Estimation of the effect of the acidosis and alkalosis on the anesthetic potency of local anesthetics by biopartitioning micellar chromatography and micellar electrokinetic chromatography.

Local anesthetics are hydrophobic compounds and weak bases with protonation constants ranged between 7.5 and 8.8. These drugs block reversibly nerve conduction near their site of application or injection and thus produce temporary loss of feeling or sensation in a limited area of the body. The efficacy of anesthetic blockade of local anesthetics depends on the charged/uncharged form ratio and the hydrophobicity of the compounds. In addition their toxicological effects have been reported to be highly dependent on the physiological pH. Biopartitioning micellar chromatography (BMC) and micellar e

Bacterial nanocellulose as a drug delivery system for local anesthetics: A novel approach for non-invasive pain management.

This study presents the development of bacterial nanocellulose (BNC) membranes derived from "Nata de fique" for local anesthesia applications. The membranes were successfully produced using fique juice as a culture medium and exhibited a characteristic nanofibrillar structure. After a purification process with KOH, they displayed a neutral pH and a high moisture content of 98.12 ± 0.54%. Following mechanical dehydration, their thickness and weight were reduced, but upon loading with articaine hydrochloride and epinephrine, the membranes recovered 87.35 ± 0.32% of their moisture. Moist Wound Healing and Eutectic Mixture of Local Anesthetics Cream for Clean Buttonhole Cannulation: An Infection Reduction Strategy.

KEY POINTS: In buttonhole cannulation, where scabs are forcibly removed, the infection rate is higher than that in the rope ladder method. The clean buttonhole method enhances scab care by combining moist wound healing, thorough washing, and eutectic mixture of local anesthetics. The buttonhole technique allows patients to maintain healthy skin, except for the two puncture sites. BACKGROUND: The buttonhole cannulation technique is recognized for its advantages in hemodialysis, including reduced cannulation errors, decreased pain, and prevention of hematoma formation. However, it has been crit

Cryosurgery of eyelid actinic keratosis and the effect of adrenaline in local anesthetics - temperature mapping using IR thermography.

PURPOSE: The efficacy of cryosurgery is believed to depend on the thaw time. The presence of adrenaline in local anesthetics affects blood perfusion, and may thus also affect the thaw time. The aim of the present study was to use IR thermography to monitor the tissue temperature during cryosurgery of actinic keratosis, and to assess the effect of adrenaline in local anesthetics. METHODS: Twelve patients with actinic keratosis in the periorbital region underwent cryosurgery. The temperature was continuously mapped using a high-precision IR camera and was also visually assessed by the surgeon.

Hyperspectral and Laser Speckle Contrast Imaging for Monitoring the Effect of Epinephrine in Local Anesthetics in Oculoplastic Surgery.

PURPOSE: Epinephrine is used in local anesthetics to induce vasoconstriction and thus reduce bleeding and prolong the anesthetic effect. Finding the optimal delay between the administration of the anesthetic and skin incision to ensure vasoconstriction and minimize bleeding is important and has recently become the subject of debate. This is the first study to assess blood perfusion and oxygen saturation (sO 2 ) simultaneously in response to a local anesthetic containing epinephrine in human oculoplastic surgery. METHODS: A local anesthetic consisting of lidocaine and epinephrine (20 mg/ml + 1

Dexmedetomidine as an adjuvant to local anesthetics in brachial plexus blocks: A meta-analysis of randomized controlled trials.

BACKGROUND: Brachial plexus block (BPB) for upper extremity surgery provides superior analgesia, but this advantage is limited by the pharmacological duration of local anesthetics. Dexmedetomidine (DEX) as a local anesthetics adjuvant for BPB has been utilized to prolong the duration of the nerve block in some randomized controlled trials (RCTs) but is far from unanimous in the efficacy and safety of the perineural route. Hence, an updated meta-analysis was conducted to assess the efficacy and safety of DEX as local anesthetic adjuvants on BPB. METHODS: A search in electronic databases was co

[Effect of local anesthetics on the transport of mannose in the microsomal membranes of Aspergillus niger van Tieghem].

The influence of various local anaesthetics has been studied on the mannose transfer enzymic system which is localized in Aspergillus niger van Tieghem microsomal membranes. The n-alkanes and tertiary amines do not seem to react on the first step of the reaction: i.e. the polyprenylphosphate-mannose biosynthesis. However two amines, the dibucaine and to a lesser extent the tetracaine, inhibit the mannose transfer from the polyprenylphosphate-mannose to the endogenous proteins.

Reverse arterial blood flow as a pathway for central nervous system toxic responses following injection of local anesthetics.

In baboons, lidocaine HCl was injected into the lingual (3 mg/kg), brachial (7 mg/kg), or femoral (7 mg/kg) arteries. Blood samples were taken from the internal carotid artery (ICA), internal jugular (IJV), external jugular (EJV), brachial (BV), or femoral (FV) vein, depending on the injection site, 6, 30, and 180 seconds after injection. Subsequently, radioactive microspheres (20 micron in diameter) were injected into the lingual artery, and the brains were obtained 48 hours later at postmortem to locate the microspheres. Six seconds after injection into the lingual artery, lidocaine concentr
