Hamsters exposed to 10 ppm NO2 for 5 hr once a week for 4 weeks while undergoing acute vitamin A deficiency showed much reduced epithelial cell regeneration in the terminal bronchioles. Quantitative analysis done by autoradiography and scintillation counting from lung tissues indicates much reduced cell kinetics occurring in terminal bronchiolar alveolar region. Alveolar necrosis was often observed and no type II cell reversion occurred. Virus particles were found within the alveolar epithelial plasma membrane.

The localization of mRNA coding for the dopamine D2 receptor was studied in the rabbit carotid body using in situ hybridization with synthetic 35S-labelled oligodeoxynucleotides. Using autoradiography on cryostat or semi-thin sections, labelling was observed over the cytoplasm of glomus cells, but not over sustentacular cells. A quantitative study showed that labelling intensity (silver grain density) was increased by haloperidol treatment. These results suggest that glomus cells express the dopamine D2 receptor gene and that this expression is regulated.

Permeabilization with cetyl trimethyl ammonium bromide was used to study the post-translational modification of the PII protein in Rhizobium leguminosarum. Upon incubation with radioactive UTP a single band was obtained after SDS-PAGE and autoradiography. RNase resistance and snake venom phosphodiesterase sensitivity showed that radioactivity was bound through a phosphodiester bond to a protein which was absorbed by an antiserum specific for the PII protein. Uridylylation of the PII protein was shown to be dependent on the modifications of the glutamine/alpha-ketoglutarate ratio.

We used quantitative in vitro autoradiography to localize and characterize 2-125I-melatonin binding sites in the rat suprachiasmatic nuclei in relation to pineal melatonin production. In a light:dark cycle of 12:12 h, binding density exhibited significant diurnal variation with a peak at the dark-light transition and a trough 12 hours later. Saturation studies suggested that the decreased binding at light-dark transition might be due to a shift of the putative melatonin receptor to a low affinity state.

The human papova (JC) virus was extracted from brain of a patient with progressive multifocal leukoencephalopathy. A single band of virus was obtained at a density of 1.345 g/ml CsCl.JC virus DNA was purified and a highly specific cRNA was generated in vitro. In situ hybridization with JC virus cRNA and autoradiography on sections of the same brain revealed silver grains over oligodendrocytes, astrocytes and possible vascular endothelial cells, indicating the presence of JC virus DNA in these different cell classes.

The diagnosis of Lesch-Nyhan syndrome was confirmed by the finding of low levels of hypoxanthine-guanine phosphoribosyl transferase in erythrocytes and by autoradiography of fibriblasts. The mother, maternal grandmother, a female sibling and a maternal aunt were identified as carriers of the X-linked mutation which was responsible for the enzyme deficiency in the two male siblings.

The monoclonal antibody TU-20 and its scFv fragment were radiolabeled with 125 I in order to develop new imaging agents against the specific neuronal marker III beta-tubulin. The reaction via chloramine-T using thiosulfate as a stopping reductant was determined as the most convenient way for radioiodination. The preserved immunological properties of radioiodinated species were estimated by ELISA, electrophoresis, and immunohistochemistry with autoradiography. Biodistribution studies revealed a different behavior of radioiodinated TU-20 and its scFv.

Local cerebral blood flow and glucose utilisation were measured in both saline (n = 10) and cocaine (10 mg/kg; n = 10) treated rats using [14C]iodoantipyrine and [14C]2-deoxyglucose quantitative autoradiography respectively. In control animals, the ratio of flow to metabolism was 1.40 (r = 0.92) for the 40 brain regions examined. Cocaine treatment altered neither the correlation (r = 0.83) nor the ratio (1.49). Thus, the fundamental relationship between CBF and metabolism remains intact following acute cocaine exposure.

A 6-year-old girl with Ehlers-Danlos syndrome (type I) who shows hyperelastic skin, hypermobile joints and delayed wound healing is reported. The growth kinetics of fibroblasts derived from her skin was examined using 3H-thymidine autoradiography. Our study showed that these fibroblasts had poor proliferative activity, compared with normal age-matched fibroblasts. Thus, we could suggest that the pathogenesis of Ehlers-Danlos syndrome was partially attributed to the lack of proliferative activity of the fibroblasts.

3H-thymidine autoradiography showed an enhanced DNA synthesis in acinar and islet cells of pancreas after partial hepatectomy in syrian golden hamsters. A significant nuclear labeling index of acinar cells was observed between 48 and 84 h and reached control levels by 120 h. An increased labeling index of islet cells was also observed, however, this increase was not statistically significant. These results indicate growth factor(s) produced after partial hepatectomy is capable of inducing DNA synthesis in pancreas.

Treatment of neonatal mice with diethylstilbestrol results in the appearance of adenosis in the uterine cervix and upper part of the vagina in later life. Using dry mount autoradiography, the uptake and distribution of [3H]-estradiol in the same regions were studied after in vivo injections with [3H]-estradiol. A technique is described for the detection of immunoglobulins secreted by individual lymphoid cells. Cells were cultured in agarose gel containing dilute radiolabelled anti-immunoglobulin antibodies. The secreted immunoglobulins produced a precipitin lattice which was made visible by washing the gel, followed by autoradiography. Immunoglobulin secreting cells were surrounded by clusters, or `galaxies' of silver grains. Galaxies did not appear in the presence of certain metabolic inhibitors, indicating that the secretion of immunoglobulin was an active process dependent upon cell metabolism.
