Multi-omics analyses of the ulcerative colitis gut microbiome link Bacteroides vulgatus proteases with disease severity.

Ulcerative colitis (UC) is driven by disruptions in host-microbiota homoeostasis, but current treatments exclusively target host inflammator … clinically active UC patients had an overabundance of proteases that originated from the bacterium Bacteroides vulgatus. To test whether B. vulgatus proteases contribute to UC disease activity, we first profiled B. vulgatus proteases found in patients and bacterial cultures. Use of a broad-spectrum protease inhibitor improved B. vulgatus-induced barrier dysf …

Metallo-β-lactamase-producing bacteroides species can shield other members of the gut microbiota from antibiotics.

Antibiotics disrupt the intestinal microbiota, rendering patients vulnerable to colonization by exogenous pathogens. Intermicrobial interactions may attenuate this effect. Incubation with ceftriaxone-resistant, ccrA-positive, β-lactamase-producing Bacteroides strains raised the minimum bactericidal concentration of ceftriaxone required to kill a susceptible Escherichia coli strain (mean change, <0.25 to 29 mg/liter; P = 0.009); incubation with ceftriaxone-resistant but non-β-lactamase-producing Bacteroides strains had no effect. The production of β-lactamase by common members of the intestinal…

Aberrant response to commensal Bacteroides thetaiotaomicron in Crohn's disease: an ex vivo human organ culture study.

BACKGROUND: Human ex vivo evidence indicating that an inappropriate immune response(s) to nonpathogenic bacteria contributes to disease pathogenesis in pediatric Crohn's disease (CD) is limited. The aim of the present study was to compare and contrast the early innate immune response of pediatric "Работает" versus CD mucosa to pathogenic, probiotic, and commensal bacteria. METHODS: "Healthy control" and CD pediatric mucosal biopsies (terminal ileum and transverse colon) were cocultured for 8 hours with E. coli O42, Lactobacillus GG (LGG), Bacteroidesthetaiotaomicron (B. theta), or stimulated w…

The survival rate of Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, and Bacteroides forsythus following 4 randomized treatment modalities.

The overall goal of this clinical study was to determine the short-term anti-infective effects of four randomized treatment modalities on Actinobacillus actinomycetemcomitans (Aa), Porphyromonas gingivalis (Pg), and Bacteroides forsythus (Bf) and determine the effects of bacterial survival on treatment outcomes in patients with adult periodontitis. Twelve adult patients requiring therapy for moderate periodontitis were selected for this study. All patients had at least one tooth in each quadrant that had an inflamed pocket of probing depth > or =5 mm with probing attachment loss that harbored …

Purification and Characterization of His-Tagged Recombinant Bacteroides fragilis Toxin-2 Variants In Vitro and In Vivo.

Bacteroides fragilis is a major commensal bacterium of the human colon. However, enterotoxigenic B. fragilis (ETBF) secretes B. fragilis toxin (BFT), a zinc-dependent metalloprotease that cleaves E-cadherin and promotes chronic inflammation and colorectal tumorigenesis. Despite extensive research, the cellular receptor for BFT remains unidentified. In this study, we developed His-tagged recombinant BFT variants including both catalytically active and inactive forms to facilitate biochemical and functional analyses. Functional assays confirmed that the active variant retained proteolytic activi…

Observations Concerning Rhizobium tropici Bacteroid Phosphorus Stress Response During Symbiosis With Phaseolus vulgaris.

Bacteroid inorganic phosphorus (Pi) metabolism in the Rhizobium-legume symbiosis differs between indeterminate and determinate legume nodules. In contrast to alfalfa bacteroids, bean (Phaseolus vulgaris) bacteroids exhibit high levels of alkaline phosphatase (AP), the native reporter enzyme for the bacterial Pi stress response. 14C and 32Pi whole plant labelling techniques were used in conjunction with diagnostic mutants (lacking AP or lacking high affinity Pi transport) to assess the relative importance of the Pi stress response in Rhizobium tropici bacteroids during symbiosis. The AP- mutant

Unique presentation of an intramuscular abscess caused by Bacteroides pyogenes in the setting of a cat bite: A case report.

Cat bites are a relatively common emergency department presentation. The most common species isolated from cat bite wounds is Pasteurella multocida. In this case report, we discuss a unique case of an intramuscular abscess caused by Bacteroides pyogenes. Antibiotic resistance in Bacteroides species is a growing concern and thus should be treated with caution. We present an 81-year-old female who presented to the emergency department with left calf pain that began 3 weeks prior to the emergency department presentation. She was initially seen by her primary care provider and was prescribed oral …

In vitro activity of eravacycline and comparator antimicrobials against 143 recent strains of Bacteroides and Parabacteroides species.

Eravacycline, a novel fluorocycline antibiotic, has been evaluated against complicated mixed aerobic/anaerobic intra-abdominal infections but scant supporting in vitro data against anaerobes has been published. We found that eravacycline had good anaerobic in vitro activity with MICs of 4 μg/ml or less against all Bacteroides and Parabacteroides strains tested, except for two B. ovatus strains that had MICs of 8 μg/ml and one strain that had an MIC of 16 μg/ml. Eravacycline was four-to-eight fold more active than tigecycline.

A comparison of IgG antibody reactive with Bacteroides forsythus and Porphyromonas gingivalis in adult and early-onset periodontitis.

Recent work has indicated that Bacteroides forsythus and Porphyromonas gingivalis are significant local risk factors for periodontitis. Several reports find that both organisms are frequently associated with periodontitis lesions and often are present together. We have previously shown that early-onset periodontitis patients seropositive for P. gingivalis have less attachment loss than seronegative patients. In this study, we determined serum IgG antibody concentrations reactive with B. forsythus in adult and early-onset periodontitis patients using an ELISA and used P. gingivalis in the same …

Susceptibility patterns and resistance to imipenem in the Bacteroides fragilis group species in Japan: a 4-year study.

A study was undertaken in Japan to evaluate the susceptibility patterns of Bacteroides fragilis group species (849 strains) isolated from December 1986 through May 1991. All of the strains, which included B. fragilis (610 strains), Bacteroides thetaiotaomicron (201 strains), and Bacteroides distasonis (38 strains), were studied for susceptibility to imipenem and 16 other antimicrobial agents by an agar dilution method. Metronidazole was the most active agent; the minimal concentration for 90% inhibition (MIC90) was 0.78 micrograms/mL, and no isolate was noted to be resistant to it during the e…

Belgian Collaborative Study of the in-vitro susceptibility of the Bacteroides fragilis group. A Belgian Collaborative Study Group.

The antimicrobial susceptibility of clinical isolates of the Bacteroides fragilis group obtained from nine large Belgian university and community hospitals was studied. Of the beta-lactam antibiotics tested, none were active against 100% of the isolates. Piperacillin was active against 93% of the strains at a breakpoint of 64 micrograms/ml. The percentage of isolates inhibited at 16 micrograms/ml (and 32 micrograms/ml) for the 7-alpha-methoxy antibiotics was: cefoxitin 84; latamoxef 87; and cefotetan 64. B. fragilis tended to be more susceptible to all beta-lactams than the other members of th…

The capsular polysaccharide of Bacteroides fragilis as a virulence factor: comparison of the pathogenic potential of encapsulated and unencapsulated strains.

The pathogenic potentials of encapsulated and unencapsulated strains of Bacteroides fragilis were compared by use of a rat model of intraabdominal sepsis. Implantation of encapsulated B. fragilis alone resulted in abscesses in most recipients, whereas unencapsulated strains seldom produced this effect unless they were combined with another organism. Implants of heat-killed, encapsulated B. fragilis also resulted in abscess formation. Subsequent experiments suggested that the abscess-potentiating ability of encapsulated B. fragilis is related to the capsular polysaccharide. Implantation of 200 …

Nutrient niche specificity for glycosaminoglycans is reflected in polysaccharide utilization locus architecture of gut Bacteroides species.

INTRODUCTION: Glycosaminoglycans (GAGs) present in the mucosal layer can be used as nutrients by certain intestinal bacteria, particularly members of the Bacteroides. GAG abundances are altered in some diseases such as inflammatory bowel diseases, which may affect microbial composition and activity, and it is therefore important to understand GAG utilization by members of the gut microbiota. METHODS: We used growth assays, transcriptomics, and comparative genomics to evaluate chondroitin sulfate (CS) and hyaluronan (HA) degradation ability by multiple gut Bacteroides species. RESULTS AND DIS…
