Nocturnal sleep architecture disturbances in early methadone treatment patients.

The subjective and objective sleep patterns of patients with opioid dependence have been previously reported, but the sleep characteristics of patients in early methadone treatment, especially the objective sleep patterns, remain largely unexamined. This study was designed to explore the nocturnal sleep structure of patients on early methadone treatment. Twenty male methadone treatment (MT) patients and 20 male age- and body mass index-matched controls were assessed with overnight limited polysomnography. Subjective sleep was assessed using the Pittsburgh Sleep Quality Index (PSQI) and the Epw

Acute severe mitral regurgitation: consideration of papillary muscle architecture.

We present a case of an individual who presented with acute severe mitral regurgitation in the setting of an inferior ST elevation myocardial infarction. Both transthoracic and transesophageal echocardiography demonstrated a posteriorly directed eccentric jet of severe mitral regurgitation with flail anterior mitral valve leaflet attached presumably to the anterior papillary muscle. Intraoperative findings demonstrated rupture of the postero-medial papillary muscle attached via chords to the anterior mitral valve leaflet. This case serves to remind us that both the anterior and posterior leafl

Three-dimensional domain architecture of the ADAM family proteinases.

A disintegrin and metalloproteinase (ADAM) family of proteins constitutes a major class of mammalian membrane-bound sheddases that are responsible for the processing of cell-surface-protein ectodomains, including the latent forms of growth factors, cytokines and their receptors. However, the molecular mechanism by which ADAMs recognize and process their substrates is largely unknown. Recent crystallographic studies on phylogenically related snake venom metalloproteinases (SVMPs) and mammalian ADAM with thrombospondin type-1 motif (ADAMTS) family proteins have shed light on the structure-functi

Competitive Workflow: novel software architecture for automating drug design.

As industrialization of laboratory processes for drug discovery continues to gather momentum, the bottleneck has moved toward exploitation of this tide of information to enable better quality decisions. The development of information-management systems to automate data and materials management can have a positive impact on productivity, as can increasingly sophisticated computer-aided molecular design approaches. However, as long as key decisions can only be taken by a small number of expert individuals working in a complex social environment, the impact of such innovations will be limited. Th

Microanatomical architecture of Dorello's canal and its clinical implications.

OBJECTIVE: We investigated the membranous architecture of the abducens nerve at the petroclival region and describe the characteristics of this area in cadaveric specimen and two children with hydrocephalus and sixth nerve palsy using magnetic resonance imaging (MRI). MATERIALS AND METHODS: Five adult cadaver heads were used to investigate the petroclival part of the abducens nerve. The heads were injected with colored latex for microsurgical dissection, and the length of the dural sleeve of the abducens nerve and its width at the apex were measured. In one cadaver head, the area between the

Novel replication complex architecture in rubella replicon-transfected cells.

Rubella virus (RUB) assembles its replication complexes (RCs) in modified organelles of endo-lysosomal origin, known as cytopathic vacuoles (CPVs). These peculiar structures are key elements of RUB factories, where rough endoplasmic reticulum, mitochondria, and Golgi are recruited. Bicistronic RUB replicons expressing an antibiotic resistance gene either in the presence or the absence of the RUB capsid (C) gene were used to study the structure of RCs in transfected cells. Confocal microscopy showed that the RUB replicase components P90 and P150 localized to CPVs, as did double-stranded RNA (ds

Color architecture in alert macaque cortex revealed by FMRI.

The contribution that different brain areas make to primate color vision, especially in the macaque, is debated. Here we used functional magnetic resonance imaging in the alert macaque, giving a whole brain perspective of color processing in the healthy brain. We identified color-biased and luminance-biased activity and color-afterimage activity. Color-biased activity was found in V1, V2, and parts of V4 and not in V3a, MT, or other dorsal stream areas, in which a luminance bias predominated. Color-biased activity and color-afterimage activity were also found in a region on the posterior bank

Microvascular architecture of the rat choroid: corrosion cast study.

This study presents the details of the microvasculature of the rat choroid visualized by scanning electron microscopy of vascular corrosion casts. Wistar Kyoto rats were anesthetized with intraperitoneal sodium pentobarbital. The vascular system was perfused with heparinized saline, and Mercox resin was injected into the cannulated carotid arteries. After polymerization of the resin, the vascular casts were macerated with potassium hydroxide, washed with water, and freeze dried. The casts were examined with a Hitachi S-2360N scanning electron microscope (SEM). Corrosion casts of the entire cho

Arteriohepatic dysplasia (Alagille's syndrome): unusual hepatic architecture and function.

BACKGROUND: Alagille's syndrome, also called arteriohepatic dysplasia, is a congenital anomaly consisting of hepatic, ocular, skeletal, and cardiac anomalies. The abdominal imaging findings were reviewed in eight patients with biopsy-proven Alagille's syndrome. One patient also had coexistent hepatocellular carcinoma. METHODS: Seven right upper quadrant sonograms, six hepatic CT studies, five hepatobiliary imaging studies, two hepatic MRI examinations, and two sulphur colloid liver spleen radionuclide studies were reviewed. RESULTS: The most striking abnormality was gross distortion of hepat

Alterations of elastic architecture in human aortic dissecting aneurysm.

Elastin of the ascending aortic media of 10 cases with type A dissecting aneurysm, 14 hypertensive cases, and 30 control cases were prepared by the treatment of aortas with hot formic acid, and three-dimensional architecture was observed by scanning electron microscopy. In the control cases, elastin showed framework-like continuous structure consisting of elastic laminae, and interlaminar fibers that interconnected the laminae. In 6 of 10 cases of dissecting aneurysm, the interlaminar fibers were apparently irregular in arrangement and shape, and decreased in number, especially in the outer me

Histometry of splenic microvascular architecture in hairy cell leukaemia.

In this study we investigated the splenic microvascular architecture in hairy cell leukaemia, in order to provide a morphological basis for the haemodynamic modifications occurring in the disease. When compared with controls, the four leukaemic spleens examined showed a set of changes involving both the arterial and the venous system. A real increase in the absolute volume, surface and length of pulp arterial vessels was present. This increase was not so great as the enlargement of the spleen, thus resulting in a reduced density of distribution of arterial vessels in the infiltrated pulp. Enla

Mnemonic Demand Reconfigures the Neural Architecture of Memory Encoding.

Memory encoding must accommodate increasingly demanding loads, yet it remains unclear whether the underlying brain state is preserved or adaptively reconfigured as load increases. Here, we recorded EEG from 111 participants performing a visual memory task in which they encoded lists of real images ranging from 1 to 128 items, spanning low to high mnemonic demands, followed by a recognition test. We measured the similarity of set-size-specific interelectrode correlation patterns to a low-load reference. Similarity decreased monotonically as list length increased: patterns at set sizes below 4 r

Cortico-striatal network architecture shapes rhythmic biases in music.

Why is human music rhythmically structured the way it is? One hypothesis is that the human brain is intrinsically wired to favor specific rhythmic organizations. To test this, we collected diffusion and resting-state functional MRI (fMRI) data from musically untrained participants and, weeks later, asked them to imitate randomly generated drumming sequences using finger taps. These imitations exhibited systematic biases toward temporal structures widespread in human music: predictable patterns characterized by isochronous organization (1:1) and other small integer ratios (2:1 and 3:1). Functio

Molecular architecture and diversity of StopGo/2A translational recoding.

Viral 2A sequences trigger a cotranslational peptide bond formation "skipping" event, termed "StopGo," to generate two separate proteins from a single open reading frame without classical termination. To investigate the mechanism of StopGo, we determined the cryo-EM structure of a mammalian ribosome positioned at the foot-and-mouth disease virus 2A (F2A) site. The structure shows how interactions between the F2A nascent chain (NC) and the ribosomal exit tunnel induce a conformational change in the peptidyl transferase center that precludes further translation elongation but instead pre-exposes

Single-Atom Migration into a Chiral Multilayer Nanocluster Architecture.

Colloidal nanocrystals are generally regarded as rigid solid entities, rarely exhibiting the structural adaptability observed in molecular cages, such as fullerenes, which can undergo carbon framework reduction and encapsulate guest cations without structural reorganization. Here, by creating two enantiomeric pairs of high-nuclearity copper sulfide nanoclusters with a mixed-valence Cu(II)/Cu(I) configuration, we endow these nanoscale assemblies with an intrinsic capacity for electron uptake under mild reducing conditions. The resulting charge imbalance provides an effective thermodynamic drivi

Physiological brain clearance architecture revealed by neuronal protein tracing.

The brain must efficiently clear protein waste to maintain homeostasis, yet physiological drainage pathways remain poorly defined. Standard tracer injection approaches may not reflect endogenous efflux. Here, we develop a non-invasive genetic system to trace neuron-derived protein clearance from the brain to cerebrospinal fluid (CSF) and border tissues. We identify distinct drainage routes and border hotspots missed by tracer injection, confirmed by bioorthogonal labeling of endogenous neuronal proteins. Pulse-chase kinetics reveal slow skull outflow versus rapid dural and nasal clearance. Tra

Molecular architecture of the fungal-specific potassium channel TOK1.

In Candida albicans, potassium (K+) channels fine-tune ionic balance under stress, contributing to host colonization. Fungal two-pore domain, outwardly rectifying potassium (TOK) channels remain insufficiently characterized despite evidence implicating them in growth and viability. Here, we describe the atomic-resolution structure of a fungal potassium channel, TOK1 from C. albicans (CaTOK), revealing an architecture defined by eight transmembrane helices and a membrane topology distinct from previously characterized K⁺ channel classes. The first four helices form a tetraspanin-like bundle res
