicators of injury are derangements in the expression of lactase-phlorizin hydrolase (LPH), which is an essential enzyme for the assimilation of milk. In an animal model of fetal alcohol syndrome, unsuckled newborn rats prenatally exposed to maternal ethanol revealed a 10- to 15-fold increase in the number of LPH mRNA molecules per absorptive enterocyte, compared with controls (Estrada et al., Alcohol. Clin. Exp. Res. 20:1662-1668, 1996).

Optic nerve hypoplasia in the fetal alcohol syndrome: a mouse model.

Optic nerve hypoplasia is commonly observed in children affected by the fetal alcohol syndrome, and is believed to contribute to their poor visual acuity. We have used a 'binge' model of alcohol abuse in an attempt to recreate this hypoplasia in a mouse model. Pregnant female (C57BL/6 x CBA)F1 mice were injected intraperitoneally with a single dose of a 25% solution of ethanol (v:w), either on d 11 or d 12 of gestation. Optic nerves were prepared for transmission electron microscopy from offspring at 3, 6, 9 and 15 wk of age (n = 64). A systematic random sampling technique was used to analyse

In utero ethanol exposure retards growth and alters morphology of cortical cultures: GM1 reverses effects.

Ethanol, a developmental neurotoxin, alters plasma membranes' physicochemical properties affecting embryogenesis, cell migration, differentiation, and synaptogenesis. In a previous study using a model for fetal alcohol effects, GM1 ganglioside treatment was shown to reduce ethanol-induced accumulation of endogenous GM1 and fatty acid ethyl esters in rat fetuses. The present study was initiated to define further the in utero effects of ethanol and the capacity of GM1 treatment to ameliorate such effects. Wistar dams were exposed to ethanol (intragastrically) on gestation day (GD) 7 and GD8 and

Alaska/Russian Far East behavioral health projects: a progress report on recent activities.

A number of "behavioral health" exchange activities between Alaska and the Russian Far East took place during 1992-93. These included visits by a psychiatrist running one of the first "private polyclinics" in Magadan and by a narcologist to discuss the latest developments in alcohol services; a Behavioral Health Conference where Native health workers from Russia and Alaska shared experiences; beginning a multi-year epidemiology project; and a project to assess needs for a fetal alcohol syndrome (FAS) prevention program in Magadan.

Treatment of pregnant alcohol-consuming rats with buspirone: effects on serotonin and 5-hydroxyindoleacetic acid content in offspring.

This laboratory previously demonstrated that in utero ethanol exposure markedly impairs the development of the serotonergic system in rat brain. Developmental abnormalities could be detected as early as G15 in the brainstem and G19 in the cortex. Because of the importance of fetal serotonin (5-HT) and 5-HT1A receptors for the normal development of 5-HT containing neurons, we initiated studies to determine whether administration of a 5-HT1A agonist, buspirone, to pregnant rats could overcome the adverse effects of in utero ethanol exposure on the developing serotonergic system in offspring. A fetal alcohol syndrome surveillance pilot project in American Indian communities in the Northern Plains.

A pilot fetal alcohol syndrome (FAS) surveillance was carried out in four American Indian communities in the Northern Plains by the Aberdeen Area Indian Health Service to determine the incidence of FAS and to evaluate the feasibility of establishing continuing surveillance for FAS. Baseline data on the incidence of FAS would be used by the Indian Health Service to develop and evaluate preventive interventions, including treatment programs for pregnant women who drink alcohol. Four of the 1,022 children included in the project were found to have FAS, a rate of 3.9 per 1,000 live births. The rat

Short- and long-term effects of combined pre- and postnatal ethanol exposure (three trimester equivalency) on the development of myelin and axons in rat optic nerve.

This study evaluated the effects of a combined gestational and 10 day postnatal alcohol exposure (human three trimester equivalency) on the development of myelin and axons in rat optic nerve. Rats were exposed during gestation via liquid diet, then their artificially reared pups were further exposed for 10 postnatal days via an ethanol-containing diet fed by gastrostomy. Control animals from pair-fed dams were artificially reared for 10 days on pair-fed isocaloric diets. Anesthetized animals were perfused with fixative on gestational days (G) 15 and 20 and postnatal days (P) 5, 10, 15, 20, and

Alcohol exposure and undernutrition: effects on lipid metabolism and alcohol partitioning in rat brain regions in vitro.

The effects of maternal alcohol consumption and undernutrition on lipid metabolism and alcohol partitioning in brain cortical and stem slices of pups were studied under in vitro conditions. Alcohol administration along with a normal diet during gestation and lactation resulted in an increase in the synthesis of cholesterol in cortical and stem slices associated with decreased entry of alcohol. Phospholipid metabolism was not affected in the cortex, whereas the incorporation of [32P] was found to be altered in the brain stem, indicating regional differences with respect to alcohol effects. Blood alcohol concentration and microencephaly: a dose-response study in the neonatal rat.

The relationships among microencephaly, peak blood alcohol concentration (BAC), and dose of alcohol were examined in a rat model of third-trimester fetal alcohol effects. Ethyl alcohol was administered to neonatal rats from postnatal day 4 to day 10 during the brain growth spurt via an artificial rearing technique. Groups of rats received one of nine doses of alcohol (0.0, 2.5, 3.3, 4.0, 4.5, 5.3, 6.6, 7.5, or 8.5 g/kg body weight) administered in 8 hours each day. BACs were determined on postnatal days 6 and 7 at times corresponding to peak and trough BACs, respectively. On postnatal day 10,

Follow-up studies of children with fetal alcohol syndrome.

Data coming from a prospective multidisciplinary study with repeated examinations of children with fetal alcohol syndrome (FAS) are reported. These patients underwent pediatric, neurological and psychiatric assessment, EEG-recordings and psychological testing. After a period of 3-4 years various subgroups of these children were re-examined. Follow-up examinations revealed that with increasing age dysmorphic signs became less apparent in children with FAS. Furthermore, neurologic performance improved and EEG-recordings revealed less pathological patterns. These positive findings were confirmed

Effect of ethanol on human umbilical artery and vein in vitro.

In vitro effects of ethanol on human umbilical artery and vein were studied. Ethanol (0.5-2.0%) elicited a dose-dependent contractile response both in the umbilical arterial and venous strips in Krebs' and Ca2+-free Krebs' solution. In umbilical artery, but not in vein, ethanol shifted the CaCl2-induced dose-response curves to the right in a parallel manner along with a significant reduction in K+-depolarized maximal tension. These result demonstrate that ethanol can elicit a potent contractile response both in human umbilical arteries and veins and could aid in explaining why ethanol can indu

Maternal age and congenital optic nerve hypoplasia: a possible clue to etiology.

The maternal age of two groups of children with congenital handicaps, cerebral palsy and fetal alcohol syndrome (FAS), were examined and compared with a group of mothers of children with congenital optic-nerve hypoplasia (CONH). The maternal age of children with CONH was significantly less than for the cerebral-palsied children which, in turn, was significantly less than for the FAS children. In addition, about one-half of the children with CONH had associated handicaps which clustered into four progressively more severe combinations. Maternal age and the occurrence of endocrine problems did n

Effects of gestational alcohol on the development of neonatal reflexes in the rat.

The offspring of rats fed a diet containing 10% (v/v) of ethanol were tested for developmental differences from controls on a variety of maturational events and reflexes. Results indicated slower development of fur onset and ear (pinnae) opening, more rapid and vigorous righting responses and shorter dowel-grasping times in the offspring of ethanol-fed dams. No differences in rooting reflex, Babinski reflex, or in the visual placing response were found. These findings were seen as consistent with the interpretation that gestational alcohol exposure results in reduction in the ability of subjec

A rat model of the fetal alcohol syndrome--preliminary histological findings.

A semipurified liquid diet was used to develop a chronic rat model of fetal alcohol syndrome. Adult female rats were fed a control or 40% ethanol (percent of total caloric intake) liquid diet. Additional controls were pair fed to the 40% experimental group. Results of the effects of these diets on the offspring included both qualitative and quantitative changes observed in Golgi study of hippocampal pyramidal neurons.

Using administrative data to examine variables affecting the mental health of siblings of children who have a developmental disability.

BACKGROUND: There is evidence that siblings of children with a developmental disability (DD) experience poorer mental health outcomes and increased stress compared to siblings of non-disabled children. The variables which contribute to this are unclear. AIMS: This study was designed to examine population-level and individual variables associated with differences in mental health outcomes among siblings of children who have a variety of developmental disabilities. METHODS: Population-level administrative health data covering 1985-2014 for the province of British Columbia, Canada were used to
